Abstract

The invention relates to a double-stranded ribonucleic acid (dsRNA) targeting a G-alpha q subunit (GNAQ) of a heterotrimeric G gene, and methods of using the dsRNA to inhibit expression of GNAQ.

Claims (18)

1. A method of inhibiting G-alpha q subunit (GNAQ) expression in a cancer cell, the method comprising:

(a) introducing into the cancer cell an isolated double-stranded ribonucleic acid (dsRNA) comprising a sense strand and an antisense strand, wherein the sense strand comprises SEQ ID NO:1409 and the antisense strand comprises SEQ ID NO:1410, and wherein each strand is 19-30 nucleotides in length; and

(b) maintaining the cancer cell produced in step (a) for a time sufficient to obtain degradation of the mRNA transcript of a GNAQ gene, thereby inhibiting expression of the GNAQ gene in the cancer cell.

2. The method of claim 1 , wherein expression is inhibited by at least 20%, 40%, 60%, or at least 80%.

3. The method of claim 1 , wherein the cancer cell is selected from A549 (lung carcinoma), A375 (malignant melanoma) and uveal melanoma cell lines GNAQmut, OMM1.3, and MEL202.

4. The method of claim 1 , wherein the dsRNA comprises at least one modified nucleotide.

5. The method of claim 1 , wherein the dsRNA comprises at least one 2′-O-methyl modified nucleotide and at least one 2′-fluoro modified nucleotide.

6. The method of claim 1 , wherein the sense strand consists of the nucleotide sequence of SEQ ID NO:1409 and the antisense strand consists of the nucleotide sequences of SEQ ID NO:1410.

7. The method of claim 6 , wherein expression is inhibited by at least 20%, 40%, 60%, or at least 80%.

8. The method of claim 6 , wherein the cancer cell is selected from A549 (lung carcinoma), A375 (malignant melanoma) and uveal melanoma cell lines GNAQmut, OMM1.3, and MEL202.

9. The method of claim 6 , wherein the dsRNA comprises at least one modified nucleotide.

10. The method of claim 6 , wherein the dsRNA comprises at least one 2′-O-methyl modified nucleotide and at least one 2′-fluoro modified nucleotide.

The method of claim 1 , wherein the sense strand consists of SEQ ID NO:1567 (cAcAAAGAuAAGAcuuGuudTdT) and the antisense strand consists of SEQ ID NO:1568 (AAcAAGUCUuAUCUUUGUGdTdT), wherein each strand includes 2′-O-methyl ribonucleotides as indicated by a lower case letter “c” and “u”, and wherein each strand includes 2′-deoxythymidine as indicated by a “dT”.

12. The method of claim 11 , wherein the dsRNA comprises at least one 2′-fluoro modified nucleotide.

A method of treating cancer mediated by GNAQ expression comprising administering to a human in need of such treatment a therapeutically effective amount of an isolated double-stranded ribonucleic acid (dsRNA) comprising a sense strand and an antisense strand, wherein the sense strand comprises SEQ ID NO:1409 and the antisense strand comprises SEQ ID NO:1410, and wherein each strand is 19-30 nucleotides in length.

14. The method of claim 13 , wherein the sense strand consists of the nucleotide sequence of SEQ ID NO:1409 and the antisense strand consists of the nucleotide sequences of SEQ ID NO:1410.

The method of claim 13 , wherein the sense strand consists of SEQ ID NO:1567 (cAcAAAGAuAAGAcuuGuudTdT) and the antisense strand consists of SEQ ID NO:1568 (AAcAAGUCUuAUCUUUGUGdTdT), wherein each strand includes 2′-O-methyl ribonucleotides as indicated by a lower case letter “c” and “u”, and wherein each strand includes 2′-deoxythymidine as indicated by a “dT”.

16. The method of claim 13 , wherein the human has uveal melanoma, cutaneous melanoma, Blue nevi, Nevi of Ota, a small lung tumor, or a neuroendocrine tumor.
