Abstract

Disclosed herein are polypeptides, polynucleotides encoding, cells and organisms comprising novel DNA-binding domains, including TALE DNA-binding domains. Also disclosed are methods of using these novel DNA-binding domains for modulation of gene expression and/or genomic editing of endogenous cellular sequences.

Claims (13)

1. An isolated polynucleotide encoding a non-naturally occurring transcription activator-like effector (TALE) DNA-binding polypeptide comprising:

two or more TALE-repeat units, the TALE repeat units comprising a repeat variable di-residue (RVD); and wherein the N-terminal region lacks at least 152 amino acids of a full-length TALE protein and the C-terminal region is truncated to residue C+63, and further wherein the polypeptide binds to DNA.

2. The polynucleotide of claim 1 , wherein at least one TALE-repeat unit comprises an atypical repeat variable di-residue (RVD).

The polynucleotide of claim 2 , wherein the polypeptide comprises an atypical RVD selected from AE, GR, TR, PR, LH, VG, RE, RG, RC, TA, AA, QR, LR, SR, GE, VR, NI, NN, CA, DA, EA, FA, GA, HA, IA, KA, LA, MA, NA, PA, QA, RA, SA, VA, WA, YA, AC, CC, DC, EC, FC, GC, HC, IC, KC, LC, MC, NC, PC, RC, SC, VC, WC, YC, AD, CD, DD, ED, FD, GD, HD, ID, KD, LD, MD, ND, PD, QD, RD, SD, TD, VD, WD, YD, AP, CP, DP, EP, FP, GP, HP, IP, KP, LP, MP, NP, PP, QP, RP, SP, TP, VP, WP, YP, AQ, CQ, DQ, EQ, FQ, GQ, HQ, IQ, KQ, LQ, MQ, NQ, PQ, QQ, RQ, SQ, TQ, VQ, WQ, YQ, AR, CR, DR, ER, FR, GR, HR, IR, KR, MR, NR, PR, RR, TR, VR, WR, YR, AH, CH, DH, EH, FH, GH, HH, IH, KH, LH, MH, NH, PH, QH, RH, SH, TH, VH, WH, YH, Al, CI, DI, EI, FI, GI, HI, IT, KI, LI, MI, PI, QI, RI, SI, TI, VI, WI, YI, AK, CK, DK, EK, FK, GK, HK, IK, KK, LK, MK, NK, PK, QK, RK, SK, TK, VK, WK, YK, AW, CW, DW, EW, FW, GW, HW, IW, KW, LW, MW, NW, PW, QW, RW, SW, TW, VW, WW, or YW.

4. The polynucleotide of claim 1 , wherein the N-terminal region of the polypeptide is truncated to residue N+134 to residue N+137.

5. The polynucleotide of claim 1 , wherein the C-cap of the polypeptide comprises a TALE repeat domain.

6. The polynucleotide of claim 1 , wherein the polypeptide further comprises a functional domain.

7. The polynucleotide of claim 6 , wherein the functional domain is a transcriptional activator or a transcriptional repressor.

8. The polynucleotide of claim 1 , wherein the functional domain comprises a nuclease domain.

9. The polynucleotide of claim 8 , wherein the nuclease domain is an endonuclease domain.

10. An isolated cell comprising the polynucleotide of claim 1 .

11. A pharmaceutical composition comprising the cell according to claim 10 .

12. A pharmaceutical composition comprising the polynucleotide of claim 1 .
