Abstract

Dosage forms, drug delivery systems, and methods related to sustained release of dextromethorphan or improved therapeutic effects are disclosed. Typically, bupropion or a related compound is orally administered to a human being to be treated with, or being treated with, dextromethorphan.

Claims (16)

1. A method of treating agitation in a human patient with Alzheimer's disease, comprising administering a drug combination comprising: a daily dose of bupropion that is about 150 mg to about 250 mg and a daily dose of dextromethorphan that is about 60 mg to about 100 mg, to the human patient for at least 8 consecutive days, and wherein administering the drug combination is more effective in treating agitation in the human patient than administering a placebo.

2. The method of claim 1 , wherein administering the drug combination is more effective in treating agitation in the human patient than administering the same amount of the dextromethorphan alone.

3. The method of claim 1 , wherein the daily dose of the bupropion is administered once daily and the daily dose of the dextromethorphan is administered once daily.

4. The method of claim 1 , wherein one half of the daily dose of the bupropion is administered twice daily and one half of the daily dose of the dextromethorphan is administered twice daily.

5. The method of claim 1 , wherein the drug combination is administered for at least 14 consecutive days.

6. The method of claim 1 , wherein the drug combination is administered for at least 30 consecutive days.

7. The method of claim 1 , wherein the dextromethorphan is formulated for immediate release.

8. The method of claim 1 , wherein the bupropion is formulated for sustained release.

9. The method of claim 1 , wherein the weight ratio of the dextromethorphan to the bupropion in the drug combination is about 0.3 to about 0.5.

10. The method of claim 1 , wherein the drug combination is administered orally.

11. The method of claim 1 , wherein the bupropion is in a solid dosage form.

12. The method of claim 1 , wherein the dextromethorphan is in a solid dosage form.

13. The method of claim 1 , wherein on the eighth day, the human patient has a C avg of bupropion that is at least about 10 ng/mL after administering the drug combination.

14. The method of claim 1 , wherein on the eighth day, the human patient has a C max of bupropion that is at least about 90 ng/mL after administering the drug combination.

15. The method of claim 1 , wherein the drug combination comprises: a daily dose of bupropion hydrochloride that is about 210 mg and a daily dose of dextromethorphan that is about 60 mg to about 100 mg.

16. The method of claim 15 , wherein administering the drug combination is more effective in treating agitation in the human patient than administering 105 mg of bupropion hydrochloride twice a day for the same number of consecutive days.
