Abstract

Four genes, A622, NBB1, PMT, and QPT, can be influenced for increasing nicotinic alkaloid levels in Nicotiana plants, as well as for synthesizing nicotinic alkaloids in non-nicotine producing plants and cells. In particular, overexpressing one or more of A622, NBB1, PMT, and QPT may be used to increase nicotine and nicotinic alkaloid levels in tobacco plants. Non-nicotine producing cells can be engineered to produce nicotine and related compounds by overexpressing A622 and NBB1.

Claims (14)

1. A tobacco plant comprising a nucleic acid construct comprising, in the 5′ to 3′ direction, a promoter operably linked to a heterologous nucleic acid encoding an NBB1 gene, wherein the NBB1 gene comprises the nucleotide sequence set forth in SEQ ID NO:1 or encodes a polypeptide having the amino acid sequence set forth in SEQ ID NO: 2, wherein the tobacco plant exhibits an increased level of nicotinic alkaloid as compared to a control non-transformed tobacco plant.

2. The tobacco plant of claim 1 , wherein the nucleic acid construct further comprises a nucleic acid encoding an enzyme selected from the group consisting of A622, quinolate phosphoribosyl transferase (QPT), putrescine methyl transferase (PMT), and any combination thereof.

3. The tobacco plant of claim 1 , wherein the tobacco plant is a Nicotiana plant.

4. The tobacco plant of claim 3 , wherein the tobacco plant is Nicotiana tabacum.

5. The tobacco plant of claim 1 , wherein the nicotinic alkaloid is nicotine.

6. Seeds from or a progeny plant of the tobacco plant of claim 1 , wherein the seeds or progeny plant comprise the nucleic acid construct.

7. A product comprising the plant of claim 1 , and having an increased level of nicotinic alkaloid as compared to a product produced from a control plant.

8. The product of claim 7 , wherein the product is selected from the group consisting of a tobacco product, a pharmaceutical, and a nutraceutical.

9. The tobacco product of claim 8 , wherein the product is selected from the group consisting of expanded tobacco, reconstituted tobacco, a cigarette, a cigar, cigar tobacco, pipe tobacco, chewing tobacco, smokeless tobacco, snuff, snus leaf tobacco, shredded tobacco, and cut tobacco.

10. The tobacco product of claim 9 , wherein the product has a two-fold or greater increase in nicotine as compared to a product produced from a control non-transformed tobacco plant.

11. The tobacco plant of claim 1 , wherein NBB1 is overexpressed as compared to a control non-transformed tobacco plant.

12. The tobacco plant of claim 1 , wherein NBB1 and A622 are overexpressed as compared to a control non-transformed tobacco plant.

13. The tobacco plant of claim 1 , wherein NBB1 and QPT are overexpressed as compared to a control non-transformed tobacco plant.

14. The tobacco plant of claim 1 , wherein NBB1 and PMT are overexpressed as compared to a control non-transformed tobacco plant.
