Abstract

The present invention provides for methods and devices suitable for producing a transplantable cellular suspension of living tissue suitable for promoting tissue regeneration in an epithelium-related procedure, as well as compositions produced therefrom. The cellular suspension can include viable and functioning cells at various stages of differentiation, including undifferentiated/progenitor cells and differentiated cells, as well as those in between. In certain embodiments, the cellular suspension can be subjected to a stress to induce a heat shock response therein, or be exposed to an exogenously supplied agent such as heat shock protein or a fragment thereof, hyaluronic acid, platelet-enriched plasma, and/or growth factors. The cellular suspension can be applied directly to a patient's recipient site for in vivo regeneration, or be cultured or seeded to a matrix for in vitro growth/regeneration.

50 . A method of preparing a cell suspension for tissue regeneration, the method comprising:

collecting a population of cells from a skin tissue sample; and

exposing the population of cells to platelet-enriched plasma.

51 . The method of claim 50 , wherein the skin tissue sample comprises an autologous epithelial tissue sample.

52 . The method of claim 50 , wherein the population of cells comprises viable and functioning cells from the skin tissue sample.

53 . The method of claim 52 , wherein the population of cells comprises keratinocytes, melanocytes, and fibroblasts.

54 . The method of claim 50 , wherein collecting the population of cells comprises exposing the skin tissue sample to an enzyme to dissociate cellular layers in the skin tissue sample.

55 . The method of claim 54 , wherein the enzyme comprises at least one enzyme selected from the group consisting of trypsin, trypsin-EDTA, dispase, collagenase, thermolysin, pronase, hyaluronidase, elastase, papain, and pancreatin.

56 . The method of claim 50 , wherein exposing the population of cells to platelet-enriched plasma comprises suspending the population of cells in a nutrient solution comprising platelet-enriched plasma.

57 . The method of claim 56 , wherein the nutrient solution comprises at least one of Hartmann's solution and physiological saline.

58 . The method of claim 50 , wherein the cell suspension is prepared without in vitro culturing.

59 . A method of promoting tissue regeneration, the method comprising:

providing a cell suspension comprising a population of cells from a skin tissue sample and platelet-enriched plasma; and

applying the cell suspension to a recipient site of a patient.

60 . The method of claim 59 , wherein the recipient site of the patient is designated for at least one of a skin wound treatment, a skin tissue graft, and a skin aesthetic procedure.

61 . The method of claim 60 , wherein the recipient site of the patient is designated for a wound treatment, wherein the wound treatment comprises at least one of a treatment of a burn wound, a laceration wound, a graft donor site, and a chronic skin wound

The method of claim 60 , wherein the recipient site of the patient is designated for a skin aesthetic procedure, wherein the skin aesthetic procedure comprises at least one of a scar remodeling procedure, treatment for a glabrous skin loss injury, treatment for vitiligo or other pigmentation condition, treatment for leukoderma, and a cosmetic rejuvenation procedure.

63 . The method of claim 59 , wherein applying the cell suspension comprises at least one of spraying, spreading, dripping, pipetting, painting, and injecting the cell suspension.

64 . The method of claim 59 , wherein the skin tissue sample comprises an autologous epithelial tissue sample.

65 . The method of claim 59 , wherein the cell suspension comprises the population of cells and platelet-enriched plasma suspended in a nutrient solution.

66 . The method of claim 65 , wherein the nutrient solution comprises at least one of Hartmann's solution and physiological saline.

67 . The method of claim 59 , wherein the cell suspension is prepared without in vitro culturing.

68 . A cell suspension for use in tissue regeneration, the cell suspension comprising:

a population of cells collected from a skin tissue sample; and

platelet-enriched plasma.

69 . The cell suspension of claim 68 , wherein the skin tissue sample comprises an autologous epithelial tissue sample.

70 . The cell suspension of claim 68 , wherein the population of cells comprises viable and functioning cells from the skin tissue sample.

71 . The cell suspension of claim 70 , wherein the population of cells comprises keratinocytes, melanocytes, and fibroblasts.

72 . The cell suspension of claim 68 , wherein the population of cells is suspended in a nutrient solution comprising the exogenous agent.

73 . The cell suspension of claim 72 , wherein the nutrient solution comprises at least one of Hartmann's solution and physiological saline.
