Abstract

The present invention is related to a beta-herpesvirus, wherein the beta-herpesvirus is spread-deficient.

Claims (20)

1. A method for the vaccination of a human or for the treatment of a disease of human comprising administering to the human a human beta-herpesvirus, wherein the beta-herpesvirus is spread-deficient and wherein the beta-herpesvirus us endotheliotropic and/or has a wild type-like virion surface, wherein the beta-herpesvirus is deficient in at least one gene product involved in primary and/or secondary envelopment, wherein the at least one gene product involved in primary involvement is encoded by a gene selected from the group consisting of UL50 and UL53 and homologs of each thereof, wherein the at least one gene product involved in secondary envelopment is encoded by a gene selected from the group consisting of UL94 and UL99 and homologs thereof, and wherein the beta-herpesvirus is human cytomegalovirus.

2. The method of claim 1 , wherein the beta-herpesvirus is endotheliotropic and has a wild type-like virion surface.

3. The method of claim 1 , wherein the beta-herpesvirus induces and immune response, wherein the immune response comprises neutralizing antibodies against beta-herpesvirus and CD4 + and CD8 + T-cells directed against epitopes of beta-herpesvirus.

4. The method of claim 3 , wherein the immune response comprises induction of neutralizing antibodies against a wild type beta-herpesvirus, wherein said antibodies are capable of inhibiting said wild type beta-herpesvirus from infecting endothelial cells and/or epithelial cells.

5. The method of claim 1 , wherein the beta-herpesvirus comprises a deletion of nucleotides 122631-134667 and 181653-189191 of SEQ ID NO: 20, and wherein SEQ ID NO: 34 is inserted into said virus to replace deleted nucleotides 12263-123667 from SEQ ID NO: 29.

6. The method of claim 1 , wherein the beta-herpesvirus further comprises a deletion of nucleotides 130671-131242 of SEQ ID NO: 20.

7. The method of claim 1 , wherein SEQ ID NO: 35 is inserted into said virus to replace deleted nucleotides 130671-131242 from SEQ ID NO: 20.

8. The method of claim 1 , wherein the beta-herpesvirus comprises a deletion of nucleotides 58443-59622, 181653-189191 of SEQ ID NO: 20, and wherein SEQ ID NO: 32 is inserted into said virus to replace deleted nucleotides 58443-59622 from SEQ ID NO: 20.

9. The method of claim 1 , wherein the beta-herpesvirus comprises a deletion of nucleotides 62130-63260 and 181653-189191 of SEQ ID NO: 20 and wherein SEQ ID NO: 33 is inserted into said virus to replace deleted nucleotides 62130-63260 from SEQ ID NO: 20.

10. The method of claim 9 , wherein the virus further comprises a deletion of nucleotides 58443-59622.

11. The method of claim 10 , wherein SEQ ID NO: 32 is inserted into said virus to replace deleted nucleotides 58443-59622 from SEQ ID NO: 20.

12. The method of claim 1 , wherein the beta-herpesvirus comprises the nucleotide sequence of SEQ ID NO: 23.

The method of claim 1 , wherein the beta-herpesvirus is deficient in at least one gene product encoded by an immune evasive gene, wherein the at least one gene product encoded by an immune evasive gene is selected from the group comprising gene products regulating MHC class I presentation and gene products regulating NK cell response.

14. The method of claim 13 , wherein the gene product regulating NK cell response is selected from the group comprising gene products encoded by the genes UL40, UL16 and UL18.

15. The method of claim 13 , wherein the gene product regulating MHC class I presentation is selected from the group consisting of US6, US3, US2, UL18, US11, UL83 and UL40.

The method of claim 1 , wherein the beta-herpesvirus encodes a heterologous nucleic acid, wherein the heterologous nucleic acid is a functional nucleic acid selected from the group consisting of antisense molecules, ribozymes and RNA interference mediating nucleic acids, or a nucleic acid a nucleic acid coding for a peptide, oligopeptide, polypeptide or protein.

17. The method of claim 1 , wherein the disease is a disease associated with human cytomegalovirus infection.

18. The method of claim 17 , wherein the disease comprises congenital inclusion disease.

19. The method of claim 1 , wherein the vaccination is against a disease, wherein the disease is a disease associated with human cytomegalovirus infection.

20. The method of claim 19 , wherein the disease comprises congenital inclusion disease.
