Composite scaffold for the repair, reconstruction, and regeneration of soft tissues
A composite scaffold having a highly porous interior with increased surface area and void volume is surrounded by a flexible support structure that substantially maintains its three-dimensional shape under tension and provides mechanical reinforcement during repair or reconstruction of soft tissue while simultaneously facilitating regeneration of functional tissue.
1. A composite scaffold comprising:
a microporous matrix having a multitude of interconnected pores opening to an exterior surface of the microporous matrix and collectively defining void space; and
a three-dimensional, textile structure supporting the microporous matrix and comprising first and second outer layers separated by an interior space therebetween and the microporous matrix within the interior space, and a plurality of spacer yarns extending through the interior space and attached to each of the first and second outer layers;
wherein a surface area of the composite scaffold is between approximately 0.3 m 2 /gram and 15.0 m 2 /gram per gram of the composite scaffold.
2. The composite scaffold of claim 1 wherein the surface area of the composite number scaffold is between approximately 0.6 m 2 /gram and 1.2 m 2 /gram.
3. The composite scaffold of claim 1 wherein the surface area of the composite scaffold is between approximately 0.7 m 2 /gram and 1.0 m 2 /gram.
4. The composite scaffold of claim 1 wherein a volume of the void space is between approximately 3.0 cm 3 /gram and 9.0 cm 3 /gram.
5. A composite scaffold having a volume and a surface area and comprising:
a microporous matrix having a multitude of interconnected pores collectively defining void space opening to an exterior surface of the microporous matrix; and
a three-dimensional, textile structure supporting the microporous matrix and comprising first and second outer layers separated by an interior space therebetween and the microporous matrix within the interior space, and
a plurality of spacer yarns extending through the interior space and attached to each of the first and second outer layers will;
wherein the composite scaffold has a permeability of between approximately 1200 and 3000 millidarcy.
6. The composite scaffold of claim 5 wherein the composite scaffold has a permeability of between approximately 1400 and 2600 millidarcy.
7. The composite scaffold of claim 5 wherein the wherein the composite scaffold has a permeability of between approximately 1500 and 2200 millidarcy.
8. The composite scaffold of claim 5 wherein a measurable volume of the microporous matrix void space per gram of the composite scaffold is between approximately 3.0 cm 3 /gram and 9.0 cm 3 /gram.
9. The composite scaffold of claim 5 wherein a surface area of the composite scaffold is between approximately 0.3 m 2 /gram and 15 m 2 /gram per gram of the composite scaffold.
10. A composite scaffold comprising:
a microporous matrix having a multitude of interconnected pores opening to an exterior surface of the microporous matrix and collectively defining void space having a void space volume; and
a three-dimensional, textile structure supporting the microporous matrix and comprising first and second outer layers separated by an interior space therebetween and the microporous matrix disposed within the interior space, and
a plurality of spacer yarns extending through the interior space and attached to each of the first and second outer layers;
wherein the multitude of interconnected pores have a tortuosity of approximately between 1 μm/μm and 50 μm/μm, wherein the tortuosity defines a ratio of actual flow path length to straight distance between first and second ends of the microporous matrix.
11. The composite scaffold of claim 10 wherein the multitude of interconnected pores have a tortuosity of approximately between 5 μm/μm and 30 μm/μm.
12. The composite scaffold of claim 10 wherein the multitude of interconnected pores have a tortuosity of approximately between 6 μm/μm and 20 μm/μm.
13. A composite scaffold comprising:
a three-dimensional, textile support structure comprising first and second outer layers defining an interior space therebetween and separated by the interior space, and further comprising a plurality of spacer yarns extending through the interior space and attached to each of the first and second outer layers; and
a microporous matrix disposed within the interior space of the support structure,
wherein the microporous matrix comprises a plurality of interconnected pores having a median pore size of between approximately 10 μm to 100 μm.
14. The composite scaffold of claim 13 wherein the microporous matrix comprises a plurality of interconnected pores having a median pore size of between approximately 12 μm to 70 μm.
15. The composite scaffold of claim 13 wherein the microporous matrix comprises a plurality of interconnected pores having a median pore size of between approximately 20 μm to 40 μm.
16. The composite scaffold of claim 1 wherein the composite scaffold has an elongation at tensile failure of between approximately 20% and 125%.
17. The composite scaffold of claim 1 wherein the composite scaffold has an elongation at yield of between approximately 5% and 50%.
18. The composite scaffold of claim 1 wherein the composite scaffold has a stiffness of approximately between 2.5 N/mm and 25 N/mm.
19. The composite scaffold of claim 1 wherein the composite scaffold has an ultimate strength of approximately between 2.5 MPa and 30 MPa.
20. The composite scaffold of claim 1 wherein the composite scaffold has an ultimate stress of approximately between 2.5 MPa and 30 MPa.
21. The composite scaffold of claim 1 wherein the composite scaffold has a modulus of approximately between 2.5 MPa and 70 MPa, wherein modulus defines stress divided by strain of a cross-sectional area of the composite scaffold, including the void space.
22. The composite scaffold of claim 1 wherein the composite scaffold has a modulus of approximately between 150 MPa and 600 MPa, wherein modulus defines stress divided by strain of bulk material from which the composite scaffold is comprised, excluding the void space.
23. The composite scaffold of claim 1 wherein the composite scaffold has an ultimate load displacement of approximately between 5 mm and 50 mm.
24. The composite scaffold of claim 1 wherein the composite scaffold has a yield displacement of approximately between 1 mm and 8 mm.
25. The composite scaffold of claim 1 wherein the composite scaffold has a yield force of approximately between approximately between 20 N and 70 N.
26. The composite scaffold of claim 1 wherein the surface area of the composite scaffold is measured using mercury intrusion porosimetry.
27. The composite scaffold of claim 1 wherein the surface area of the composite scaffold includes surface area of pores having a diameter approximately equal to or greater than 1 μm in size.
28. The composite scaffold of claim 10 wherein a volume of the void space is between approximately 3.0 cm 3 /gram and 9.0 cm 3 /gram.
29. The composite scaffold of claim 10 wherein a surface area of the composite scaffold is between approximately 0.3 m 2 /gram and 1.5 m 2 /gram.
30. The composite scaffold of any of claim 1 wherein the surface area of the composite scaffold is measured using gas adsorption.
31. The scaffold structure of claim 1 wherein each of the plurality of spacer yarns comprise one of monofilament fiber and multifilament fibers.
32. The scaffold structure of claim 1 wherein each the plurality of spacer yarns has a length extending between the first and second outer layers of between approximately 100 μm and 5000 μm.
