Abstract

A sensor (e.g., an optical sensor) that may be implanted within a living animal (e.g., a human) and may be used to measure an analyte (e.g., glucose or oxygen) in a medium (e.g., interstitial fluid, blood, or intraperitoneal fluid) within the animal. The sensor may include a sensor housing, an analyte indicator covering at least a portion of the sensor housing, and one or more compounds containing boronate or boronic acid moieties that reduce degradation of the analyte indicator.

Claims (86)

1. A sensor for measurement of an analyte in a medium within a living animal, the sensor comprising:

a sensor housing;

an analyte indicator covering at least a portion of the sensor housing, wherein the analyte indicator comprises indicator molecules comprising a boronate group; and

A) characterized in that the sensor further comprises:

one or more compounds selected from Formula I and Formula II:

R—B(OH) 2   [Formula I],

wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, a heteroaryl, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y=alkyl, or

a combination thereof; or

B) characterized in that the analyte indicator comprises a polymer comprising co-monomers of four monomers according to Formula III:

A x B y C z D w   [Formula III],

wherein A is a monomer of the indicator molecules, B is a methacrylate monomer, C is a polyethylene glycol monomer, and D is one or more compounds selected from Formula I, a phenylboronic acid compound, and Formula II:

R—B(OH) 2   [Formula I],

wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

a phenylboronic acid compound

wherein one or more R in the phenylboronic acid compound is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y=alkyl;

wherein A is 0.01 to 10% by weight, B is 1 to 99% by weight, Cis 1 to 99% by weight, D is 0.01 to 99% by weight of the total polymer, a weight of A, B, C, and D combined is not more than 100% by total weight of the polymer, and x, y, z, and w are integers greater than 0;

wherein the one or more compounds of Formula I, the phenylboronic acid compound, and Formula II interact or react with a degradative species so as to neutralize, bind to, and/or sequester the degradative species so as to inhibit, reduce, or prevent degradation of the indicator molecules of the analyte indicator resulting from reactions of the indicator molecules with the degradative species, wherein the degradative species is hydrogen peroxide, a reactive oxygen species, a reactive nitrogen species, an enzyme, a free radical, or a metal ion.

2. The sensor of claim 1 , wherein the sensor is implantable within the living animal.

3. The sensor of claim 1 , wherein the one or more compounds further comprise a phenylboronic acid compound

wherein one or more R in the phenylboronic acid compound is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group.

4. The sensor of claim 3 , wherein the one or more compounds are selected from 2-acrylamidophenylboronic acid, 3-(Acrylamido)phenylboronic acid, 4-vinylboronic acid containing moieties, and (3-methacrylamidophenyl) boronic acid.

5. The sensor of claim 1 , wherein the one or more compounds are co-monomers with the analyte indicator in a hydrogel.

6. The sensor of claim 1 , wherein the one or more compounds are entrapped in a hydrogel covering at least a portion of the sensor housing.

7. The sensor of claim 1 , wherein the one or more compounds reduce chemical degradation and/or oxidation of the analyte indicator.

8. The sensor of claim 7 , wherein the one or more compounds reduce a degradation rate of the analyte indicator.

9. The sensor of claim 7 , wherein the one or more compounds sequester the degradative species so as to reduce, and/or prevent degradation of the analyte indicator by the degradative species.

10. The sensor of claim 1 , wherein the sensor comprises one or more compounds selected from: a compound of formula I:

R—B(OH) 2 [Formula I], wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, a heteroaryl, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, or

a boronate compound of Formula II:

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y =alkyl, or

a combination thereof.

11. The sensor of claim 1 , wherein the analyte indicator comprises a polymer comprising co-monomers of four monomers according to Formula III: A x B y C z D w [Formula III],

wherein A is a monomer of the indicator molecules, B is a methacrylate monomer, C is a polyethylene glycol monomer, and D is one or more compounds selected from Formula I, a phenylboronic acid compound, and Formula II:

R—B(OH) 2   [Formula I],

wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

a phenylboronic acid compound

wherein one or more R in the phenylboronic acid compound is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y=alkyl, wherein A is 0.01 to 10% by weight, B is 1 to 99% by weight, C is 1 to 99% by weight, D is 0.01 to 99% by weight of the total polymer, a weight of A, B, C, and D combined is not more than 100% by total weight of the polymer, and x, y, z, and w are integers greater than 0.

12. The sensor of claim 1 , wherein the one or more compounds are provided at a molar ratio of 0.1 to 100 to a monomer of the indicator molecules.

13. The sensor of claim 1 , wherein the sensor comprises at least one drug eluting polymer matrix that covers at least the portion of the sensor housing.

