Abstract

Disclosed are chemical entities which are inhibitors of spleen tyrosine kinase (SYK), namely, chemical entities comprising 6-((1R,2S)-2-aminocyclohexylamino)-7-fluoro-4-(1-methyl-1H-pyrazol-4-yl)-1H-pyrrolo[3,4-c]pyridine-3(2H)-one and certain solid state forms thereof. Also disclosed are methods of using the chemical entities to treat disorders such as a cancer.

Claims (22)

1. A method of treating a cancer comprising administering to a subject having a cancer a crystal form of 6-((1R,2S)-2-aminocyclohexylamino)-7-fluoro-4-(1-methyl-1H-pyrazol-4-yl)-1H-pyrrolo[3,4-c]pyridine-3(2H)-one citrate, which is in substantially crystalline form, and is characterized by an X-ray powder diffraction (XRPD) pattern using Cu Kα radiation comprising peaks at 2θ angles of 9.4, 16.6, 17.4, 18.9, 19.2, and 20.7°±0.2 degrees.

A method of treating a cancer comprising administering to a subject having a cancer a pharmaceutical composition comprisinf a crystal form of 6-((1R,2S)-2-aminocyclohexylamino)-7-fluoro-4-(1-methyl-1H-pyrazol-4-yl)-1H-pyrrolo[3,4-c]pyridine-3(2H)-one citrate, which is in substantially crystalline form, and is characterized by an X-ray powder diffraction (XRPD) pattern using Cu Kα radiation comprising peaks at 2θ angles of 9.4, 16.6, 17.4, 18.9, 19.2, and 20.7°±0.2 degrees and one or more pharmaceutically accepable carriers.

3. The method of claim 1 , wherein the cancer is a leukemia or lymphoma.

The method of claim 1 , wherein the cancer is selected from indolent non-Hodgkin's lymphoma (iNHL), peripheral T-cell lymphoma (PTCL), diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), mantle cell lymphoma (MCL), chronic lymphocytic leukemia (CLL), acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), nasopharyngeal carcinoma, gastric carcinoma, breast cancer, ovarian cancer, lung cancer and a post-transplant lymphoproliferative disorder (PT-LPD).

5. The method of claim 1 , wherein the cancer is selected from indolent non-Hodgkin's lymphoma (iNHL), mantle cell lymphoma, post-transplant lymphoproliferative disorder (PT-LPD), diffuse large B-cell lymphoma (DLBCL), chronic lymphocytic leukemia (CLL), and acute myeloid leukemia (AML).

6. The method of claim 1 , wherein the cancer is selected from diffuse large B-cell lymphoma (DLBCL) and acute myeloid leukemia (AML).

7. The method of claim 2 , wherein the cancer is a leukemia or lymphoma.

The method of claim 2 , wherein the cancer is selected from indolent non-Hodgkin's lymphoma (iNHL), peripheral T-cell lymphoma (PTCL), diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), mantle cell lymphoma (MCL), chronic lymphocytic leukemia (CLL), acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), nasopharyngeal carcinoma, gastric carcinoma, breast cancer, ovarian cancer, lung cancer and a post-transplant lymphoproliferative disorder (PT-LPD).

9. The method of claim 2 , wherein the cancer is selected from indolent non-Hodgkin's lymphoma (iNHL), mantle cell lymphoma, post-transplant lymphoproliferative disorder (PT-LPD), diffuse large B-cell lymphoma (DLBCL), chronic lymphocytic leukemia (CLL), and acute myeloid leukemia (AML).

10. The method of claim 2 , wherein the cancer is selected from diffuse large B-cell lymphoma (DLBCL) and acute myeloid leukemia (AML).

11. The method of claim 1 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 16.6, 17.4, 18.9, 19.2, 20.7, and 23.0°±0.2 degrees.

12. The method of claim 1 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 13.0, 13.8, 14.1, 16.6, 17.4, 18.4, 18.9, 19.2, 20.7, 23.0, 23.3, 23.6, and 25.0°±0.2 degrees.

13. The method of claim 1 , wherein the crystal form is not hygroscopic.

14. The method of claim 2 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 16.6, 17.4, 18.9, 19.2, 20.7, and 23.0°±0.2 degrees.

15. The method of claim 2 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 13.0, 13.8, 14.1, 16.6, 17.4, 18.4, 18.9, 19.2, 20.7, 23.0, 23.3, 23.6, and 25.0°±0.2 degrees.

16. The method of claim 2 , wherein the crystal form is not hygroscopic.

17. The method of claim 4 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 16.6, 17.4, 18.9, 19.2, 20.7, and 23.0°±0.2 degrees.

18. The method of claim 4 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 13.0, 13.8, 14.1, 16.6, 17.4, 18.4, 18.9, 19.2, 20.7, 23.0, 23.3, 23.6, and 25.0°±0.2 degrees.

19. The method of claim 4 , wherein the crystal form is not hygroscopic.

20. The method of claim 8 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 16.6, 17.4, 18.9, 19.2, 20.7, and 23.0°±0.2 degrees.

21. The method of claim 8 , wherein the crystal form is characterized by an XRPD pattern using Cu Kα radiation comprising peaks at 2θ angles of 4.7, 9.4, 13.0, 13.8, 14.1, 16.6, 17.4, 18.4, 18.9, 19.2, 20.7, 23.0, 23.3, 23.6, and 25.0°±0.2 degrees.

22. The method of claim 8 , wherein the crystal form is not hygroscopic.
