Abstract

The present disclosure provides nucleic acids and vectors for use with methanotrophic bacteria. Related host cells and methods for using such nucleic acids and vectors for expressing polypeptides or other genetic manipulation of methanotrophic bacteria are also provided.

Claims (28)

1. A non-naturally occurring polynucleotide, comprising:

(a) a regulated promoter operably linked to a first ribosomal binding sequence and a nucleic acid molecule encoding a target protein, wherein the regulated promoter is functional in a methanotrophic bacterium; and

(b) a constitutive promoter operably linked to a second ribosomal binding sequence and a nucleic acid molecule encoding an activator protein or a repressor protein, wherein the constitutive promoter is functional in a methanotrophic bacterium,

wherein the first and second ribosomal binding sequences are each an altered methanol dehydrogenase (MDH) ribosomal binding sequence comprising the polynucleotide sequence as set forth in any one of SEQ ID NOS:20, 22, 24, 26, 27, 32, and 41, or a native MDH ribosomal binding sequence.

2. The non-naturally occurring polynucleotide of claim 1 , wherein the regulated promoter comprises an operator of the lac operon, and the nucleic acid molecule encoding an activator protein or a repressor protein encodes a LacI protein.

3. The non-naturally occurring polynucleotide of claim 1 , wherein the regulated promoter comprises an MDH promoter flanked by two or more operators of the lac operon, and the nucleic acid molecule encoding an activator protein or a repressor protein encodes a LacI protein.

The non-naturally occurring polynucleotide of claim 1 , wherein the constitutive promoter comprises an MDH promoter, a 30S ribosomal protein S16 promoter, a 30S ribosomal protein S12 promoter, a 50S ribosomal protein L13 promoter, a hexulose 6-phosphate synthase promoter, a T5 promoter, or a Trc promoter.

5. The non-naturally occurring polynucleotide of claim 4 , wherein the constitutive promoter comprises:

(a) an MDH promoter sequence from the promoter portion of SEQ ID NO.:14;

(b) a 30S ribosomal protein S16 promoter sequence from the promoter portion of SEQ ID NO.:15;

(c) a 30S ribosomal protein S12 promoter sequence from the promoter portion of SEQ ID NO.:43;

(d) a 50S ribosomal protein L13 promoter sequence from the promoter portion of SEQ ID NO.:44;

(e) a hexulose 6-phosphate synthase promoter sequence from the promoter portion of SEQ ID NO.:16;

(f) a T5 promoter sequence from the promoter portion of SEQ ID NO.:17; or

(g) a Trc promoter sequence from the promoter portion of SEQ ID NO.:18.

6. The non-naturally occurring polynucleotide of claim 1 , wherein the target protein comprises:

(a) a marker or reporter protein;

(b) a protein capable of improving production of a desired chemical or metabolite; or

(c) an enzyme.

The non-naturally occurring polynucleotide of claim 1 , wherein the regulated promoter comprises an MDH promoter flanked by two or more operators of the lac operon; the first ribosomal binding sequence comprises an altered MDH ribosomal binding sequence or native MDH ribosomal binding sequence; the constitutive promoter comprises a 30S ribosomal protein S16 promoter; the second ribosomal binding sequence comprises an altered MDH ribosomal binding sequence or native MDH ribosomal binding sequence; and the encoded activator protein or repressor protein comprises a LacI protein.

8. The non-naturally occurring polynucleotide of claim 1 , wherein the regulated promoter comprises a promoter region of the benzoate metabolic operon, and the nucleic acid molecule encoding an activator protein or a repressor protein encodes a BenR protein.

9. A vector comprising the non-naturally occurring polynucleotide of claim 1 .

10. A host cell comprising the non-naturally occurring polynucleotide of claim 1 .

11. The host cell of claim 10 , wherein the non-naturally occurring polynucleotide is integrated into the genome of the host cell.

12. The host cell of claim 10 , wherein the non-naturally occurring polynucleotide is present in a vector.

13. The host cell of claim 10 , wherein the host cell is Methylococcus capsulatus Bath or Methylosinus trichosporium OB3b.

14. A method for producing a target protein, comprising culturing the host cell of claim 10 under conditions and for a time sufficient to express the target protein.

The non-naturally occurring polynucleotide of claim 7 , wherein the regulated promoter comprises an MDH promoter flanked by two or more operators of the lac operon; the first ribosomal binding sequence comprises a native MDH ribosomal binding sequence; the constitutive promoter comprises a 30S ribosomal protein S16 promoter; the second ribosomal binding sequence comprises a native MDH ribosomal binding sequence; and the encoded activator protein or repressor protein comprises a LacI protein.
