Abstract

Methods, systems and techniques for the accurate measurement of breath-borne biomarkers are disclosed. Such methods, systems and techniques may be used for the purposes of detection and/or measurement in breath samples of biomarkers, e.g., biomarkers relating to a disease or a physiological condition.

Claims (37)

1. A point of care biomarker detection device, comprising:

at least one impactor configured to capture, at an impaction surface, a plurality of aerosol droplets contained in an exhaled breath sample from a subject via impaction;

a microfluidic plate that is fluidly coupled to the at least one impactor to receive a sample of the plurality of aerosol droplets, the microfluidic plate including at least one reaction channel having an acetone-specific reagent immobilized thereon; and

one or more processors that are operable to:

detect, at the microfluidic plate in fluid communication with the at least one impactor, an amount of an indicator biomarker comprising acetone from the captured plurality of aerosol droplets, the indicator biomarker associated with an existence of a physiological condition in the subject; and

determine, based on the detected amount of indicator biomarker exceeding a threshold concentration level of acetone, the existence of the physiological condition in the subject.

2. The device of claim 1 , wherein the one or more processors are further configured to detect an amount of a reference biomarker in the captured aerosol droplets.

3. The device of claim 2 , wherein the one or more processors are further configured to: determine the existence of the physiological condition in the subject based at least in part on a comparison of the detected indicator biomarker and an amount of the reference biomarker.

4. The device of claim 1 , wherein the detected amount of indicator biomarker is detected using one or more of a chemiluminescence assay, an immunoassay, an enzymatic assay, and electrochemical detection.

5. The device of claim 1 , wherein the detected amount of indicator biomarker is detected via an optical detection means.

6. The device of claim 1 , wherein the point of care device is portable.

7. The device of claim 1 , wherein the physiological condition is diabetes.

8. A point of care device to detect an existence of a physiological condition in a subject, the device comprising:

at least one impactor configured to capture, at an impaction surface, a plurality of aerosol droplets contained in an exhaled breath sample from a subject via impaction;

a microfluidic plate that is fluidly coupled to the at least one impactor to receive a sample of the plurality of aerosol droplets, the microfluidic plate including at least one reaction channel having an acetone-specific reagent immobilized thereon; and

one or more processors that are operable to:

measure an amount of an indicator biomarker in the captured plurality of aerosol droplets from the subject, the indicator biomarker comprising acetone and being associated with existence of a physiological condition in the subject;

measure an amount of a reference biomarker in the captured plurality of aerosol droplets; and

determine the existence of the physiological condition in the subject based at least in part upon the amount of the indicator biomarker exceeding the amount of the reference biomarker.

9. The device of claim 8 , wherein the point of care device is portable.

10. The device of claim 8 , wherein the measured amount of indicator biomarker is detected using one or more of a chemiluminescence assay, an immunoassay, an enzymatic assay, and electrochemical detection.

11. The device of claim 8 , wherein the physiological condition is diabetes.

12. A method for detecting a presence of a physiological condition in a human at a point of care, the method comprising:

separating a plurality of aerosol droplets from a captured breath sample from the human via at least one impactor;

detecting, with a microfluidic plate an amount of a target chemical comprising acetone that is present in the separated aerosol droplets, wherein the target chemical is associated with an existence of a physiological condition in the human, wherein the microfluidic plate is fluidly coupled to the at least one impactor to receive a sample of the plurality of aerosol droplets, and wherein the microfluidic plate includes at least one reaction channel having an acetone-specific reagent immobilized thereon; and

determining, based on the amount of the target chemical being detected, the presence of the physiological condition.

13. The method of claim 12 , wherein the target chemical comprising acetone is a biomarker or a chemical indicative of a physiological condition related to diabetes.

14. The method of claim 12 , further comprising detecting an amount of a reference biomarker in the separated aerosol droplets.

15. A method for determining an existence of a physiological condition in a subject at a point of care of the subject, the method comprising:

non-invasively capturing a biological sample from a subject;

separating a plurality of aerosol droplets included with the biological sample via at least one impactor;

measuring, with a microfluidic plate, an amount of an indicator biomarker comprising acetone in the separated aerosol droplets, wherein the indicator biomarker is associated with an existence of a physiological condition in the human, wherein the microfluidic plate is fluidly coupled to the at least one impactor to receive a sample of the plurality of droplets, and wherein the microfluidic plate includes at least one reaction channel having an acetone-specific reagent immobilized thereon;

measuring an amount of a reference biomarker in the separated aerosol droplets, the reference biomarker known to exist at a stable concentration in the subject; and

determining, based at least in part on the amount of the indicator biomarker exceeding the amount of the reference biomarker, the existence of the physiological condition in the subject.

16. The method of claim 15 , wherein measuring the amount of the indicator biomarker in the separated aerosol droplets includes measuring a plurality of indicator biomarkers comprising acetone associated with the existence of a plurality of physiological conditions in the subject.

17. The method of claim 15 , wherein the biological sample is an exhaled breath sample.

18. The method of claim 15 , wherein the measuring is conducted using one or more of a chemiluminescence assay, an immunoassay, an enzymatic assay, electrochemical detection, or optical detection.
