Method of performing droplet-based assays
Method of performing a droplet-based assay. The method may include obtaining droplets encapsulated by an immiscible liquid and packed closely together in a monolayer, performing a reaction in the droplets while packed closely together in the monolayer; and collecting data related to an analyte from a plurality of the droplets while the droplets remain closely packed together in the monolayer.
1. A method of performing a droplet-based assay, the method comprising:
partitioning a sample-containing mixture to form obtaining droplets each encapsulated by an immiscible liquid;
collecting the droplets in a chamber such that the droplets are packed closely together in a two-dimensional arrangement as a monolayer, wherein the average separation between adjacent pairs of droplets in the monolayer is less than an average diameter of the droplets;
performing a reaction in the droplets while packed closely together in the monolayer; and
collecting data related to an analyte from a plurality of the droplets while the droplets remain closely packed together in the monolayer.
2. The method of claim 1 , wherein partitioning a sample-containing mixture includes forming monodisperse droplets.
3. The method of claim 1 , wherein performing a reaction includes heating the droplets.
4. The method of claim 3 , wherein heating the droplets includes thermally cycling the droplets.
5. The method of claim 4 , wherein thermally cycling the droplets includes amplifying one or more nucleic acid targets in the droplets, and wherein collecting data includes collecting data related to amplification of the one or more targets in individual droplets.
6. The method of claim 5 , further comprising processing the data to determine whether a target of the one or more targets is present or absent in individual droplets.
7. The method of claim 1 , wherein collecting data includes capturing an image of droplets in the monolayer.
8. The method of claim 1 , wherein capturing an image includes capturing a fluorescence image of droplets in the monolayer.