14. The sensor of claim 1 , wherein the one or more compounds are a boronate compound of Formula II:

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y =alkyl.

15. D) A method of fabricating a sensor for measurement of an analyte in a medium within a living animal, the method comprising:

applying an analyte indicator to a sensor housing of the sensor such that the applied analyte indicator covers at least a portion of the sensor housing, wherein the analyte indicator comprises indicator molecules comprising a boronate group; and

A) characterized in that the method further comprises:

incorporating into the analyte indicator one or more compounds selected from Formula I and Formula II:

R—B(OH) 2   [Formula I],

wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, a heteroaryl, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y=alkyl, or

a combination thereof; or

B) characterized in that the method further comprises:

incorporating the analyte indicator into a polymer comprising co-monomers of four monomers according to Formula III:

A x B y C z D w   [Formula III],

wherein A is a monomer of the indicator molecules, B is a methacrylate monomer, C is a polyethylene glycol monomer, and D is one or more compounds selected from Formula I, a phenylboronic acid compound, and Formula II:

R—B(OH) 2   [Formula I],

wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

a phenylboronic acid compound

wherein one or more R in the phenylboronic acid compound is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y=alkyl;

wherein A is 0.01 to 10% by weight, B is 1 to 99% by weight, C is 1 to 99% by weight, D is 0.01 to 99% by weight of the total polymer, a weight of A, B, C, and D combined is not more than 100% by total weight of the polymer, and x, y, z, and w are integers greater than 0;

wherein the one or more compounds of Formula I, the phenylboronic acid compound, and Formula II interact or react with a degradative species so as to neutralize, bind to, and/or sequester the degradative species so as to inhibit, reduce, or prevent degradation of the indicator molecules of the analyte indicator resulting from reactions of the indicator molecules with the degradative species, wherein the degradative species is hydrogen peroxide, a reactive oxygen species, a reactive nitrogen species, an enzyme, a free radical, or a metal ion.

16. The method of claim 15 , wherein the one or more compounds further comprise a phenylboronic acid compound

wherein one or more R in the phenylboronic acid compound is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group.

17. The method of claim 16 , wherein the one or more compound are selected from 2-acrylamidophenylboronic acid, 3-(Acrylamido)phenylboronic acid, 4-vinylboronic acid containing moieties, and (3-methacrylamidophenyl)boronic acid.

18. The method of claim 15 , wherein the one or more compounds are co-monomers with the analyte indicator in a hydrogel.

19. The method of claim 15 , wherein the one or more compounds are entrapped in a hydrogel covering at least a portion of the sensor housing.

20. The method of claim 15 , wherein the one or more compounds reduce chemical degradation and/or oxidation of the analyte indicator.

21. The method of claim 15 , wherein the one or more compounds reduce a degradation rate of the analyte indicator.

22. The method of claim 15 , wherein the one or more compounds bind to the degradative species.

23. The method of claim 15 , wherein the one or more compounds sequester the degradative species so as to reduce, and/or prevent degradation of the analyte indicator by the degradative species.

24. The method of claim 15 , wherein the method comprises incorporating into the analyte indicator one or more compounds selected from: a compound of formula I:

R—B(OH) 2 [Formula I], wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, a heteroaryl, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, or

a boronate compound of Formula II:

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y =alkyl, or

a combination thereof.

25. The method of claim 15 , wherein the method comprises incorporating the analyte indicator into a polymer comprising co-monomers of four monomers according to Formula III:

ABCD  [Formula III],

wherein A is a monomer of the analyte indicator, B is a methacrylate monomer, C is a polyethylene glycol monomer, and D is one or more compounds selected from Formula I, a phenylboronic acid compound, and Formula II:

R—B(OH) 2   [Formula I],

wherein R in Formula I is selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 , wherein R 1 and R 2 may be identical or different and each may represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

a phenylboronic acid compound

wherein one or more R in the phenylboronic acid compound is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group; or

wherein R in Formula II is independently selected from hydrogen, hydroxyl, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, and/or NR 1 R 2 wherein R 1 and R 2 , identical or different, each represent a hydrogen atom, a hydroxyl group, an alkyl group, an alkoxy group, an amino group, an aryl group, a heteroaryl, a cyclic group, a multicyclic group, a carboxylic acid, a vinyl group, an acrylate group, an acryloyl group, or a methacrylate group, and wherein X and Y=alkyl, wherein A is 0.01 to 10% by weight, B is 1 to 99% by weight, C is 1 to 99% by weight, and D is 0.01 to 99% by weight of the total polymer.

26. The method of claim 15 , wherein the one or more compounds are provided at a molar ratio of 0.1 to 100 to a monomer of the indicator molecules.
