Abstract

Provided herein are binding polypeptides that specifically bind PD-1. More specifically, provided herein are fusion proteins, including multivalent and/or multispecific constructs and chimeric antigen receptors, that bind PD-1. Also provided are pharmaceutical compositions containing the polypeptides, nucleic acid molecules encoding the polypeptides and vectors and cells thereof, and methods of use and uses of the provided PD-1 binding polypeptides for treating diseases and conditions, such as cancer.

Claims (36)

1. A PD-1-binding polypeptide construct, comprising at least one heavy chain only variable domain (PD-1 VHH domain) that specifically binds PD-1, wherein the at least one VHH domain comprises a CDR1, CDR 2 and CDR 3 comprising:

the amino acid sequence set forth in SEQ ID NOs: 268, 278 and 283, respectively;

the amino acid sequence set forth in SEQ ID NOs: 272, 278 and 283, respectively;

the amino acid sequence set forth in SEQ ID NOs: 313, 314 and 315, respectively; or

the amino acid sequence set forth in SEQ ID NOs: 273, 278 and 283, respectively.

2. The PD-1-binding polypeptide construct of claim 1 , comprising at least one heavy chain only variable domain (PD-1 VHH domain) that specifically binds PD-1 and comprises one or more additional binding domain that binds to a target other than PD-1.

3. The PD-1-binding polypeptide construct of any of claim 1 or 2 , wherein the PD-1 VHH domain that specifically binds PD-1, blocks the interaction of PD-1 and PD-L 1.

4. The PD-1-binding polypeptide construct of claim 2 , wherein the one or more additional binding domain binds a co-stimulatory molecule.

5. The PD-1-binding polypeptide construct of claim 2 , wherein the one or more additional binding domain binds a T-cell surface marker.

6. The PD-1-binding polypeptide construct of claim 5 , wherein the T-cell surface marker is CD8.

7. The PD-1-binding polypeptide construct of claim 5 , wherein the T-cell surface marker is CD4.

8. The PD-1-binding polypeptide construct of claim 2 , wherein the one or more additional binding domain binds an immune checkpoint other than PD1.

9. The PD-1-binding polypeptide construct of claim 2 , wherein the one or more additional binding domains binds to an activating receptor on an immune cell.

10. The PD-1-binding polypeptide construct of claim 2 , wherein the one or more additional domains binds to a tumor associated antigen (TAA).

11. The PD-1-binding polypeptide construct of claim 2 , wherein the one or more additional binding domain binds to a cytokine receptor.

12. The PD-1-binding polypeptide construct of any of claims 1 or 2 wherein the polypeptide comprises an immunoglobulin Fc region.

The PD-1-binding polypeptide construct of any of claims 1 or 2 wherein the at least one PD-1 VHH domain comprises the amino acid sequence set forth in any of SEQ ID NOS: 251-267, 284 or 312, or a sequence of amino acids that exhibits at least 85%, 86%, 87%, 88%, 89%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, or 99% sequence identity to amino acid sequence set forth in any of SEQ ID NOS: 251-267, 284 or 312 and binds PD-1.

14. An isolated single domain antibody that binds PD-1, comprising a complementarity determining region 1 (CDR1); a complementarity determining region 2 (CDR2); and a complementarity determining region 3 (CDR3) comprising:

the amino acid sequence set forth in SEQ ID NOs: 268, 278 and 283, respectively;

the amino acid sequence set forth in SEQ ID NOs: 272, 278 and 283, respectively;

the amino acid sequence set forth in SEQ ID NOs: 313, 314 and 315, respectively; or

the amino acid sequence set forth in SEQ ID NOs: 273, 278 and 283, respectively.

The isolated single domain antibody of claim 14 , comprising the amino acid sequence set forth in any of SEQ ID NOS: 251-267 or 284, or a sequence of amino acids that exhibits at least 85%, 86%, 87%, 88%, 89%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, or 99% sequence identity to amino acid sequence set forth in any of SEQ ID NO: 251-267, 284 or 312 and binds PD-1.

16. The isolated single domain antibody of claim 15 , wherein the sdAb comprises the sequence of amino acid sequence set forth in any of SEQ ID NO: 251, 252, 253, 254, 255, 256, 257, 258, 259, 260, 261, 262, 263, 264, 265, 266, 267, 284 or 312.

17. A polynucleotide encoding the PD-1-binding polypeptide of claim 1 .

18. A polynucleotide encoding the single domain antibody of claim 14 .

19. A vector, comprising the polynucleotide of claim 17 .

20. A cell, comprising a polynucleotide of claim 17 .

21. A method of producing a polypeptide construct, the method comprising introducing into a cell a polynucleotide of claim 17 and culturing the cell under conditions to produce the polypeptide construct.

22. An engineered immune cell, comprising a binding molecule comprising the PD-1-binding polypeptide construct binding molecule of claim 1 , optionally wherein the binding molecule is secretable from the cell.

23. A pharmaceutical composition comprising the PD-1-binding polypeptide construct of claim 1 .

24. A cell, comprising a vector of claim 19 .

25. A method of producing a polypeptide, the method comprising introducing into a cell a vector of claim 19 and culturing the cell under conditions to produce the polypeptide.

26. An engineered immune cell, comprising a binding molecule comprising the single domain antibody of claim 14 , optionally wherein the binding molecule is secretable from the cell.

27. A pharmaceutical composition comprising the single domain antibody of claim 14 .

28. A pharmaceutical composition comprising the engineered immune cell of claim 22 .
