Abstract

According to various embodiments of this disclosure, pharmaceutical formulations comprising solubilized estradiol are provided. In various embodiments, such formulations are encapsulated in soft capsules which may be vaginally inserted for the treatment of vulvovaginal atrophy.

Claims (26)

1. A method for the treatment of one or more symptoms of vulvovaginal atrophy (VVA) comprising:

administering a vaginal suppository intravaginally twice weekly to a female subject having VVA, the vaginal suppository comprising a liquid composition comprising about 1 μg to about 25 μg 17β-estradiol and a solubilizing agent comprising a medium chain oil;

wherein the vaginal suppository further comprises a gelatin capsule comprising a gel mass, wherein the gel mass is selected from the group consisting of:

(i) Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, GELITA (hydrolyzed collagen) 3.0% w/w, and Purified Water 35.0% w/w;

(ii) Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, citric acid 0.1% w/w, and Purified Water 37.9% w/w;

(iii) Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, citric acid 0.5% w/w, and Purified Water 37.5% w/w;

(iv) Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, citric acid 1% w/w, and Purified Water 37.0% w/w;

(v) Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 18.0% w/w, GELITA (hydrolyzed collagen) 3.0% w/w, and Purified Water 36.0% w/w; and

(vi) Gelatin, NF (150 Bloom, Type B) 43.0% w/w, Glycerin 99.7%, USP 18.0% w/w, citric acid 0.1% w/w, and Purified Water 38.9% w/w; and

wherein the gel mass further comprises a colorant;

wherein the viscosity of the liquid composition is from about 50 cps to about 1000 cps at 25° C.;

wherein the twice weekly administration follows two weeks of once daily administration of the vaginal suppository;

wherein 17β-estradiol is the only active hormone in the vaginal suppository.

2. The method of claim 1 , wherein the medium chain oil comprises at least one C6-C12 fatty acid or a monoglyceride, diglyceride, or triglyceride ester thereof.

3. The method of claim 1 , wherein the liquid composition further comprises a thickening agent.

4. The method of claim 1 , wherein the gel mass comprises the following components: Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, GELITA (hydrolyzed collagen) 3.0% w/w, and Purified Water 35.0% w/w.

5. The method of claim 1 , wherein the gel mass comprises the following components: Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, citric acid 0.1% w/w, and Purified Water 37.9% w/w.

6. The method of claim 1 , wherein the gel mass comprises the following components: Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, citric acid 1 w/w, and Purified Water 37.0% w/w.

7. The method of claim 1 , wherein the gel mass comprises the following components: Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 6.0% w/w, Sorbitol Special, USP 15.0% w/w, citric acid 1 w/w, and Purified Water 37.0% w/w.

8. The method of claim 1 , wherein the gel mass comprises the following components: Gelatin, NF (150 Bloom, Type B) 41.0% w/w, Glycerin 99.7%, USP 18.0% w/w, GELITA (hydrolyzed collagen) 3.0% w/w, and Purified Water 36.0% w/w.

9. The method of claim 1 , wherein the gel mass comprises the following components: Gelatin, NF (150 Bloom, Type B) 43.0% w/w, Glycerin 99.7%, USP 18.0% w/w, citric acid 0.1% w/w, and Purified Water 38.9% w/w.

10. The method of any one of claims 4 - 9 , wherein the gel mass completely dissolves in about 42 to about 76 minutes when measured in water using a USP type II dissolution vessel at 50 rpm and at a temperature of about 37° C.

11. The method of claim 5 , wherein the gel mass completely dissolves in about 48 minutes.

12. The method of claim 5 , wherein the gel mass completely dissolves in about 70 to about 84 minutes when measured in a buffer at pH 4 using a USP type II dissolution vessel at 50 rpm and at a temperature of about 37° C.

13. The method of claim 6 , wherein the gel mass completely dissolves in about 70 minutes.

14. The method of claim 1 , wherein the symptoms of VVA are selected from the group consisting of vaginal dryness, dyspareunia, vaginal or vulvar irritation, burning or itching, dysuria, and vaginal bleeding associated with sexual activity.
