Abstract

An optical system for acquiring fast spectra from spatially channel arrays includes a light source for producing a light beam that passes through the microfluidic chip or the channel to be monitored, one or more lenses or optical fibers for capturing the light from the light source after interaction with the particles or chemicals in the microfluidic channels, and one or more detectors. The detectors, which may include light amplifying elements, detect each light signal and transducer the light signal into an electronic signal. The electronic signals, each representing the intensity of an optical signal, pass from each detector to an electronic data acquisition system for analysis. The light amplifying element or elements may comprise an array of phototubes, a multianode phototube, or a multichannel plate based image intensifier coupled to an array of photodiode detectors.

Claims (21)

1. A method for processing particles, comprising:

flowing particles through a detection region of each of a plurality of microchannels in a microfluidic chip;

illuminating the particles with an electromagnetic radiation source using two or more light beams of same wavelength;

capturing an optical signal from the detection region of at least one of the plurality of microchannels using a lens of an optical system;

separating the optical signal into a plurality of color signals corresponding to one or more characteristics of the particles using a spectral separation element of the optical system; and

detecting the plurality of color signals using an array of detectors, each detector in the array disposed along an optical path associated with a color signal in the plurality of color signals.

2. The method of claim 1 , further comprising

transmitting a first color signal in the plurality of color signals to a first detector in the array of detectors using a first optical fiber; and

transmitting a second color signal in the plurality of color signals to a second detector in the array of detectors using a second optical fiber.

3. The method of claim 1 , wherein the spectral bandwidth of a first color signal in the plurality of color signals is approximately 10 nm.

4. The method of claim 1 , wherein separating the optical signal includes using a spectral dispersive element to receive the optical signal from the lens; the method further comprising:

transmitting each color signal of the plurality of color signals using a respective optical fiber of the plurality of optical fibers, the plurality of optical fibers being operatively associated with the spectral dispersive element.

5. The method of claim 1 , further comprising masking each of the two or more illuminating light beams using a mask before the light beams reach the microfluidic chip.

6. The method of claim 1 , wherein the lens is a high numerical aperture lens.

7. The method of claim 1 , wherein the array of detectors is an array of fluorescence detectors and detecting the plurality of color signals includes detecting fluorescence color signals.

8. The method of claim 1 , further comprising receiving optical signals associated with each of the plurality of microchannels using the optical system.

9. The method of claim 1 , further comprising filtering each color signal in the plurality of color signals using a spectral filter disposed in the optical path associated with the color signal.

10. The method of claim 9 , wherein filtering includes passing wavelength bands or blocking wavelength bands using the spectral filter.

11. The method of claim 1 , wherein separating the optical signal includes bending each of the plurality of color signals in a manner related to the wavelength of the particular color signal using the spectral separation element.

12. The method of claim 1 , further comprising detecting an optical extinction signal from the detection region using an extinction light detector.

13. The method of claim 1 , wherein separating the optical signal includes directing a first color signal of the plurality of color signals along a first path and directing a second color signal of the plurality of color signals along a second path.
