Abstract

The present invention is related to HMGB1 antagonists and, in particular, HMGB1 tetramer peptidomimetics which have been stabilized with at least one azatide linkage, such as K883. The present invention is also related to pharmaceutical compositions comprising HMGB1 antagonists such as K883.

Claims (25)

1. A pharmaceutical composition comprising a peptidomimetic small molecule of the formula:

wherein:

R 1 is CH or N; and

R 2 is CH or N,

provided that at least one of R 1 or R 2 is N,

and at least one pharmaceutically acceptable excipient.

2. The pharmaceutical composition of claim 1 , wherein R 1 is CH and R 2 is N.

3. The pharmaceutical composition of claim 1 , wherein R 1 is N and R 2 is CH.

4. The pharmaceutical composition of claim 1 , wherein the peptidomimetic small molecule is of the formula:

The pharmaceutical composition of claim 1 , which is a dosage form selected from the group consisting of an oral dosage form, a parenteral dosage form, a buccal dosage form, a sublingual dosage form, a nasal dosage form, an inhaler, a nebulizer, a topical dosage form, a transdermal dosage form and a suppository.

6. The pharmaceutical composition of claim 1 , wherein the pharmaceutical excipient comprises PBS:PEG 300:propylene glycol:polysorbate 80.

7. The pharmaceutical composition of claim 1 , wherein the pharmaceutical excipient comprising PBS:PEG 300:propylene glycol:polysorbate 80 in a ratio of about 50:40:5:5.

8. The pharmaceutical composition of claim 6 , wherein the composition is an oral liquid dosage form or a parenteral dosage form.

9. The pharmaceutical composition of claim 1 , wherein the pharmaceutical excipient is selected from the group consisting of 1) phosphate buffered saline, 2) polyethylene glycol, 3) propylene glycol and 4) polysorbate 80 and 5) combinations thereof.

10. The pharmaceutical composition of claim 9 , wherein the polyethylene glycol is PEG 300.

11. The pharmaceutical composition of claim 1 , wherein the pharmaceutical composition is an oral dosage form.

12. The pharmaceutical composition of claim 1 , wherein the pharmaceutical composition is a parenteral dosage form.

13. The pharmaceutical composition of claim 1 , wherein the aqueous solubility of the peptidomimetic molecule is greater than 1 mg/ml.

14. The pharmaceutical composition of claim 1 , wherein the peptidomimetic molecule stable for greater than 60 minutes in plasma or simulated stomach acid.

15. The pharmaceutical composition of claim 4 , wherein the peptidomimetic molecule stable for greater than 60 minutes in plasma or simulated stomach acid.

The pharmaceutical composition of claim 4 , which is a dosage form selected from the group consisting of an oral dosage form, a parenteral dosage form, a buccal dosage form, a sublingual dosage form, a nasal dosage form, an inhaler, a nebulizer, a topical dosage form, a transdermal dosage form and a suppository.

17. The pharmaceutical composition of claim 4 , wherein the pharmaceutical excipient comprises PBS:PEG 300:propylene glycol:polysorbate 80.

18. The pharmaceutical composition of claim 4 , wherein the pharmaceutical excipient comprising PBS:PEG 300:propylene glycol:polysorbate 80 in a ratio of about 50:40:5:5.

19. The pharmaceutical composition of claim 17 , wherein the composition is an oral liquid dosage form or a parenteral dosage form.

20. The pharmaceutical composition of claim 4 , wherein the pharmaceutical composition is an oral dosage form or a parenteral dosage form.
