Treatment or prevention of diseases and disorders associated with cells that express IL-6 with Anti-IL-6 antibodies
The present invention is directed to antibodies and fragments thereof and humanized versions thereof having binding specificity for IL-6. Another embodiment of this invention relates to the antibodies described herein, and binding fragments thereof, comprising the sequences of the V H , V L and CDR polypeptides described herein, and the polynucleotides encoding them. The invention also contemplates conjugates of anti-IL-6 antibodies and binding fragments thereof conjugated to one or more functional or detectable moieties. The invention also contemplates methods of making the anti-IL-6 antibodies and binding fragments thereof. Embodiments of the invention also pertain to the use of anti-IL-6 antibodies, and binding fragments thereof, for the diagnosis, assessment and treatment of diseases and disorders associated with IL-6. These antibodies may bind at least one of soluble IL-6, cell surface expressed IL-6, IL-6/IL-6R and/or prevent the association of IL-6 and IL-6R, the association of IL-6/IL-6R and gp130 and or the formation of IL-6/IL-6R/gp130 multimers and thereby inhibit a biological effect associated with any of the foregoing.

A method of treatment of adult respiratory distress syndrome (ARDS), severe acute respiratory syndrome (SARS), sepsis, or systemic inflammatory response syndrome (SIRS) in a subject in need thereof comprising administering to said subject a therapeutically effective amount of an anti-human interleukin-6 (IL-6) antibody or anti-human IL-6 antibody fragment, wherein the antibody or antibody fragment comprises: a variable light chain (VL) polypeptide comprising the complementarity determining regions (CDRs) of SEQ ID Nos: 4, 5, and 6, and a variable heavy chain (VH) polypeptide comprising the CDRs of SEQ ID Nos: 7, 8 or 120, and 9.
2. The method of claim 1 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 20 and a VH polypeptide of SEQ ID NO: 19.
3. The method of claim 2 , wherein the antibody comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
4. The method of claim 1 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 18, 19, 656, 657 or 658.
5. The method of claim 4 , wherein the antibody comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
6. The method of claim 1 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 20 and a VH polypeptide of SEQ ID NO: 18, 19, 656, 657 or 658.
7. The method of claim 6 , wherein the antibody comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
8. The method of claim 1 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 657.
9. The method of claim 8 , wherein the antibody comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
10. The method of claim 1 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 658.
11. The method of claim 10 , wherein the antibody comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
12. The method of claim 1 , wherein the antibody comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
13. A method of treatment of graft versus host disease (GVHD), reperfusion injury, or transplant rejection in a subject in need thereof comprising administering to said subject a therapeutically effective amount of an anti-human interleukin-6 (IL-6) antibody or anti-human IL-6 antibody fragment, wherein the antibody or antibody fragment comprises: a variable light chain (VL) polypeptide comprising the complementarity determining regions (CDRs) of SEQ ID Nos: 4, 5, and 6, and a variable heavy chain (VH) polypeptide comprising the CDRs of SEQ ID Nos: 7, 8 or 120, and 9.
14. The method of claim 13 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 20 and a VH polypeptide of SEQ ID NO: 19.
15. The method of claim 13 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 18, 19, 656, 657 or 658.
16. The method of claim 13 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 20 and a VH polypeptide of SEQ ID NO: 18, 19, 656, 657 or 658.
17. The method of claim 13 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 657.
18. The method of claim 13 , wherein the antibody comprises a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 658.

The method of claim 13 , wherein the antibody comprises: (a) a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 657 and further comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588; or (b) a VL polypeptide of SEQ ID NO: 651 and a VH polypeptide of SEQ ID NO: 658 and further comprises a light chain constant region polypeptide of SEQ ID NO: 586 and a heavy chain constant region polypeptide of SEQ ID NO: 588.
