Diphosphomevalonate decarboxylase variant and method for producing olefin compound by using the same

Abstract

Mutations involving amino acid substitution were introduced into various sites of diphosphomevalonate decarboxylase (MVD), thus preparing a large number of MVD variants. Then, the variants were each evaluated in terms of a catalytic activity for production of olefin compounds such as isoprene. As a result, it was found that substitution of glycine at position with a different amino acid resulted in improvement in the catalytic activity. In addition, it was found that the MVD in which arginine at position and threonine at position in addition to the position were further substituted with different amino acids, respectively, also had the high catalytic activity.

Claims (16)

1. A method for producing an olefin compound, the method comprising the step of reacting ATP and a substrate compound represented by the following Formula (1) in the presence of a diphosphomevalonate decarboxylase, wherein the diphosphomevalonate decarboxylase is a protein consisting of the amino acid sequence of SEQ ID NO: 2 in which the glycine at position 152 is mutated to a different amino acid, and in which no more than 40 amino acid residues of the amino acid sequence of SEQ ID NO: 2 are substituted, deleted, added, and/or inserted other than at position 152

wherein in the Formula (1), R 1 and R 2 each independently represent a hydrogen atom, an alkyl group having 1 to 10 carbon atoms, an alkenyl group having 2 to 15 carbon atoms, an aryl group having 6 to 20 carbon atoms, or a halogen atom, wherein the alkyl group and the alkenyl group may be each independently optionally substituted with a hydroxy group and/or a carboxy group, and at least one of R 1 and R 2 is a methyl group.

2. The production method according to claim 1 , wherein the reaction step is performed by culturing a host cell comprising a DNA encoding the diphosphomevalonate decarboxylase, or a vector comprising the DNA.

The production method according to claim 1 , wherein, in the diphosphomevalonate decarboxylase, the glycine at position 152 in the amino acid sequence of SEQ ID NO: 2 is substituted with cysteine, leucine, methionine, proline, glutamine, threonine, valine, arginine, lysine, tyrosine, isoleucine, or tryptophan.

4. The production method according to claim 1 , wherein, in the diphosphomevalonate decarboxylase, the arginine at position 74 in the amino acid sequence of SEQ ID NO: 2 is mutated to a different amino acid.

5. The production method according to claim 1 , wherein, in the diphosphomevalonate decarboxylase,

the arginine at position 74 in the amino acid sequence of SEQ ID NO: 2 is substituted to histidine, and

the threonine at position 209 in the amino acid sequence of SEQ ID NO: 2 is substituted with arginine.

6. The production method according to claim 1 , wherein:

the olefin compound is isoprene, and

the substrate compound is 3-hydroxy-3-methylpent-4-enotate.

7. The production method according to claim 1 , wherein:

the olefin compound is isobutene, and

the substrate compound is β-hydroxyisovaleric acid.

8. The production method according to claim 1 , wherein in the diphosphomevalonate decarboxylase, the glycine at position 152 in the amino acid sequence of SEQ ID NO: 2 is substituted with cysteine, leucine, methionine, proline, glutamine, or threonine.

9. The production method according to claim 1 , wherein in the diphosphomevalonate decarboxylase, the glycine at position 152 in the amino acid sequence of SEQ ID NO: 2 is substituted with cysteine, leucine, or methionine.
