Abstract

The present invention relates to combination therapies for the treatment of malignancy, particularly myeloid malignancy such as acute myeloid leukemia (AML). The combination therapies may include an antibody molecule that binds to CD70 and at least one antibody molecule that binds to a leukemic stem cell target. Preferred leukemic stem cell targets are TIM-3, IL1R3/IL1RAP and CD47.

Claims (74)

1. A combination comprising a first antibody molecule that specifically binds to CD70 and a second antibody molecule that specifically binds to TIM-3,

wherein the first antibody molecule comprises a first variable heavy chain domain (VH) comprising HCDR1, HCDR2 and HCDR3 regions, and a first variable light chain domain (VL) comprising LCDR1, LCDR2 and LCDR3 regions, wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3 regions of the first VH and the first VL comprise the amino acid sequences set forth in SEQ ID NOs: 1 2, 3, 5, 6, and 7, respectively, and

wherein the second antibody molecule comprises a second VH comprising HCDR1, HCDR2 and HCDR3 regions, and a second VL comprising LCDR1, LCDR2 and LCDR3 regions, wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3 regions of the second VH and the second VL comprise:

(i) SEQ ID NOs: 39, 40, 41, 78, 79, and 80, respectively;

(ii) SEQ ID NOs: 39, 42, 43, 81, 82, and 83, respectively;

(iii) SEQ ID NOs: 44, 45, 46, 84, 85, and 86, respectively;

(iv) SEQ ID NOs: 47, 48, 49, 87, 82, and 88, respectively;

(v) SEQ ID NOs: 50, 51, 52, 89, 90, and 91, respectively;

(vi) SEQ ID NOs: 53, 54, 55, 92, 93, and 94, respectively;

(vii) SEQ ID NOs: 56, 57, 58, 95, 96, and 97, respectively;

(viii) SEQ ID NOs: 50, 59, 60, 98, 99, and 100, respectively;

(ix) SEQ ID NOs: 61, 62, 63, 101, 102, and 103, respectively;

(x) SEQ ID NOs: 39, 64, 65, 104, 105, and 106, respectively;

(xi) SEQ ID NOs: 50, 66, 67, 107, 108, and 109, respectively;

(xii) SEQ ID NOs: 50, 68, 69, 110, 111, and 112, respectively;

(xiii) SEQ ID NOs: 70, 71, 72, 113, 114, and 115, respectively;

(xiv) SEQ ID NOs: 50, 73, 74, 116, 111, and 117, respectively; or

(xv) SEQ ID NOs: 75, 76, 77, 118, 119, and 120, respectively.

2. The combination of claim 1 , wherein the first VH comprises an amino acid sequence at least 70% identical to the amino acid sequence of SEQ ID NO: and the first VL comprises an amino acid sequence at least 70% identical to the amino acid sequence of SEQ ID NO: 8.

3. The combination of claim 2 , wherein the first antibody molecule is ARGX-110.

4. The combination of claim 1 , wherein the second antibody molecule inhibits the interaction of TIM-3 with one or more TIM-3 interacting moieties.

5. The combination of claim 4 , wherein the TIM-3 interacting moieties are selected from the group consisting of: CEACAM-1, HMGB-1, phosphatidylserine, Galectin-9, LILRB2, and any combination thereof.

6. The combination of claim 1 , wherein:

the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 9, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 10;

(ii) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 11, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 12;

(iii) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 13, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 14;

(iv) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 15, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 16;

(v) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 17, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 18;

(vi) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 19, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 20;

(vii) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 21, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 22;

(viii) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 23, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 24;

(ix) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 25, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 26;

(x) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 27, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 28;

(xi) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 29, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 30;

(xii) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 31, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 32;

(xiii) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 33, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 34;

(xiv) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 35, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 36; or

(xv) the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 37, and the second VL comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 38.

7. The combination of claim 1 , wherein the first and second antibody molecules of the combination are combined in a multispecific antibody.

8. The combination of claim 7 , wherein the multispecific antibody is a bispecific antibody comprising the first antibody molecule and the second antibody molecule.

9. The combination of claim 1 , wherein the first and second antibody molecules of the combination are co-formulated.

10. The combination of claim 1 , wherein the first and second antibody molecules of the combination are provided separately.

11. The combination of claim 1 , wherein the combination additionally comprises an agent that inhibits SIRPα signaling.

12. The combination of claim 11 , wherein the agent that inhibits SIRPα signaling is selected from the group consisting of: (i) an antibody molecule that binds CD47 and inhibits interaction between CD47 and SIRPα; (ii) an antibody molecule that binds SIRPα and inhibits interaction between CD47 and SIRPα; and (iii) an SIRPα-antibody molecule fusion protein.

