Cytotoxic and anti-mitotic compounds, and methods of using the same
Compounds having cytotoxic and/or anti-mitotic activity are disclosed. Methods associated with preparation and use of such compounds, as well as pharmaceutical compositions comprising such compounds, are also disclosed. Also disclosed are compositions having the structure: (T)-(L)-(D), wherein (T) is a targeting moiety, (L) is an optional linker, and (D) is a compound having cytotoxic and/or anti-mitotic activity.
1. A conjugate having the following structure:
(T)-(L)-(PT) (VII)
wherein (T) is a targeting moiety, wherein the targeting moiety comprises an antibody or an antigen-binding antibody fragment,
(L) is a linker, and
(PT) is a hemiasterlin analog, and
wherein (T)-(L)-(PT) has the following structure:
wherein:
R 15 is selected from the group consisting of optionally substituted alkyl, optionally substituted alkylamino, optionally substituted cycloalkyl, optionally substituted aryl, optionally substituted heterocyclyl and optionally substituted heteroaryl;
R 16 and R 17 are independently selected from the group consisting of H and C 1-6 alkyl;
R 18 and R 30 are independently selected from the group consisting of H, C 1-6 alkyl and —SH, with the proviso that R 18 and R 30 cannot both be H;
R 32 is:
Z is selected from the group consisting of: —OH; —OR; —SH; —SR; —NH 2 ; —NRCH(R 11 )COOH; and —NHCH(R 11 )COOH, wherein R 11 is R or —(CH 2 ) n NR 12 R 13 , wherein n=1−4 and R 12 and R 13 are independently selected from the group consisting of: H, R and —C(NH)(NH 2 );
R is a saturated or unsaturated linear or branched alkyl containing one to ten carbon atoms, or a cycloalkyl or heterocyclyl containing three to ten carbon atoms and zero to four nitrogen atoms, wherein the carbon atoms are optionally independently substituted with: ═O, ═S, OH, —OR 10 , —O 2 CR 10 , —SH, —SR 10 , —SOCR 10 , —NH 2 , —NHR 10 , —N(R 10 ) 2 , —NHCOR 10 , —NR 10 COR 10 , —I, —Br, —Cl, —F, —CN, —CO 2 H, —CO 2 R 10 , —CHO, —COR 10 , —CONH 2 , —CONHR 10 , —CON(R 10 ) 2 , —COSH, —COSR 10 , —NO 2 , —SO 3 H, —SOR 10 , or —SO 2 R 10 , wherein R 10 is a linear or branched one to ten carbon saturated or unsaturated alkyl group or a three to ten carbon cycloalkyl;
Y is a linear, saturated or unsaturated, one to six carbon alkyl group, optionally substituted with R, and
R 14 is selected from the group consisting of optionally substituted alkyl, optionally substituted alkylamino, optionally substituted cycloalkyl, optionally substituted aryl, optionally substituted aralkyl, optionally substituted heterocyclyl and optionally substituted heteroaryl.
2. The conjugate according to claim 1 , wherein (T)-(L)-(PT) has the following structure:

3. The conjugate according to claim 2 , wherein R 15 is optionally substituted aryl or optionally substituted heteroaryl.
4. The conjugate according to claim 2 , wherein Z is OH, SH or NH 2 .

13. The conjugate according to claim 1 , wherein (L) is a cleavable linker.
14. The conjugate according to claim 1 , wherein (L) is a cleavable linker comprising a self-immolative component.
15. The conjugate according to claim 14 , wherein the self-immolative component comprises p-aminobenzylcarbamoyl (PABC).
16. The conjugate according to claim 1 , wherein (L) comprises sulfosuccinimidyl 6-[3′(2-pyridyldithio)-propionamido]hexanoate (sulfo-LC-SPDP), succinimidyl 4[N-maleimidomethyl]cyclohexane-1-carboxylate (SMCC) or maleimidocaproyl-valine-citrulline-PABC (MC—VC-PABC).
17. The conjugate according to claim 1 , wherein (T)-(L)-(PT) has one of the following structures:
18. The conjugate according to claim 1 , wherein (T) is an antibody.
19. The conjugate according to claim 1 , wherein the antibody or antibody fragment is a bispecific antibody or antibody fragment, or a multispecific antibody or antibody fragment.
20. The conjugate according to claim 1 , wherein the antibody or antibody fragment specifically binds a cancer antigen.
21. A pharmaceutical composition comprising the conjugate according to claim 1 and a pharmaceutically acceptable carrier, diluent or excipient.
22. A method of inhibiting cancer, relieving cancer, or relieving the symptoms of cancer in a mammal comprising administering to a mammal in need thereof an effective amount of the conjugate according to claim 1 .
23. A method of inhibiting tumor growth in a mammal comprising administering to a mammal in need thereof an effective amount of the conjugate according to claim 1 .
