Engineered phytases in animal feed
Methods for enhancing phytase thermal stability by fusing binding elements to target phytases are provided. Engineered phytases that include binding elements fused to target phytases to cause cyclization of the engineered phytases and enhance thermal stability of the target phytases are described. Engineered nucleic acids encoding engineered phytases and hosts engineered to express engineered nucleic acids are also provided. Methods for incorporating engineered phytases in animal feed and animal feed including the same are described.
1. An animal feed comprising a cyclic engineered phytase comprising a target phytase, a first binding element, and a second binding element, wherein
the first binding element is fused to the N-terminus of the target phytase and the second binding element is fused to the C-terminus of the target phytase;
the first binding comprises a tag domain and the second binding element comprises a catcher domain;
the target phytase has at least 95% identity to SEQ ID NO: 53;
the tag domain is an amino acid sequence with at least 90% identity to the amino acid sequence set forth in SEQ ID NO: 33 or SEQ ID NO: 34 that is capable of interacting with the catcher domain;
the catcher domain is an amino acid sequence with at least 90% identity to the amino acid sequence set forth in SEQ ID NO: 35 or SEQ ID NO: 36 that is capable of interacting with the tag domain;
the engineered phytase has at least 95% identity with SEQ ID NO: 119;
the tag domain and catcher domain are covalently linked resulting in the cyclic engineered phytase; and
wherein the cyclic engineered phytase has enhanced thermal stability compared to the target phytase.
2. The animal feed of claim 1 , wherein the cyclic engineered phytase has enhanced thermostability at a temperature in a range from 70° C. to 90° C., compared to the target phytase.
3. The animal feeds of claim 1 further comprising a feed supplement.
4. The animal feed of claim 3 , wherein the feed supplement is a non-transgenic plant or an engineered plant.
5. The animal feed of claim 3 , wherein the supplement includes at least one component selected from the group consisting of: corn meal, corn pellets, wheat meal, wheat pellets, wheat grain, barley grain, barley pellets, soybean meal, soybean oilcake, sorghum grain and sorghum pellets.
6. The animal feed of claim 3 , wherein the feed supplement includes one or more exogenous enzymes.
7. The animal feed of claim 6 , wherein the one or more exogenous enzymes includes a hydrolytic enzyme selected from the group consisting of: xylanase, endoglucanase, cellulase, protease, glucanase, amylase and mannanase.
8. The animal feed of claim 3 , wherein the feed supplement includes at least one component selected from the group consisting of: soluble solids derived from dry milling of corn plants, the combination of fat and vermiculite, limestone, salt, one or more vitamins, and one or more minerals.
9. The animal fee of claim 8 , wherein the salt is selected from the group consisting of dicalcium phosphate, choline chloride, and sodium chloride.
10. The animal feed of claim 8 , wherein the one or more minerals comprises selenium.
11. The animal feed of claim 3 , wherein the feed supplement is DL-methionine, L-lysine, and/or L-threonine.
12. The animal feed of claim 3 , wherein the feed supplement is the antibiotic monensin.
