Abstract

The disclosure relates to anti-cancer compounds derived from a payload which binds to poly(ADP-ribose) polymerase (PARP) and a nuclear steroid-targeting epitope (B of Formula I), which components are covalently linked via a linker, to products containing the same, as well as to methods of their use and preparation.

1. A compound of Formula A-L-B, wherein:

L is L1, L2, L3, L4, L5, L6, L7, L8, L9, L10, L11, L12, L13, L14, L15, L16, L17, L18, L19, L20, L21, L22, L23, L24, L25, L26, L27, L28, L29, L30, L31, L32, L33, L34, L35, L36, L37, L38, L39, L40, L41, L42, L43, L44, L45, or L46, wherein:

and;

B is B1, B2, B3, B4, B5, B6, B7, B8, B9, B10, B11, B12, B13, B14, B4-5-B16, B17, B18, or B19, wherein:

and further wherein the “*” in A1 and A4 and each L moiety denotes a covalent bond therebetween, and the wavy line in each L and B denotes a covalent bond therebetween;

3. The compound of claim 1 , wherein the compound is selected from:

4. A pharmaceutical composition comprising a compound of claim 1 and a pharmaceutically acceptable excipient.

5. A method of treating an androgen receptor overexpressing cancer, comprising administering an effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt thereof, to an individual in need thereof.

6. The method of claim 5 , wherein the cancer is prostate, breast, triple negative breast cancer, bladder, or liver cancer.

7. A method of treating an estrogen and/or progesterone receptor overexpressing cancer, comprising administering an effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt thereof, to an individual in need thereof.

8. The method of claim 7 , wherein the cancer is breast, uterine, or ovarian cancer.

9. A method of treating a glucocorticoid receptor overexpressing cancer, comprising administering an effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt thereof, to an individual in need thereof.

10. The method of claim 9 , wherein the cancer is prostate, breast, uterine, or ovarian cancer.
