Recombinant virus products and methods for inhibition of expression of DUX4
The present invention relates to RNA interference-based methods for inhibiting the expression of the DUX4 gene, a double homeobox gene on human chromosome 4q35. Recombinant adeno-associated viruses of the invention deliver DNAs encoding microRNAs that knock down the expression of DUX4. The methods have application in the treatment of muscular dystrophies such as facioscapulohumeral muscular dystrophy.

A method of treating a subject suffering from a facioscapulohumeral muscular dystrophy associated with overexpression of DUX4 in the subject, the method comprising administering intramuscularly to the subject a recombinant adeno-associated virus comprising a DNA operably linked to a muscle specific control element, the DNA encoding a DUX4 miRNA comprising an miRNA antisense guide strand, wherein the miRNA antisense guide strand consists of the nucleotide sequence of

2. The method of claim 1 , wherein the recombinant adeno-associated virus lacks rep and cap genes.
3. The method of claim 1 , wherein the adenovirus recombinant adeno-associated virus (rAAV) is rAAV1, rAAV2, rAAV3, rAAV4, rAAV5, rAAV6, rAAV7, rAAV8, rAAV9, rAAV10 or rAAV11.
4. The method of claim 3 , wherein the rAAV is rAAV6.

A method of inhibiting DUX4 expression by a cell comprising the DUX4 gene, the method comprising administering to the cell a recombinant adeno-associated virus comprising a DNA operably linked to a tissue specific control element, the DNA encoding a DUX4 miRNA comprising an miRNA antisense guide strand, wherein the miRNA antisense guide strand consists of the nucleotide sequence of

7. The method of claim 5 , wherein the recombinant adeno-associated virus (rAAV) is rAAV1, rAAV2, rAAV3, rAAV4, rAAV5, rAAV6, rAAV7, rAAV8, rAAV9, rAAV10 or rAAV11.
8. The method of claim 7 , wherein the rAAV is rAAV6.
9. The method of claim 5 , wherein the cell is a muscle cell.
10. The method of claim 5 , wherein the tissue specific control element is muscle specific.
