Regulating alkaloids
MPO1 and MPO2 can be regulated for either decreasing or increasing alkaloid levels in plants, in particular in Nicotiana plants. In particular, suppressing or overexpressing one or more of MPO1 and MPO2 may be used to decrease or increase nicotine and nicotinic alkaloid levels in tobacco plants. Suppression or overexpression of one or more of MPO1 and MPO2 may be used in combination with modification of expression of other genes encoding enzymes on the nicotinic alkaloid biosynthetic pathway such as A622, NBB1, PMT, and QPT.
1. A tobacco plant produced by a method comprising:
(a) mutagenizing a plurality of tobacco plant cells to comprise one or more mutations in a nucleotide sequence set forth in SEQ ID NO: 1 or SEQ ID NO: 5, thereby producing a plurality of mutagenized cells comprising the one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1 or SEQ ID NO: 5;
(b) screening plants generated from the mutagenized cells comprising the one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1 or SEQ ID NO: 5 for reduced expression of MPO1; and
(c) selecting one or more of the progeny comprising the one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1 or SEQ ID NO: 5, wherein the progeny has reduced MPO1 expression and reduced nicotine content as compared to a control tobacco plant,
thereby resulting in a tobacco plant having reduced MPO1 expression and reduced nicotine content as compared to a control non-mutagenized tobacco plant.
2. A tobacco product produced from the tobacco plant of claim 1 , wherein:
(a) the tobacco product is selected from the group consisting of smoking cessation products, cigarettes, cigarette tobacco, cigars, cigar tobacco, snus, pipe tobacco, and chewing tobacco, and
(b) the tobacco product comprises the mutagenized cells of claim 1 .
3. A product produced from the tobacco plant of claim 1 , wherein:
(a) the product is selected from the group consisting of a food product, food ingredient, feed product, feed ingredient, nutritional supplement, and biofuel, and
(b) the product comprises the mutagenized cells of claim 1 .
4. A tobacco plant comprising plant cells into which one or more mutations are introduced into a region of SEQ ID NO: 1 or SEQ ID NO: 5 that encodes a polypeptide having the amino acid sequence set forth in SEQ ID NO: 2, wherein the plant exhibits reduced expression of a gene product encoded by SEQ ID NO: 1 or SEQ ID NO: 5 as compared to a control non-mutagenized tobacco plant.
5. The tobacco plant of claim 4 , wherein the tobacco plant exhibits a reduced nicotine level as compared to a control non-mutagenized tobacco plant.
6. Seeds from or a progeny plant of the tobacco plant of claim 4 , wherein the seeds or progeny plant comprise in their genomes the one or more mutations.
7. A product comprising the plant of claim 5 , and having a reduced level of nicotine as compared to a product produced from a control non-mutagenized plant, wherein the product is selected from the group consisting of a tobacco product, food product, food ingredient, feed product, feed ingredient, nutritional supplement, and biofuel.
8. The tobacco product of claim 7 , wherein the product is selected from the group consisting of smoking cessation products, cigarettes, cigarette tobacco, cigars, cigar tobacco, snus, pipe tobacco, and chewing tobacco.
