Abstract

Systems and methods for automated laser microdissection are disclosed including automatic slide detection, position detection of cutting and capture lasers, focus optimization for cutting and capture lasers, energy and duration optimization for cutting and capture lasers, inspection and second phase capture and/or ablation in a quality control station and tracking information for linking substrate carrier or output microdissected regions with input sample or slide.

Claims (23)

1. A method for using a laser microdissection instrument comprising:

loading a carrier having a transfer film attached into the laser microdissection instrument, wherein initial tracking information unique to the carrier is associated with the carrier;

introducing a substrate with a sample of biological material mounted on the substrate into the laser microdissection instrument, wherein initial tracking information unique to the substrate is associated with the substrate;

moving the carrier into juxtaposition with a target location on the substrate wherein tracking information unique to the substrate is associated with the carrier;

capturing biological material from the target location to the carrier, wherein tracking information related to processing the sample of biological material on the substrate is associated with the carrier; and

recording tracking information unique to the carrier including a carrier number, a carrier style, and a date and a time of microdissection.

2. The method of claim 1 , further comprising outputting the at least one tracking information upon end user request.

3. The method of claim 1 , wherein the at least one tracking information includes information related to a location of the carder in a station on a worksurface of the laser microdissection instrument.

4. The method of claim 3 , wherein the location of the carrier is recorded from initial tracking information related to a location in a cap station on the worksurface of the laser microdissection instrument.

5. The method of claim 3 , wherein the location of the carder is recorded from tracking information related to a location of a substrate in a slide station on the worksurface of the laser microdissection instrument.

6. The method of claim 1 , after capturing biological material from the target location, the method further comprises:

moving the carrier to a quality control station;

performing a quality control operation on the carrier; and

updating tracking information associated with the carrier to include information associated with performing a quality control operation on the carrier.

7. The method of claim 6 , wherein updating tracking information associated with the carrier includes information related to a location of the carrier in the quality station, an identification of biological material on the transfer film requiring further ablation, and an image of the carrier.

8. The method of claim 7 , further comprising outputting updated tracking information related to the carrier in the quality control station upon end user request.

9. The method of claim 1 , after capturing biological material from the target location, the method further comprises:

moving the carrier to an unload station;

positioning the carrier in a location defined by a set of coordinates in the unload station; and

updating the tracking information associated with the carrier to include information related to the set of coordinates for locating the carrier.

10. The method of claim 9 , further comprising outputting updated tracking information related to the carrier in the unload station upon end user request.

The method of claim 1 , wherein recording tracking information further comprises recording initial tracking information unique to the substrate, including a substrate number, a substrate name, a type of substrate, and a tissue type for the sample of biological material mounted on the substrate.

The method of claim 1 , wherein recording tracking information further comprises recording tracking information related to processing the sample of biological material on the substrate, including a number of captures, a total area of captures, and an image of the substrate from which a tissue sample was captured.
