Abstract

The present disclosure provides immunoassay methods of detecting a cleaved high molecular weight kininogen (HMWK) with high sensitivity and specificity and isolated antibodies that specifically bind cleaved HMWK.

Claims (5)

1 . An isolated antibody, which specifically binds a cleaved high molecular weight kininogen (HMWK),

wherein the antibody comprises a heavy chain complementarity determining region (CDR) 1 sequence FSFYVMV (SEQ ID NO: 84), a heavy chain CDR2 sequence GISPSGGNTAYADSVK (SEQ ID NO: 85), and a heavy chain CDR3 sequence KLFYYDDTKGYFDF (SEQ ID NO: 86) and a light chain CDR1 sequence SGSSSNIGSNYVY (SEQ ID NO: 87), a light chain CDR2 sequence RNNQRPS (SEQ ID NO: 88), and a light chain CDR3 sequence AWDDSLNGRV (SEQ ID NO: 89).

2 . The isolated antibody of claim 1 , wherein the antibody comprises a heavy chain variable region (VH) comprising the amino acid sequence of SEQ ID NO: 4 and a light chain variable region (VL) comprising the amino acid sequence of SEQ ID NO: 5.

3 . A kit for detecting a cleaved high molecular weight kininogen (HMWK), the kit comprising a first agent that specifically binds a cleaved HMWK; wherein the first agent is the isolated antibody of claim 1 .

4 . The kit of claim 3 , wherein the kit further comprises a second agent that binds HMWK, a support member, or both.
