Abstract

Provided herein are liposomal vesicles comprising at least a first lipid bilayer and a second lipid bilayer, and a plurality of crosslinkages between the first lipid bilayer and the second lipid bilayer, wherein the plurality of crosslinkages comprise boronic ester or thioketal bonds. Also provided are pharmaceutical compositions comprising the liposomal vesicles described herein and a pharmaceutically acceptable excipient. Also provided are methods of making and using the liposomal vesicles. Thus, a method of treating a subject with a disease comprising administering to the subject a pharmaceutical composition comprising the liposomal vesicles is described herein. Methods of making multilamellar liposomal vesicles responsive to reactive oxygen species are also provided.

Claims (29)

1. A method of administering a pharmaceutical composition to a subject, the pharmaceutical composition comprising:

a liposomal vesicle comprising:

a first lipid bilayer and a second lipid bilayer,

a plurality of reversible crosslinkages between the first lipid bilayer and the second lipid bilayer, wherein:

each crosslinkage of the plurality of reversible crosslinkages comprises boronic ester bonds,

each crosslinkage is formed using a crosslinker,

the crosslinker comprises two terminal ends, and

each terminal end of the two terminal ends comprises a boronate center, and

one or more active agents; and

a pharmaceutically acceptable excipient.

2. The method of claim 1 , wherein the one or more active agents are encapsulated within the liposomal vesicle.

3. The method of claim 2 , further comprising releasing the one or more active agents in the presence of reactive oxygen species.

4. The method of claim 1 , wherein the pharmaceutical composition comprises a plurality of liposomal vesicles.

5. The method of claim 1 , wherein the subject has a disease characterized by production of reactive oxygen species.

6. The method of claim 5 , wherein the disease characterized by production of reactive oxygen species is cancer, an inflammatory disease, or a neurodegenerative disease.

7. The method of claim 6 , wherein the liposomal vesicle targets inflammatory macrophages.

8. The method of claim 1 , wherein each crosslinkage of the plurality of reversible crosslinkages is reversed in the presence of reactive oxygen species.

9. The method of claim 1 , wherein the crosslinker comprises an aryl boronic acid.

10. The method of claim 1 , wherein each crosslinkage comprises two boronic ester bonds.

11. The method of claim 1 , wherein administering to the subject the pharmaceutical composition comprises delivering the pharmaceutical composition intranasally.

12. The method of claim 1 , wherein administering to the subject the pharmaceutical composition comprises delivering the pharmaceutical composition orally.

13. The method of claim 1 , wherein administering to the subject the pharmaceutical composition comprises delivering the pharmaceutical composition parenterally.

14. The method of claim 6 , wherein the disease is cancer.

15. The method of claim 6 , wherein the disease is the inflammatory disease.

16. The method of claim 15 , wherein the one or more active agents comprise an anti-inflammatory agent.

17. The method of claim 6 , wherein the disease is the neurodegenerative disease.

18. The method of claim 1 , wherein the crosslinker comprises a phenylboronate.

19. The method of claim 1 , wherein the crosslinker comprises a pyridylboronate.

20. The method of claim 1 , wherein the crosslinker comprises a cyclohexylboronate.
