Pyrazole compounds and methods for making and using the compounds
Disclosed embodiments concern novel interleukin receptor associated kinases (IRAK) inhibitors and compositions comprising such inhibitors. Also disclosed are methods of making and using the compounds and compositions. The disclosed compounds and/or compositions may be used to treat or prevent an IRAK-associated disease or condition.
1. A method for treating a disease or condition for which an IRAK inhibitor is indicated, comprising administering to a subject in need thereof an amount of a compound effective to treat the disease or condition, wherein the compound has a formula
or salt thereof, wherein:
R is monocyclic heteroaryl;
R 1 is H, aliphatic or heteroaliphatic;
R 2 is H, aliphatic, heteroaliphatic, heterocycloaliphatic, aryl, amide, heterocyclyl or araliphatic;
each R 3 independently is H, aliphatic, halogen, heteroaliphatic, —O-aliphatic, heterocyclyl, aryl, araliphatic, —O-heterocyclyl, hydroxyl, nitro, cyano, carboxyl, carboxyl ester, acyl, amide, amino, sulfonyl, sulfonamide, sulfanyl, sulfinyl, haloalkyl, alkylphosphate, or alkylphosphonate, wherein R 3 is not pyridinyl;
y is from 1 to 6; and
Het-1 is furanyl or thiazolyl, and is substituted with pyrazolyl; or

2. A method for inhibiting an IRAK protein, comprising contacting the IRAK protein with an effective amount of a compound wherein the compound has a formula
or salt thereof, wherein:
R is monocyclic heteroaryl;
R 1 is H, aliphatic or heteroaliphatic;
R 2 is H, aliphatic, heteroaliphatic, heterocycloaliphatic, aryl, amide, heterocyclyl or araliphatic;
each R 3 independently is H, aliphatic, halogen, heteroaliphatic, —O-aliphatic, heterocyclyl, aryl, araliphatic, —O-heterocyclyl, hydroxyl, nitro, cyano, carboxyl, carboxyl ester, acyl, amide, amino, sulfonyl, sulfonamide, sulfanyl, sulfinyl, haloalkyl, alkylphosphate, or alkylphosphonate, wherein R 3 is not pyridinyl;
y is from 1 to 6; and

3. The method of claim 2 , wherein the compound has a formula

R 2 is H, aliphatic, heteroaliphatic, heterocycloaliphatic, aryl, amide, heterocyclyl or araliphatic;
each R 3 independently is H, aliphatic, halogen, heteroaliphatic, —O-aliphatic, heterocyclyl, aryl, araliphatic, —O-heterocyclyl, hydroxyl, nitro, cyano, carboxyl, carboxyl ester, acyl, amide, amino, sulfonyl, sulfonamide, sulfanyl, sulfinyl, haloalkyl, alkylphosphate, or alkylphosphonate, providing R 3 is not pyridinyl;
x is from 1 to 6;
y is from 1 to 6;
z is from 1 to 6;
Het-1 is furanyl or thiazolyl;
Het-2 is monocyclic heteroaryl; and
Het-3 is pyrazolyl.
4. The method of claim 3 , wherein Het-2 is pyridinyl, pyrimidinyl, pyrazinyl, oxadiazolyl or thiazolyl.
5. The method of claim 4 , wherein the compound has a formula
wherein R 4 , R 5 , R 6 and R 7 are each independently H, aliphatic, heteroaliphatic, alkoxy, heterocyclyl, aryl, araliphatic, —O-heterocyclyl, hydroxyl, haloalkyl, halogen, nitro, cyano, carboxyl, carboxyl ester, acyl, amide, amino, sulfonyl, sulfonamide, sulfanyl or sulfinyl.
6. The method of claim 5 , wherein each of R 4 , R 6 , and R 7 independently is H, F or CF 3 , and R 5 is H, F, CF 3 , methoxy, morpholin-4-yl, 1-methylpiperidin-4-yl, —O—CH 2 C(CH 3 ) 2 OH, or —O-(oxetan-3-yl).
7. The method of claim 5 , wherein the compound has a formula
wherein:
R 8 and R 9 are each independently H, aliphatic, heteroaliphatic, aryl, heterocyclyl, sulfonyl, nitro, halogen, haloalkyl, carboxyl ester, cyano or amino; and
R 10 is H, aliphatic, alkoxy, heteroaliphatic, carboxyl ester, araliphatic, NO 2 , CN, OH, haloalkyl, acyl, alkyl phosphate or alkylphosphonate.
8. The method of claim 7 , wherein R 8 and R 9 are each independently H, halogen, haloalkyl, or alkyl.
9. The method of claim 7 , wherein R 10 is H, alkyl, carboxyl ester, acyl, alkyl phosphate, alkyl phosphonate or aralkyl.

11. The method of claim 2 , comprising inhibiting an IRAK protein in vivo.
12. The method of claim 11 , wherein the IRAK protein is in a subject.
13. The method of claim 12 , wherein the method is a method for treating a disease or condition for which an IRAK inhibitor is indicated.

The method of claim 13 , wherein the disease is an auto-immune disease, inflammatory disorder, cardiovascular disease, neurodegenerative disorder, allergic disorder, multi-organ failure, kidney disease, platelet aggregation, cancer, transplantation, sperm motility, erythrocyte deficiency, graft rejection, lung injury, respiratory disease, ischemic condition, bacterial infection, viral infection, immune regulatory disorder or a combination thereof.
15. The method of claim 14 , wherein the disease or condition is an auto-immune disease, inflammatory disorder, cardiovascular disease, cancer, ischemic condition, immune regulatory disorder or a combination thereof.
16. The method of claim 14 , wherein the disease or condition is an auto-immune disease or inflammatory disorder.
17. The method of claim 14 , wherein the cancer is breast cancer, lung cancer, melanoma, leukemia, large B-cell lymphoma, Waldenström's macroglobulinemia, myelodysplastic syndromes, Kaposi's sarcoma, or a combination thereof.
