Abstract

Dosage forms, drug delivery systems, and methods related to sustained release of dextromethorphan or improved therapeutic effects are disclosed. Typically, bupropion or a related compound is orally administered to a human being to be treated with, or being treated with, dextromethorphan.

Claims (23)

1. A method of treating nicotine addiction, comprising administering a drug combination comprising: a daily dose of a bupropion that is about 200 mg to about 250 mg of bupropion hydrochloride, or a molar equivalent amount of the free base form of bupropion or another salt form of bupropion, and a daily dose of a dextromethorphan that is about 80 mg to about 110 mg of dextromethorphan hydrobromide, or a molar equivalent amount of the free base form of dextromethorphan or another salt form of dextromethorphan, to a human being in need thereof; and wherein administering the drug combination is more effective in treating nicotine addiction than administering a placebo.

2. The method of claim 1 , wherein administering the drug combination is more effective in treating nicotine addiction than administering the same daily dose of the dextromethorphan alone.

3. The method of claim 1 , wherein the daily dose of the bupropion is administered once daily and the daily dose of the dextromethorphan is administered once daily.

4. The method of claim 1 , wherein one half of the daily dose of the bupropion is administered twice daily and one half of the daily dose of the dextromethorphan is administered twice daily.

5. The method of claim 1 , wherein the drug combination is administered for at least 8 consecutive days.

6. The method of claim 1 , wherein the drug combination is administered for at least 14 consecutive days.

7. The method of claim 1 , wherein the drug combination is administered for at least 30 consecutive days.

8. The method of claim 1 , wherein the dextromethorphan is formulated for immediate release.

9. The method of claim 1 , wherein the bupropion is formulated for sustained release.

10. The method of claim 1 , wherein the weight ratio of the dextromethorphan to the bupropion in the drug combination is about 0.3 to about 0.5.

11. The method of claim 1 , wherein the drug combination is administered orally.

12. The method of claim 1 , wherein the bupropion is in a solid dosage form.

13. The method of claim 1 , wherein the dextromethorphan is in a solid dosage form.

14. The method of claim 1 , wherein the human being has a C avg of bupropion that is at least about 10 ng/mL after the drug combination is administered.

15. The method of claim 1 , wherein the human being has a C max of bupropion that is at least about 90 ng/mL after the drug combination is administered.

16. The method of claim 1 , wherein the drug combination comprises a daily dose of bupropion hydrochloride that is about 210 mg and a daily dose of dextromethorphan hydrobromide that is about 90 mg.

17. The method of claim 16 , wherein administering the drug combination is more effective in treating nicotine addiction than administering 105 mg of bupropion hydrochloride twice a day for a corresponding number of consecutive days.

18. The method of claim 1 , wherein the drug combination is administered once a day for 1 to 7 days, and then twice a day for at least 4 consecutive days.

19. The method of claim 1 , wherein the drug combination is administered once a day for 1 to 3 days, and then twice a day for at least 4 consecutive days.

20. The method of claim 1 , wherein the drug combination is administered once a day for 3 days, and then twice a day for at least 4 consecutive days.

21. The method of claim 1 , wherein the human being has at least a 50% reduction in expired carbon monoxide level as compared to baseline after the drug combination is administered.

22. The method of claim 1 , wherein the human being smokes at least 7 fewer cigarettes a day as compared to baseline after the drug combination is administered.

23. The method of claim 1 , wherein the human being has at least a 20% greater reduction in the number of cigarettes smoked per day as compared to baseline after the drug combination is administered than the number of cigarettes that would have been smoked per day as compared to baseline by administering the same daily dose of the bupropion alone.
