Abstract

A sensor (e.g., an optical sensor) that may be implanted within a living animal (e.g., a human) and may be used to measure an analyte (e.g., glucose or oxygen) in a medium (e.g., interstitial fluid, blood, or intraperitoneal fluid) within the animal. The sensor may include a sensor substrate, electrode or housing, an analyte indicator covering at least a portion of the sensor, and one or more compounds that reduce degradation of the analyte indicator.

Claims (43)

1. A sensor for measurement of an analyte in a medium within a living animal, the sensor comprising:

an analyte indicator; and

one or more compounds that reduce degradation of the analyte indicator, wherein the one or more compounds are selected from one or more of Formulae I-III:

wherein in the compound of Formula I, X is a halide, each R3, R4, R5, and R6 is independently selected from H, C1-C20 alkyl, C1-C20 alkoxy, carboxy, aryl, alkoxy, halide, SH, aryloxy, alkylthio, amino, substituted amino, alkoxycarbonyl, alkanoylamido, aroylamido, heterocyclocarbonylamido, heteroaroylamido, alkanoyl(alkylsubstituted) amido, aroyl(alkylsubstituted)amido, heteroaroyl(alkylsubstituted)amido, and heterocyclocarbonyl(alkylsubstituted)amido, and wherein formula I may be optionally substituted with C1-5 alkyl, alkoxy, cyano, halo and/or trifluoromethyl at any position;

wherein in the compound of Formula III, X is O, N, S, and each of R and R 1 is independently selected from H, C1-C20 alkyl, C1-C20 alkoxy, carboxy, aryl, alkoxy, halide, SH, aryloxy, alkylthio, amino, substituted amino, alkoxycarbonyl, alkanoylamido, aroylamido, heterocyclocarbonylamido, heteroaroylamido, alkanoyl(alkylsubstituted) amido, aroyl(alkylsubstituted)amido, heteroaroyl(alkylsubstituted)amido, and heterocyclocarbonyl(alkylsubstituted)amido, and formula I may be optionally substituted with C1-5 alkyl, alkoxy, cyano, halo and/or trifluoromethyl at any position, or wherein each of R and R1 is independently absent, or a bond or linker for polymerizing the compound of Formula III to a polymer backbone.

2. The sensor of claim 1 , further comprising a sensor housing, wherein the analyte indicator covers at least a portion of the sensor housing.

3. The sensor of claim 2 , wherein the one or more compounds of Formulae I-III are entrapped in a hydrogel covering at least a portion of the sensor housing.

4. The sensor of claim 2 , further comprising at least one drug eluting region that covers a portion of the sensor housing.

5. The sensor of claim 2 , further comprising at least one drug eluting polymer matrix that covers a portion of the sensor housing.

6. The sensor of claim 1 , further comprising a sensor substrate or sensor electrode, wherein the analyte indicator covers at least a portion of the sensor substrate or sensor electrode.

7. The sensor of claim 1 , wherein the sensor is implantable within the living animal.

8. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III are co-monomers with the analyte indicator.

9. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III are co-monomers with the analyte indicator in a hydrogel.

10. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III reduce chemical degradation and/or oxidation of the analyte indicator.

11. The sensor of claim 10 , wherein the one or more compounds of Formulae I-III reduce a degradation rate of the analyte indicator.

12. The sensor of claim 10 , wherein the one or more compounds of Formulae I-III bind to the degradative species.

13. The sensor of claim 10 , wherein the one or more compounds of Formulae I-III sequester the degradative species so as to reduce, and/or prevent degradation of the analyte indicator by the degradative species.

14. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III are selected from the compound of Formula I and the compound of Formula III.

15. The sensor of claim 1 , wherein the analyte indicator comprises a polymer comprising co-monomers of four monomers according to Formula V: A-B-C-D [Formula V],

wherein A is an analyte indicator monomer, B is a methacrylate monomer, C is a polyethylene glycol monomer, and D is the one or more compounds of Formulae I-III, wherein A is 0.01 to 10% by weight, B is 1 to 99% by weight, C is 1 to 99% by weight, and D is 0.01 to 99% by weight of the total polymer.

16. The sensor of claim 2 , wherein the one or more compounds of Formulae I-III are provided at a molar ratio of 0.1 to 100 to analyte indicator monomer.

17. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III comprise the compound of Formula I.

18. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III comprise the compound of Formula II.

