Abstract

The invention generally relates to a microfluidic platforms or “chips” for testing and conducting experiments on the International Space Station (ISS). More specifically, microfluidic Brain-On-Chip, comprising neuronal and vascular endothelial cells, will be analyzed in both healthy and inflamed states to assess how the circumstances of space travel affect the human brain.

Claims (19)

1. A method of recirculating culture media through microfluidic devices, comprising:

1) providing an assembly, comprising

a) one or more reservoirs comprising cell culture media in fluidic communication with one or more microfluidic devices, said one or more microfluidic devices comprising

i) first and second channels attached mammalian cells on a surface of said first or second channels and

ii) inlet and outlet ports associated with said first and second channels, said inlet and outlet ports in fluidic communication with a recirculation pathway, said cells perfused with fluid; and

2) causing culture media in said one or more reservoirs to flow into said one or more microfluidic devices, thereby causing fluid to exit as effluent from said outlet port of said first or second channels of one or more microfluidic devices, enter said recirculation pathway and directly enter an inlet port of said first or second channel, so that said effluent becomes incoming fluid.

2. The method of claim 1 , wherein said surface is a microchannel.

3. The method of claim 1 , wherein said surface is a membrane.

4. A method of recirculating culture media through a microfluidic device, comprising:

a) providing an assembly, comprising

i) an inlet reservoir and ii) an outlet reservoir connected by iii) a shortcut channel, said shortcut channel containing iv) a single valve, and said inlet and outlet reservoirs in fluidic communication with v) a microfluidic device, said inlet reservoir comprising fluid, said microfluidic device comprising inlet and outlet ports, one of said ports linked to one of said reservoirs through a resistor; and

b) causing fluid in said inlet reservoir to flow into said inlet port of said microfluidic device while said valve is closed, thereby causing fluid to exit said outlet port of said one or more microfluidic devices and enter said outlet reservoir as recirculated fluid;

c) causing said valve to open, thereby permitting said recirculated fluid in said outlet reservoir to move through said shortcut channel to said inlet reservoir; and

d) causing said valve to close;

e) flowing said recirculated fluid in said inlet reservoir into said inlet port of said microfluidic device.

5. The method of claim 4 , wherein said fluid is moved in step b) with pressure.

6. The method of claim 4 , wherein said single valve is a one-way check valve.

7. The method of claim 4 , wherein the majority of fluid in step c) moves into the inlet reservoir through the open check valve as the path of least resistance.

8. The method of claim 4 , wherein said microfluidic device further comprises cells.
