Abstract

The inventive technology relates to novel paratransgenic strategies for the biocontrol of pathogens in animal systems using interfering RNA molecules expressed in genetically modified bacteria that may be configured to colonize a target host. In one preferred embodiment, the invention includes novel paratransgenic strategies for the biocontrol of pathogens in aquatic organisms raised in aquaculture environments.

Claims (20)

1. A composition for the biocontrol of white spot syndrome comprising a genetically modified bacteria expressing at least one heterologous nucleotide sequence having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 15, encoding at least one inhibitory polynucleotide that downregulates expression of the vp19 gene of the white spot syndrome virus (WSSV).

2. The composition of claim 1 , wherein said inhibitory polynucleotide comprises an inhibitory RNA polynucleotide selected from the group consisting of: SEQ ID NOs. 11-14, or a sequence having at least 95% homology with SEQ ID NOs. 11-14.

The composition of claim 1 , wherein said inhibitory polynucleotide comprises a first inhibitory RNA polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 11, and a second complementary polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 12, wherein said first and said second RNA polynucleotides form an inhibitory double stranded RNA molecule (dsRNA) that downregulates expression of the vp19 gene of WSSV.

The composition of claim 1 , wherein said inhibitory polynucleotide comprises a first inhibitory RNA polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 13, and a second complementary polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 14, wherein said first and said second RNA polynucleotides form an inhibitory dsRNA configured to downregulate expression of the vp19 gene of WSSV.

5. The composition of claim 1 , wherein said genetically modified bacteria comprises a genetically modified bacteria selected from the group consisting of: Bacillus subtilis, Enterobacter , a shrimp probiotic bacteria, a shrimp enteric bacteria.

6. The composition of claim 5 , wherein said genetically modified bacteria comprises an RNaseIII deficient genetically modified bacteria.

7. The composition of claim 1 , wherein said genetically modified bacteria is added to a commercial feed forming a treated feed for an aquatic animal that is infected with WSSV, or susceptible to infection by WSSV.

8. The composition of claim 1 , wherein the biocontrol of white spot syndrome comprises the biocontrol of white spot syndrome in shrimp.

9. The composition of claim 8 , wherein said shrimp comprise shrimp raised in an aquaculture environment.

The composition of claim 9 , and further comprising a therapeutically effective amount of said genetically modified bacteria administered to said shrimp such that said heterologous inhibitory polynucleotide downregulates expression of said vp19 gene and further results in a reduction in mortality due to WSSV in said shrimp that are administered a therapeutically effective amount of said genetically modified bacteria.

A composition for the biocontrol of white spot syndrome in shrimp comprising a genetically modified bacteria expressing a first inhibitory RNA polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 11, and a second complementary polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 12, wherein said first and said second RNA polynucleotides form an inhibitory dsRNA downregulate expression of the vp19 gene of the white spot syndrome virus (WSSV).

The composition of claim 11 , wherein said genetically modified bacteria comprises a genetically modified bacteria selected from the group consisting of: Bacillus subtilis, Enterobacter , a shrimp probiotic bacteria, a shrimp enteric bacteria, and an RNaseIII deficient genetically modified bacteria.

13. The composition of claim 11 , wherein said genetically modified bacteria is added to a commercial shrimp feed forming a treated feed for shrimp that is infected with WSSV, or susceptible to infection by WSSV.

14. The composition of claim 11 , wherein said shrimp comprise shrimp raised in an aquaculture environment.

15. The composition of claim 14 , wherein said genetically modified bacteria is transmitted to shrimp progeny through vertical transmission.

The composition of 15 , and further comprising a therapeutically effective amount of said genetically modified bacteria administered to said shrimp such that said heterologous inhibitory polynucleotide downregulating expression of said essential gene in a WSSV, and further results in a reduction in mortality due to WSSV in said shrimp.

A composition for the biocontrol of white spot syndrome in shrimp comprising a genetically modified bacteria expressing a first inhibitory RNA polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 13, and a second complementary polynucleotide having at least 95% homology with the nucleotide sequence according to SEQ ID NO. 14, wherein said first and said second RNA polynucleotides form an inhibitory dsRNA configured to downregulate expression of the vp19 gene of WSSV.

18. The composition of 15 , wherein said genetically modified bacteria comprises a genetically modified bacteria selected from the group consisting of: Bacillus subtilis, Enterobacter , a shrimp probiotic bacteria, a shrimp enteric bacteria, and RNaseIII deficient genetically modified bacteria.

19. The composition of 16 , wherein said genetically modified bacteria is added to a commercial shrimp feed forming a treated feed for shrimp that is infected with WSSV, or susceptible to infection by WSSV.

The composition of 15 , and further comprising a therapeutically effective amount of said genetically modified bacteria administered to shrimp such that said heterologous inhibitory polynucleotide downregulating expression of said essential gene in a WSSV, and further results in a reduction in mortality due to WSSV in said shrimp.