19. The sensor of claim 1 , wherein the one or more compounds of Formulae I-III comprise compound of Formula III.

20. A method of fabricating a sensor for measurement of an analyte in a medium within a living animal, the method comprising:

applying an analyte indicator to a sensor such that the applied analyte indicator covers at least a portion of the sensor, wherein the analyte indicator comprises one or more compounds that reduce degradation of the analyte indicator, and the one or more compounds are selected from one or more of Formula I-III:

wherein in the compound of Formula I, X is a halide, each R3, R4, R5, and R6 is independently selected from H, C1-C20 alkyl, C1-C20 alkoxy, carboxy, aryl, alkoxy, halide, SH, aryloxy, alkylthio, amino, substituted amino, alkoxycarbonyl, alkanoylamido, aroylamido, heterocyclocarbonylamido, heteroaroylamido, alkanoyl(alkylsubstituted) amido, aroyl(alkylsubstituted)amido, heteroaroyl(alkylsubstituted)amido, and heterocyclocarbonyl(alkylsubstituted)amido, and wherein formula I may be optionally substituted with C1-5 alkyl, alkoxy, cyano, halo and/or trifluoromethyl at any position;

wherein in the compound of Formula III, X is O, N, S, and each of R and R 1 is independently selected from H, C1-C20 alkyl, C1-C20 alkoxy, carboxy, aryl, alkoxy, halide, SH, aryloxy, alkylthio, amino, substituted amino, alkoxycarbonyl, alkanoylamido, aroylamido, heterocyclocarbonylamido, heteroaroylamido, alkanoyl(alkylsubstituted) amido, aroyl(alkylsubstituted)amido, heteroaroyl(alkylsubstituted)amido, and heterocyclocarbonyl(alkylsubstituted)amido, and formula I may be optionally substituted with C1-5 alkyl, alkoxy, cyano, halo and/or trifluoromethyl at any position, or wherein each of R and R1 is independently absent, or a bond or linker for polymerizing the compound of Formula III to a polymer backbone.

21. The method of claim 20 , wherein the one or more compounds of Formulae I-III are co-monomers with the analyte indicator.

22. The method of claim 20 , wherein the one or more compounds of Formulae I-III are co-monomers with the analyte indicator in a hydrogel.

23. The method of claim 20 , further comprising a sensor housing, wherein the one or more compounds of Formulae I-III are entrapped in a hydrogel covering at least a portion of the sensor housing.

24. The method of claim 20 , wherein the one or more compounds of Formulae I-III reduce chemical degradation and/or oxidation of the analyte indicator.

The method of claim 20 , wherein the one or more compounds of Formulae I-III interact or react with a degradative species without compromising signal integrity or performance of the sensor device, wherein the degradative species is hydrogen peroxide, a reactive oxygen species, a reactive nitrogen species, enzymes, free radical or metal ions.

26. The method of claim 25 , wherein the one or more compounds of Formulae I-III reduce a degradation rate of the analyte indicator.

27. The method of claim 25 , wherein the one or more compounds of Formulae I-III bind to the degradative species.

28. The method of claim 25 , wherein the one or more compounds of Formulae I-III sequester the degradative species so as to reduce, and/or prevent degradation of the analyte indicator by the degradative species.

29. The method of claim 20 , wherein the one or more compounds of Formulae I-III are one or more compounds selected from the compound of Formula I and the compound of Formula III.

30. The method of claim 20 , wherein the analyte indicator comprises a polymer comprising co-monomers of four monomers according to Formula V: A-B-C-D [Formula V],

wherein A is an analyte indicator monomer, B is a methacrylate monomer, C is a polyethylene glycol monomer, and D is the one or more compounds of Formulae I-III, wherein A is 0.01 to 10% by weight, B is 1 to 99% by weight, C is 1 to 99% by weight, and D is 0.01 to 99% by weight of the total polymer.

31. The method of claim 20 , wherein the one or more compounds of Formulae I-III are provided at a molar ratio of 0.1 to 100 to analyte indicator monomer.

32. The method of claim 20 , wherein the one or more compounds of Formulae I-III comprise the compound of Formula I.

33. The method of claim 20 , wherein the one or more compounds of Formulae I-III comprise the compound of Formula II.

34. The method of claim 20 , wherein the one or more compounds of Formulae I-III comprise compound of Formula III.
