Abstract

The present invention describes methods and compositions for the treatment of neurodegenerative conditions including Alzheimer's disease (AD) and aging. The present invention discloses an anaplerotic diet therapy for the treatment of AD. The high fat, low carbohydrate diet described in the various embodiments of the present invention increasing circulating ketone bodies, reduces amyloid-β (Aβ) deposition in AD patients, thereby improving cognition and locomotive skills.

31 . A method of treating Alzheimer's disease comprising orally administering to a subject with Alzheimer's disease a therapeutically effective amount of a composition, wherein the composition comprises a C7 fatty acid.

32 . The method of claim 31 , wherein the composition comprises triheptanoin.

33 . The method of claim 31 , wherein the therapeutically effective amount is 35% kcalories of the daily dietary caloric requirement for an adult.

34 . The method of claim 31 , wherein the therapeutically effective amount is 0.1-2 g/kg body weight of the subject.

35 . The method of claim 31 , wherein the therapeutically effective amount is sufficient to increase circulating levels of C5-ketone bodies in the subject's blood.

36 . The method of claim 35 , wherein the C5-ketone bodies are β-hydroxypentanoate and β-ketopentanoate.
