Methods and compositions for selecting siRNA of improved functionality
View Patent ↗Efficient sequence specific gene silencing is possible through the use of siRNA technology. Be selecting particular siRNAs by rational design, one can maximize the generation of an effective gene silencing reagent, as well as methods for silencing genes. Methods compositions, and kits generated through rational design of siRNAs are disclosed, including those directed to the nucleotide sequences for COX-2.
1. An RNA molecule capable of inducing the RNA interference pathway, wherein said RNA molecule consists of:
(a) a duplex region; and
(b) either no overhang regions or at least one overhang region, wherein each overhang region contains six or fewer nucleotides,
wherein the duplex region consists of a sense region and an antisense region, wherein said sense region and said antisense region together form said duplex region and said sense region and said antisense region are each 18-30 nucleotides in length, and said antisense region comprise a sequence of at least 18 contiguous bases wherein the sequence of at least 18 contiguous bases of the antisense region is complementary to at least 18 contiguous bases of SEQ ID NO: 502.
2. The RNA molecule of claim 1 , wherein said duplex region is 18-25 nucleotides in length.
3. The RNA molecule of claim 2 , wherein said antisense region and said sense region are each 25 nucleotides in length.
4. The RNA molecule of claim 2 , wherein said antisense region and said sense region are each 19 nucleotides in length.
5. The RNA molecule of claim 1 , wherein said RNA molecule has at least one overhang region.
6. The RNA molecule of claim 1 , wherein said RNA molecule has no overhang regions.
7. The RNA molecule of claim 2 , wherein said RNA molecule has at least one overhang region.
8. The RNA molecule of claim 2 , wherein said RNA molecule has no overhang regions.
9. The RNA molecule of claim 3 , wherein said RNA molecule has at least one overhang region.
10. The RNA molecule of claim 3 , wherein said RNA molecule has no overhang regions.
11. An RNA molecule capable of inducing the RNA interference pathway, wherein said RNA molecule consists of:
(a) a duplex region; and
(b) either no overhang regions or at least one overhang region, wherein each overhang region contains six or fewer nucleotides,
wherein the duplex region consists of a sense region and an antisense region, wherein said sense region and said antisense region together form said duplex region and said sense region and said antisense region are each 18-30 nucleotides in length, and said antisense region comprise a sequence of 18 bases, wherein the sequence of 18 bases is at least 90% complementary SEQ ID NO: 502.
12. The RNA molecule of claim 11 , wherein said duplex region is 18-25 nucleotides in length.
13. The RNA molecule of claim 12 , wherein said antisense region and said sense region are each 25 nucleotides in length.
14. The RNA molecule of claim 12 , wherein said antisense region and said sense region are each 19 nucleotides in length.
15. The RNA molecule of claim 11 , wherein said RNA molecule has at least one overhang region.
16. The RNA molecule of claim 11 , wherein said RNA molecule has no overhang regions.
17. The RNA molecule of claim 12 , wherein said RNA molecule has at least one overhang region.
18. The RNA molecule of claim 12 , wherein said RNA molecule has no overhang regions.
19. The RNA molecule of claim 13 , wherein said RNA molecule has at least one overhang region.
20. The RNA molecule of claim 13 , wherein said RNA molecule has no overhang regions.
