Abstract

The majority of the mortality observed in young pigs occurs between three and five weeks post-weaning for S. suis infections and between four and six weeks post-weaning for H. parasuis and Actinobacillus suis infections. Clinical disease control associated with S. suis, A. suis and H. parasuis has been attempted using antibiotic treatment, by controlled exposure with live organisms, and by vaccination, using either inactivated commercial or autogenous bacterins administered parenterally. A similar lack of protection in very young pigs is observed with various viruses including swine influenza virus, porcine reproductive and respiratory syndrome virus, porcine epidemic diarrhea virus and rotavirus. Disclosed herein is an immunogenic composition comprising inactivated antigens and a mucosal adjuvant. The composition may be administered to subjects, such as animals, particularly piglets from pre-weaning through the nursery phase, such as from birth or from three to five days of age, to protect from these diseases.

Claims (36)

1. A method, comprising:

administering a first composition mucosally to a mammal, the first composition being an aqueous solution comprising inactivated antigens obtained from a bacteria or virus, and a single adjuvant such that a concentration of adjuvant in the first composition is from 5% to 70% w/w, wherein the single adjuvant is a polyacrylic acid-based mucoadhesive adjuvant; and

parenterally administering to the mammal, a second composition comprising inactivated antigens obtained from the bacteria or virus.

2. The method of claim 1 , comprising administering the first composition intranasally.

3. The method of claim 1 , comprising administering the second composition intramuscularly, subcutaneously, or intraperitoneally.

4. The method of claim 1 , wherein the second composition is administered from two to six weeks after the first composition is administered.

5. The method of claim 1 , wherein the first composition is administered from zero to 10 days after birth of the subject.

6. The method of claim 1 , wherein the first composition and the second composition each comprise inactivated SIV antigens.

7. The method of claim 1 , wherein the first composition and the second composition each comprise inactivated PRRSV antigens.

8. The method of claim 1 , wherein the mammal is a pig.

9. The method of claim 1 , wherein the second composition comprises a non-mucoadhesive adjuvant.

10. The method of claim 1 , wherein the second composition comprises an oil-based adjuvant.

11. The method of claim 1 , wherein the method is a method of producing an immune response in a pig, comprising:

administering the first composition intranasally to the pig at from zero to 10 days old, the first composition comprising inactivated viral antigens; and

administering the second composition intramuscularly to the pig from two to six weeks after the first composition is administered, the second composition comprising an oil-based adjuvant and inactivated viral antigens;

wherein the inactivated viral antigens in the first and second compositions are inactivated PRRSV antigens, or the inactivated viral antigens in the first and second compositions are inactivated SIV antigens.

12. A method of vaccinating a pig, comprising:

administering a first immunogenic composition intranasally to the pig, the first immunogenic composition being an aqueous solution comprising inactivated antigens obtained from a virus, and a single adjuvant such that a concentration of adjuvant in the composition is from 5% to 70% w/w, wherein the single adjuvant is a polyacrylic acid-based mucoadhesive adjuvant; and

administering a second immunogenic composition intramuscularly or subcutaneously to the pig, the second immunogenic composition comprising inactivated antigens obtained from the virus, and a non-mucoadhesive adjuvant.

13. The method of claim 12 , wherein the non-mucoadhesive adjuvant is an oil-based adjuvant.

14. The method of claim 12 , wherein the second immunogenic composition is administered from two to six weeks after administration of the first immunogenic composition.

15. The method of claim 12 , wherein the inactivated viral antigens are inactivated PRRSV antigens.

16. The method of claim 12 , wherein the inactivated viral antigens are inactivated SIV antigens.

17. The method of claim 12 , comprising:

administering the first immunogenic composition to the pig, the first immunogenic composition comprising inactivated PRRSV antigens; and

administering the second immunogenic composition to the pig from two to six weeks after the first immunogenic composition is administered, the second immunogenic composition comprising inactivated PRRSV antigens and an oil-based adjuvant.

18. The method of claim 12 , comprising:

administering the first immunogenic composition to the pig, the first vaccine composition comprising inactivated SIV antigens; and

administering the second immunogenic composition to the pig from two to six weeks after the first immunogenic composition is administered, the second immunogenic composition comprising inactivated SIV antigens and an oil-based adjuvant.

19. A method of producing an immune response in a pig, comprising:

administering a first composition mucosally to a mammal, the first composition being an aqueous suspension comprising inactivated antigens obtained from a bacteria or virus, cell fragments, and a single adjuvant such that a concentration of adjuvant in the first composition is from 5% to 70% w/w, wherein the single adjuvant is a polyacrylic acid-based mucoadhesive adjuvant; and

parenterally administering to the mammal, a second composition comprising inactivated antigens obtained from the bacteria or virus.

20. The method of claim 19 , wherein the inactivated antigens are inactivated PRRSV antigens, inactivated SIV antigens, or a combination thereof.

21. The method of claim 19 , comprising:

administering the first composition intranasally to the pig at from zero to 10 days old, the first composition comprising inactivated viral antigens selected from inactivated PRRSV antigens, inactivated SIV antigens, or a combination thereof; and

administering the second composition intramuscularly to the pig from two to six weeks after the first composition is administered, the second composition comprising an oil-based adjuvant and inactivated viral antigens selected from inactivated PRRSV antigens, inactivated SIV antigens, or a combination thereof.
