Abstract

Systems and methods are provided for a UV-VIS spectrophotometer, such as a UV-VIS detector unit included in a high-performance liquid chromatography system. In one example, a system for the UV-VIS detector unit may include a first light source, a signal detector, a flow path positioned intermediate the first light source and the signal detector, a second light source, and a reference detector. The first light source, the signal detector, and the flow path may be aligned along a first axis, and the second light source and the reference detector may be aligned along a second axis, different than the first axis.

Claims (58)

1. A system, comprising:

a first light source;

a first signal detector;

a first flow path positioned intermediate the first light source and the first signal detector, where the first flow path, the first light source, and the first signal detector are aligned along a first axis;

a second light source;

a second signal detector;

a second flow path positioned intermediate the second light source and the second signal detector, where the second flow path, the second light source, and the second signal detector are aligned along a second axis, and the first flow path is fluidically coupled to the second flow path; and

a controller including executable instructions stored in non-transitory memory that, when executed, cause the controller to:

activate the first light source at a first activation time; and

activate the second light source at a second activation time, wherein the second activation time is determined based on a flow rate of a sample from the first flow path to the second flow path.

2. The system of claim 1 , wherein the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

determine a first sample transmittance signal based on an output from the first signal detector while the first light source is activated at the first activation time; and

calculate a first absorbance of the sample in the first flow path based on the first sample transmittance signal.

3. The system of claim 2 , wherein the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

determine a second sample transmittance signal based on an output from the second signal detector while the second light source is activated at the second activation time; and

calculate a second absorbance of a second sample in the second flow path based on the second sample transmittance signal.

The system of claim 3 , further comprising coupling optics positioned intermediate the first light source and the first flow path and positioned intermediate the second light source and the second flow path, wherein the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

determine a first reference signal based on the output of the second signal detector while the second light source is off and the first light source is activated; and

determine a second reference signal based on the output of the first signal detector while the first light source is off and the second light source is activated.

5. The system of claim 4 , wherein the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

calculate the first absorbance of the sample in the first flow path based on the first sample transmittance signal and further based on the first reference signal; and

calculate the second absorbance of the sample in the second flow path based on the second sample transmittance signal and further based on the second reference signal.

6. The system of claim 1 , wherein the first light source is configured to emit light of a first wavelength, and the second light source is configured to emit light of a second wavelength.

7. The system of claim 1 , wherein the first light source comprises a first type of light emitter, and the second light source comprises a second type of light emitter.

The system of claim 7 , wherein the first type of light emitter is one of a laser diode, a light emitting diode (LED), an organic LED, and a plasma discharge light source, and the second type of light emitter is another of the laser diode, the LED, the organic LED, and the plasma discharge light source.

9. The system of claim 1 , wherein the second axis is parallel to the first axis, and the first light source and the second light source each comprises one or more light emitters, each of the one or more light emitters configured to emit light of a different wavelength.

10. A light detector system, comprising:

a light source comprising at least two light emitters;

a signal detector;

a flow path positioned intermediate the light source and the signal detector; and

a controller including executable instructions stored in non-transitory memory that, when executed, cause the controller to:

modulate one or more of the at least two light emitters based on parameters of the light detector system, including:

during a first mode, activate a first light emitter of the at least two light emitters, maintain a second light emitter of the at least two light emitters deactivated, and determine a first sample transmittance signal based on an output from the signal detector; and

during a second mode, activate the second light emitter, maintain the first light emitter deactivated, and determine a second sample transmittance signal based on the output from the signal detector.

11. The light detector system of claim 10 , wherein the light source comprises four light emitters arranged in a two by two array, and wherein each of the four light emitters is positioned to emit light along at least a portion of the flow path.

The light detector system of claim 11 , wherein the flow path is configured to flow a sample along a longitudinal axis of the flow path, and wherein the flow path and the light source are aligned such that light emitted from each of the four light emitters is transmitted through the flow path along a respective axis parallel to the longitudinal axis.

13. The light detector system of claim 10 , wherein to modulate the one or more of the at least two light emitters, the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

during a third mode, alternately activate and deactivate the first light emitter of the at least two light emitters and the second light emitter of the at least two light emitters, and determining a third sample transmittance signal based on the output from the signal detector.

The light detector system of claim 10 , wherein to modulate the one or more of the at least two light emitters based on the parameters of the light detector system, the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

adjust one or more of a frequency and pulse width of the one or more of the at least two light emitters based on a respective efficiency of each of the at least two light emitters.

The light detector system of claim 10 , wherein to modulate the one or more of the at least two light emitters, the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to simultaneously activate the at least two light emitters.

16. A system, comprising:

a first light source;

a first signal detector;

a first flow path positioned intermediate the first light source and the first signal detector;

a second light source;

a second signal detector;

a second flow path positioned intermediate the second light source and the second signal detector, the first flow path fluidically coupled to the second flow path; and

a controller including executable instructions stored in non-transitory memory that, when executed, cause the controller to:

determine a first sample transmittance signal based on output from the first signal detector as a sample flows through the first flow path;

calculate a first absorbance of the sample based on the first sample transmittance signal;

determine a second sample transmittance signal based on output from the second signal detector as the sample flows through the second flow path;

calculate a second absorbance of the sample based on the second sample transmittance signal;

determine a reference transmittance signal based on output from the first signal detector as the sample flows through the second flow path, and not through the first flow path; and

correct the second absorbance of the sample based on the reference transmittance signal.

17. The system of claim 16 , wherein the reference transmittance signal is a first reference transmittance signal, and wherein the controller includes further executable instructions stored in non-transitory memory that, when executed, cause the controller to:

determine a second reference transmittance signal based on output from the second signal detector as the sample flows through the first flow path, and not through the second flow path; and

correct the first absorbance of the sample based on the second reference transmittance signal.
