Abstract

Provided herein are systems and methods for simultaneous imaging and stimulation using a microscope system. The microscope system can have a relatively small size compared to an average microscope system. The microscope can comprise in part an imaging light source and a stimulation light source. Light from the imaging light source and the stimulation light source can be spectrally separated to reduce cross talk between the stimulation light and the imaging light.

Claims (27)

1. A microscope system, comprising:

a memory device; and

one or more processors that execute, for an optical assembly comprising (i) an imaging light source that directs imaging light to tissue within a field-of-view, (ii) a stimulation light source that directs stimulation light to the tissue, and (iii) an image sensor that receives light from the tissue to generate an image of the tissue within the field-of-view, wherein the optical assembly mounts to a subject comprising the tissue, one or more software-encoded instructions in the memory device selected from the group consisting of:

(1) modulating the imaging light in a time-dependent or spatially-dependent manner;

(2) modulating the stimulation light in a time-dependent or spatially-dependent manner; and

(3) varying focal depth in a time-dependent manner,

wherein the one or more processors further execute one or more software-encoded instructions to: (i) modulate the stimulation light to deliver the stimulation to one or more objects of interests of the tissue in a pattern, (ii) receive a feedback image of the tissue as a response to the pattern, and (iii) modify the pattern based on the feedback image.

2. The microscope system of claim 1 , wherein the one or more processors execute software-encoded instructions for modulating the stimulation light in a time-dependent or spatially dependent manner to induce a neurological response in the brain tissue.

3. The microscope system of claim 1 , wherein the one or more processors execute software-encoded instructions for modulating the stimulation light in a time-dependent or spatially dependent manner to induce a therapeutic effect in the subject.

4. The microscope system of claim 1 , wherein the optical assembly mounts on a freely mobile subject.

5. The microscope system of claim 1 , wherein the one or more processors execute one or more software-encoded instructions to determine the pattern.

6. The microscope system of claim 5 , wherein the one or more processors execute one or more software-encoded instructions to receive and process an input image of the tissue from the optical assembly for use in determining the pattern.

7. The microscope system of claim 1 , wherein the one or more processors execute one or more software-encoded instructions to modulate the imaging light in a user-defined spatial pattern or user-defined temporal pattern.

8. The microscope system of claim 1 , wherein the one or more processors execute one or more software-encoded instructions to modulate the stimulation light in a user-defined spatial pattern or user-defined temporal pattern.

9. A method for a microscope system, comprising:

(a) executing, with aid of one or more processors in operable communication with a memory device, for an optical assembly comprising (i) an imaging light source that directs imaging light to tissue within a field-of-view, (ii) a stimulation light source that directs stimulation light to the tissue, and (iii) an image sensor that receives light from the tissue to generate an image of the tissue within the field-of-view, the optical assembly mounted to a subject comprising the tissue, one or more software-encoded instructions in the memory device selected from the group consisting of:

(1) modulating the imaging light in a time-dependent or spatially-dependent manner;

(2) modulating the stimulation light in a time-dependent or spatially-dependent manner; and

(3) varying focal depth in a time-dependent manner; and

(b) executing, with aid of the one or more processors software-encoded instructions for (i) modulating the stimulation light to deliver the stimulation to one or more objects of interests of the tissue in a pattern, (ii) receiving a feedback image of the tissue as a response to the pattern, and (iii) modifying the pattern based on the feedback image.

10. The method of claim 9 , further comprising executing with aid of the one or more processors software-encoded instructions for modulating the stimulation light in a time-dependent or spatially dependent manner to induce a neurological response in the brain tissue.

11. The method of claim 9 , further comprising executing with aid of the one or more processors software-encoded instructions for modulating the stimulation light in a time-dependent or spatially dependent manner to induce a therapeutic effect in the subject.

12. The method of claim 9 , wherein the optical assembly is mounted on a freely mobile subject.

13. The method of claim 9 , further comprising executing with aid of the one or more processors software-encoded instructions for determining the pattern.

14. The method of claim 13 , further comprising executing with aid of the one or more processors software-encoded instructions for receiving and processing an input image of the tissue from the optical assembly for use in determining the pattern.

15. The method of claim 9 , further comprising executing with aid of the one or more processors software-encoded instructions for modulating the imaging light in a user-defined spatial pattern or user-defined temporal pattern.

16. The method of claim 9 , further comprising executing with aid of the one or more processors software-encoded instructions for modulating the stimulation light in a user-defined spatial pattern or user-defined temporal pattern.