13. A combination comprising a first antibody molecule that specifically binds to CD70 and a second antibody molecule that specifically binds to TIM-3,

wherein the first antibody molecule comprises a first VH comprising HCDR1, HCDR2 and HCDR3 regions, and a first VL comprising LCDR1, LCDR2 and LCDR3 regions, wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3 regions of the first VH and the first VL comprise the amino acid sequences set forth in SEQ ID NOs: 1, 2, 3, 5, 6, and 7, respectively, and

wherein the second antibody molecule comprises a (second VH comprising HCDR1, HCDR2 and HCDR3 regions, and a (second VL comprising LCDR1, LCDR2 and LCDR3 regions, wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2 and LCDR3 regions of the second VH and the second VL comprise the amino acid sequences set forth in SEQ ID NOs: 56, 57, 58, 95, 96, and 97, respectively.

14. The combination of claim 13 ,

wherein the second VH comprises an amino acid sequence at least 80% identical to the amino acid sequence of SEQ ID NO: 21, and the second VL comprises an amino acid sequence having at least 80% identical to the amino acid sequence of SEQ ID NO: 22.

15. The combination of claim 14 , wherein the second VH comprises the amino acid sequence of SEQ ID NO: 21, and the second VL comprises the amino acid sequence of SEQ ID NO: 22.

16. The combination of claim 13 , wherein the first VH comprises an amino acid sequence at least 70% identical to the amino acid sequence of SEQ ID NO: 4, and the first VL comprises an amino acid sequence at least 70% identical to the amino acid sequence of SEQ ID NO: 8.

17. The combination of claim 16 , wherein the first VH comprises the amino acid sequence of SEQ ID NO: 4, and the first VL comprises the amino acid sequence of SEQ ID NO: 8.

18. The combination of claim 13 , wherein at least one of the antibody molecules comprises a defucosylated antibody domain.

19. The combination of claim 16 , wherein the first antibody molecule is ARGX-110.

20. The combination of claim 13 , wherein the second antibody molecule is an IgG1 antibody molecule.

21. The combination of claim 1 , wherein at least one of the antibody molecules comprises a defucosylated antibody domain.

22. The combination of claim 1 , wherein the second antibody molecule is an IgG1 antibody molecule.

23. The combination of claim 2 , wherein the first VH comprises the amino acid sequence of SEQ ID NO: 4, and the first VL comprises the amino acid sequence of SEQ ID NO: 8.

24. The combination of claim 6 , wherein:

the second VH comprises the amino acid sequence of SEQ ID NO: 9, and the second VL comprises the amino acid sequence of SEQ ID NO: 10;

(ii) the second VH comprises the amino acid sequence of SEQ ID NO: 11, and the second VL comprises the amino acid sequence of SEQ ID NO: 12;

(iii) the second VH comprises the amino acid sequence of SEQ ID NO: 13, and the second VL comprises the amino acid sequence of SEQ ID NO: 14;

(iv) the second VH comprises the amino acid sequence of SEQ ID NO: 15, and the second VL comprises the amino acid sequence of SEQ ID NO: 16;

(v) the second VH comprises the amino acid sequence of SEQ ID NO: 17, and the second VL comprises the amino acid sequence of SEQ ID NO: 18;

(vi) the second VH comprises the amino acid sequence of SEQ ID NO: 19, and the second VL comprises the amino acid sequence of SEQ ID NO: 20;

(vii) the second VH comprises the amino acid sequence of SEQ ID NO: 21, and the second VL comprises the amino acid sequence of SEQ ID NO: 22;

(viii) the second VH comprises the amino acid sequence of SEQ ID NO: 23, and the second VL comprises the amino acid sequence of SEQ ID NO: 24;

(ix) the second VH comprises the amino acid sequence of SEQ ID NO: 25, and the second VL comprises the amino acid sequence of SEQ ID NO: 26;

(x) the second VH comprises the amino acid sequence of SEQ ID NO: 27, and the second VL comprises the amino acid sequence of SEQ ID NO: 28;

(xi) the second VH comprises the amino acid sequence of SEQ ID NO: 29, and the second VL comprises the amino acid sequence of SEQ ID NO: 30;

(xii) the second VH comprises the amino acid sequence of SEQ ID NO: 31, and the second VL comprises the amino acid sequence of SEQ ID NO: 32;

(xiii) the second VH comprises the amino acid sequence of SEQ ID NO: 33, and the second VL comprises the amino acid sequence of SEQ ID NO: 34;

(xiv) the second VH comprises the amino acid sequence of SEQ ID NO: 35, and the second VL comprises the amino acid sequence of SEQ ID NO: 36; or

(xv) the second VH comprises the amino acid sequence of SEQ ID NO: 37, and the second VL comprises the amino acid sequence of SEQ ID NO: 38.
