Abstract

A glycophospholipid polymer is disclosed. The glycophospholipid polymer comprises a reaction product of one or more glycosaminoglycans and one or more phospholipids. Also disclosed is a glycophospholipid polymeric network comprising a reaction product of one or more glycosaminoglycans, one or more phospholipids and, one or more crosslinking agents.

Claims (47)

1. A crosslinked glycophospholipid polymeric network comprising a reaction product of one or more glycosaminoglycans having one or more reactive functionality groups, one or more phospholipids having one or more reactive functionality groups and one or more first crosslinking agents, wherein the one or more first crosslinking agents comprise a bi- or polyfunctional crosslinking agent comprising two or more functional groups that are complementary to the one or more reactive functionality groups of the one or more glycosaminoglycans and the one or more phospholipids, wherein one of the two or more functional groups of the bi- or polyfunctional crosslinking agent is a glycidyl ether functional group and a bond between the glycidyl ether functional group of the bi- or polyfunctional crosslinking agent and at least one of the one or more reactive functionality groups of the one or more glycosaminoglycans is an ether bond.

2. The crosslinked glycophospholipid polymeric network of claim 1 , wherein the one or more glycosaminoglycans are selected from the group consisting of chondroitin, chondroitin sulfate, dermatan, dermatan sulfate, heparin, heparan sulfate, hyaluronan and hyaluronic acid.

The crosslinked glycophospholipid polymeric network of claim 1 , wherein the one or more phospholipids are selected from the group consisting of a phosphorylcholine-containing phospholipid, a phosphorylhistidine-containing phospholipid, a phosphorylproline-containing phospholipid, a phosphorylserine-containing phospholipid, α-phosphatidylcholine, α-phosphatidylethanolamine, α-phosphatidyl-L-serine, α-phosphatidylinositol, α-phosphatidic acid, α-phosphatidyl-DL-glycerol, α-lysophosphatidylcholine, sphingomyelin, and cardiolipin.

4. The crosslinked glycophospholipid polymeric network of claim 1 , wherein the one or more glycosaminoglycans are hyaluronic acid or chondroitin sulfate and the one or more phospholipids comprise a phosphorylhistidine-containing phospholipid or a phosphorylcholine-containing phospholipid.

5. The glycophospholipid polymeric network of claim 1 , wherein the one or more phospholipids are a phosphorylcholine-containing phospholipid comprising a zwitterionic group represented by the following structure:

wherein n is an integer of 1 to 5 and R′, R″ and R′″ independently of each other are a C 1 to C 8 alkyl group, a C 1 to C 8 hydroxyalkyl group or a hetero atom-containing group.

6. The crosslinked glycophospholipid polymeric network of claim 1 , wherein the reaction product is derived from a solution comprising:

about 0.01 to about 50 wt. %, based on the total weight of the solution, of the one or more glycosaminoglycans; and

about 0.01 to about 50 wt. %, based on the total weight of the solution, of the one or more phospholipids.

7. The crosslinked glycophospholipid polymeric network of claim 1 , wherein one or more additional glycosaminoglycans are crosslinked with the reaction product with one or more second crosslinking agents.

8. The crosslinked glycophospholipid polymeric network of claim 7 , wherein the one or more second crosslinking agents are the same as the one or more first crosslinking agents.

9. The crosslinked glycophospholipid polymeric network of claim 7 , wherein the one or more second crosslinking agents are different than the one or more first crosslinking agents.

10. The crosslinked glycophospholipid polymeric network of claim 1 , wherein the reaction product is derived from a solution comprising:

about 0.01 to about 50 wt. %, based on the total weight of the solution, of the one or more glycosaminoglycans;

about 0.01 to about 50 wt. %, based on the total weight of the solution, of the one or more phospholipids; and

about 0.05 to about 10 wt. %, based on the total weight of the solution, of one or more first crosslinking agents.

11. The crosslinked glycophospholipid polymeric network of claim 1 , wherein the reaction product comprises the one or more glycosaminoglycans crosslinked with the one or more phospholipids.

12. The crosslinked glycophospholipid polymeric network of claim 11 , wherein the one or more glycosaminoglycans crosslinked with the one or more phospholipids has a weight average molecular weight ranging from about 20,000 to about 6,000,000 Da.

13. A packaging system for the storage of an ophthalmic device comprising a sealed container containing one or more unused ophthalmic devices immersed in an aqueous packaging solution comprising one or more crosslinked glycophospholipid polymeric networks;

wherein the one or more crosslinked glycophospholipid polymeric networks comprise a reaction product of one or more glycosaminoglycans having one or more reactive functionality groups, one or more phospholipids having one or more reactive functionality groups and one or more crosslinking agents, wherein the one or more crosslinking agents comprise a bi- or polyfunctional crosslinking agent comprising two or more functional groups that are complementary to the one or more reactive functionality groups of the one or more glycosaminoglycans and the one or more phospholipids, wherein a bond between at least one of the two or more functional groups of the bi- or polyfunctional crosslinking agents and at least one of the one or more reactive functionality groups of the one or more glycosaminoglycans is an ether bond; and

wherein the solution has an osmolality of at least about 200 mOsm/kg, a pH of about 6 to about 9 and is sterilized.

14. The packaging system of claim 13 , wherein the one or more unused ophthalmic devices are a contact lens.

15. The packaging system of claim 13 , wherein in the one or more crosslinked glycophospholipid polymeric networks, one or more additional glycosaminoglycans are crosslinked with the reaction product with one or more second crosslinking agents.

