Abstract

The invention provides antibodies that bind specifically to human indoleamine 2,3-dioxygenase 1 (IDO1), nucleic acids encoding the same, and methods of using the same. The antibodies are capable of binding to a sequence comprising SEQ ID NO: 1 and specifically bind to human IDO1 in formalin fixed paraffin embedded tissues. The antibodies are useful in a number of different analytical techniques, including immunohistochemistry (IHC) and immunocytochemistry (ICC).

Claims (37)

1. A method of detecting the presence or expression level of indoleamine 2,3-dioxygenase 1 (IDO1) in a biological sample, the method comprising contacting the biological sample with an anti-IDO1 antibody and detecting a presence of the antibody bound to the biological sample, wherein the anti-IDO1 antibody comprises the following heavy chain hypervariable regions (HVR-H) and light chain hypervariable regions (HVR-L):

(a) an HVR-H1 comprising the amino acid sequence of SEQ ID NO: 2;

(b) an HVR-H2 comprising the amino acid sequence of SEQ ID NO: 3;

(c) an HVR-H3 comprising the amino acid sequence of SEQ ID NO: 4;

(d) an HVR-L1 comprising the amino acid sequence of SEQ ID NO: 9;

(e) an HVR-L2 comprising the amino acid sequence of SEQ ID NO: 10; and

(f) an HVR-L3 comprising the amino acid sequence of SEQ ID NO: 11.

2. The method of claim 1 , wherein the anti-IDO1 antibody further comprises the following heavy chain variable domain framework regions (FR-H):

(g) FR-H1 comprising the amino acid sequence of SEQ ID NO: 5 or SEQ ID NO: 19;

(h) FR-H2 comprising the amino acid sequence of SEQ ID NO: 6;

(i) FR-H3 comprising the amino acid sequence of SEQ ID NO: 7 or SEQ ID NO: 20; and

(j) FR-H4 comprising the amino acid sequence of SEQ ID NO: 8.

3. The method of claim 1 , wherein the anti-IDO1 antibody further comprises the following light chain variable domain framework regions (FR-L):

(k) FR-L1 comprising the amino acid sequence of SEQ ID NO: 12;

(l) FR-L2 comprising the amino acid sequence of SEQ ID NO: 13;

(m) FR-L3 comprising the amino acid sequence of SEQ ID NO: 14; and

(n) FR-L4 comprising the amino acid sequence of SEQ ID NO: 15.

4. The method of claim 1 , wherein the anti-IDO1 antibody further comprises the following heavy chain variable domain and light chain variable domain framework regions (FR):

(a) FR-H1 comprising the amino acid sequence of SEQ ID NO: 5;

(b) FR-H2 comprising the amino acid sequence of SEQ ID NO: 6;

(c) FR-H3 comprising the amino acid sequence of SEQ ID NO: 7;

(d) FR-H4 comprising the amino acid sequence of SEQ ID NO: 8;

(e) FR-L1 comprising the amino acid sequence of SEQ ID NO: 12;

(f) FR-L2 comprising the amino acid sequence of SEQ ID NO: 13;

(g) FR-L3 comprising the amino acid sequence of SEQ ID NO: 14; and

(h) FR-L4 comprising the amino acid sequence of SEQ ID NO: 15.

5. The method of claim 1 , wherein the sample is from a subject having or predisposed to cancer or autoimmune disease.

6. The method of claim 1 , wherein the anti-IDO1 antibody is a monoclonal antibody.

7. The method of claim 6 , wherein the monoclonal antibody is a rabbit monoclonal antibody.

8. The method of claim 1 , wherein the detecting comprises immunohistochemistry (IHC) or immunocytochemistry (ICC).

9. The method of claim 8 , wherein the sample comprises a fixed tissue.

10. The method of claim 9 , wherein the fixed tissue is a formalin fixed paraffin embedded (FFPE) tissue.

11. The method of claim 1 , wherein the anti-IDO1 antibody comprises a variable domain of the heavy chain (V H ) sequence having at least 95% sequence identity to the amino acid sequence of SEQ ID NO: 16.

12. The method of claim 11 , wherein the V H sequence comprises SEQ ID NO: 16.

13. The method of claim 12 , wherein the V L sequence comprises SEQ ID NO: 17.

14. The method of claim 11 , wherein the anti-IDO1 antibody further comprises a variable domain of the light chain (VL) sequence having at least 95% sequence identity to the amino acid sequence of SEQ ID NO: 17.

15. The method of claim 14 , wherein the V L sequence comprises SEQ ID NO: 17.