16. The packaging system of claim 15 , wherein the one or more unused ophthalmic devices are a contact lens.

An aqueous ophthalmic composition comprising one or more of crosslinked glycophospholipid polymeric networks, wherein the one or more crosslinked glycophospholipid polymeric networks comprise a reaction product of one or more glycosaminoglycans having one or more reactive functionality groups, one or more phospholipids having one or more reactive functionality groups and one or more first crosslinking agents, wherein the one or more first crosslinking agents comprise a bi- or polyfunctional crosslinking agent comprising two or more functional groups that are complementary to the one or more reactive functionality groups of the one or more glycosaminoglycans and the one or more phospholipids, wherein one of the two or more functional groups of the bi- or polyfunctional crosslinking agent is a glycidyl ether functional group and a bond between the glycidyl ether functional group of the bi- or polyfunctional crosslinking agent and at least one of the one or more reactive functionality groups of the one or more glycosaminoglycans is an ether bond; and

wherein the aqueous ophthalmic composition has an osmolality in a range from about 200 mOsmol/kg to about 400 mOsmol/kg.

18. The aqueous ophthalmic composition of claim 17 , in the form of an eye care or a contact lens care product selected from the group consisting of eye drops, a contact lens preservative solution, a contact lens cleaning solution, and a contact lens multi-purpose solution.

19. The aqueous ophthalmic composition of claim 17 , in the form of a multi-purpose solution or rewetting drops.

20. An aqueous ophthalmic composition comprising one or more of the crosslinked glycophospholipid polymeric networks of claim 7 , wherein the aqueous ophthalmic composition has an osmolality in a range from about 200 mOsmol/kg to about 400 mOsmol/kg.

21. The aqueous ophthalmic composition of claim 20 , in the form of an eye care or a contact lens care product selected from the group consisting of eye drops, a contact lens preservative solution, a contact lens cleaning solution, and a contact lens multi-purpose solution.

22. The aqueous ophthalmic composition of claim 20 , in the form of a multi-purpose solution or rewetting drops.

23. The aqueous ophthalmic composition of claim 17 , wherein the one or more crosslinked glycophospholipid polymeric networks are present in the aqueous ophthalmic composition in an amount of from about 0.001 to about 10% w/w.

24. The packaging system of claim 13 , wherein the one or more crosslinked glycophospholipid polymeric networks are present in the aqueous packaging solution in an amount of from about 0.01 to about 20% w/w.

25. The packaging system of claim 13 , wherein the one or more glycosaminoglycans are hyaluronic acid or chondroitin sulfate and the one or more phospholipids comprise a phosphorylhistidine-containing phospholipid or a phosphorylcholine-containing phospholipid.

26. The packaging system of claim 13 , wherein the one or more phospholipids are a phosphorylcholine-containing phospholipid comprising a zwitterionic group represented by the following structure:

wherein n is an integer of 1 to 5 and R′, R″ and R′″ independently of each other are a C 1 to C 8 alkyl group, a C 1 to C 8 hydroxyalkyl group or a hetero atom-containing group.

27. The packaging system of claim 13 , wherein the bi- or polyfunctional crosslinking agent is a bi- or polyfunctional epoxide crosslinking agent.

28. The packaging system of claim 27 , wherein the bi- or polyfunctional epoxide crosslinking agent comprises two or more glycidyl ether functional groups.

29. The packaging system of claim 15 , wherein the one or more crosslinked glycophospholipid polymeric networks are present in the aqueous packaging solution in an amount of from about 0.01 to about 20% w/w.

30. The packaging system of claim 15 , wherein the one or more glycosaminoglycans are hyaluronic acid or chondroitin sulfate and the one or more phospholipids comprise a phosphorylhistidine-containing phospholipid or a phosphorylcholine-containing phospholipid.

31. The packaging system of claim 15 , wherein the one or more phospholipids are a phosphorylcholine-containing phospholipid comprising a zwitterionic group represented by the following structure:

wherein n is an integer of 1 to 5 and R′, R″ and R′″ independently of each other are a C 1 to C 8 alkyl group, a C 1 to C 8 hydroxyalkyl group or a hetero atom-containing group.

32. The packaging system of claim 15 , wherein the bi- or polyfunctional crosslinking agent is a bi- or polyfunctional epoxide crosslinking agent.

33. The packaging system of claim 32 , wherein the bi- or polyfunctional epoxide crosslinking agent comprises two or more glycidyl ether functional groups.

34. A crosslinked glycophospholipid polymeric network comprising a reaction product of one or more glycosaminoglycans having one or more reactive functionality groups, one or more phospholipids having one or more reactive functionality groups and one or more first crosslinking agents, wherein the one or more first crosslinking agents comprise a bi- or polyfunctional crosslinking agent comprising a first and at least a second functional group that are each complementary to the one or more reactive functionality groups of the one or more glycosaminoglycans and the one or more phospholipids, wherein a bond between the first functional group of the bi- or polyfunctional crosslinking agent and one of the at least one or more reactive functionality groups of the one or more glycosaminoglycans is an ether bond, and a bond between the second functional group of the bi- or polyfunctional crosslinking agent and another one of the at least one or more reactive functionality groups of the one or more glycosaminoglycans is an ether bond.

The crosslinked glycophospholipid polymeric network of claim 34 , wherein the one or more glycosaminoglycans are hyaluronic acid or chondroitin sulfate, the one or more phospholipids comprise a phosphorylhistidine-containing phospholipid or a phosphorylcholine-containing phospholipid and the bi- or polyfunctional crosslinking agent is a bi- or polyfunctional epoxide crosslinking agent.

36. An aqueous ophthalmic composition comprising one or more of the crosslinked glycophospholipid polymeric networks of claim 34 , wherein the aqueous ophthalmic composition has an osmolality in a range from about 200 mOsmol/kg to about 400 mOsmol/kg.
