Compounds useful for treating a Mannheimia haemolytica or Histophilus somni infection
The present invention discloses compounds that are useful in the treatment of respiratory diseases of animals, especially Bovine or Swine Respiratory disease (BRD and SRD).
1. Compound according to the formula (I):

R 1 is selected from the group consisting of —C(R 11 R 12 R 13 ), C(═O)R 11 , —C(═NR 14 )R 11 ,
R 11 is selected from the group consisting of H, and C 1-6 -alkyl;
R 12 is selected from the group consisting of
H, C 1-6 -alkyl, and C 1-6 -alkyl substituted with a substituent from the group consisting of —SR 8 , —OR 9 , —C(═O)OR 9 , —NR 9 R 10 , —SO 2 NR 9 R 10 , —SO 2 R 8 ;
R 13 is selected from the group consisting of
H, C 1-6 -alkyl, aryl, —SR 8 , —OR 9 , —NR 9 R 10 , —SO 2 R 8 , nitro, —C(═O)NR 9 R 10 , and C 1-6 -alkyl substituted with a substituent selected from the group consisting of —SR 8 , —C(═O)NR 9 R 10 , —SO 2 NR 9 R 10 , —SO 2 R 8 , nitro, cyano, —OR 9 , —C(═O)OR 9 , —NR 9 C(═NR 14 )NR 9 R 10 ;
wherein when R 13 is OH or NH 2 and R 12 is methyl then R 11 cannot be H and when R 13 is NH 2 and R 12 is methyl then R 11 cannot be methyl; and
wherein when R 13 is OH and R 12 is methyl then R 11 cannot be methyl and wherein when R 13 is NH 2 and R 12 is H then R 11 cannot be H;
R 14 is selected from the group consisting of H, C 1-6 -alkyl, or —OR 9 ;
R 2 , R 3 is H;
R 4 is —OR 9 ;
R 5 is H;
R 6 , R 7 are independently selected from the group consisting of
H, C 1-6 -alkyl, C 2-6 -alkenyl, C 2-6 -alkynyl, C 3-10 -cycloalkyl, aryl, heterocyclyl, heteroaryl, C 1-6 -alkyloxy-C 1-6 -alkyl, C 1 -C 6 -alkyl substituted with aryl, C 1 -C 6 -alkyl substituted with heteroaryl, C 1 -C 6 -alkyl substituted with heterocyclyl, or NR 6 R 7 is NO 2 or
R 6 , R 7 together with the N atom to which they are attached can form a saturated or unsaturated heterocyclic ring having 3 to 12 ring atoms, wherein 1 ring atom is N and 0, 1, 2, or 3 further ring atoms are selected from N, S, and O, the rest of the ring atoms being C;
wherein the alkyl, alkenyl, alkynyl, cycloalkyl, aryl, heterocyclyl, heteroaryl, alkyloxy or the heterocyclic ring formed by R 6 , R 7 together with the N atom to which they are attached is optionally substituted with a substituent selected from the group consisting of
C 1-6 -alkyl, C 3-8 -cycloalkyl, C 1-6 -alkyloxy, —NR 9 R 10 , carbonyl, —C(═O)OR 9 , halogen atom, C 1-6 -alkyl substituted with halo, C 1-6 -alkyloxy-C 1-6 -alkyl, aryl, heteroaryl, C 1 -C 6 -alkyl substituted with aryl, cyano, hydroxyl, —SR 8 , —SO 2 R 8 , SO 2 NR 9 R 10 , —C(═O)NR 9 R 10 , C 1-6 -alkyl substituted with hydroxyl;
R 8 is selected from the group consisting of H, C 1-6 -alkyl;
R 9 , R 10 are independently selected from the group consisting of H, and C 1-6 -alkyl;
L is C 1-6 -alkyl;
M is selected from the group consisting of aryl, heteroaryl, C 2-4 alkenyl;
G is —C≡C—;
Y is aryl; and
X is —C(═O)—.
2. A compound according to claim 1 , wherein R 6 , R 7 are independently selected from the group consisting of
H, C 1-6 -alkyl, C 2-6 -alkenyl, C 3-10 -cycloalkyl, C 1-6 -alkyloxy-C 1-6 -alkyl, C 1 -C 6 -alkyl substituted with aryl, and C 1 -C 6 -alkyl substituted with heteroaryl, or NR 6 R 7 is NO 2 or R 6 , R 7 together with the N atom to which they are attached can form a saturated or unsaturated heterocyclic ring having 3 to 12 ring atoms, wherein 1 ring atom is N and wherein 0, 1, 2, or 3 further ring atoms are selected from N, S, and O;
wherein the alkyl, alkenyl, cycloalkyl, aryl, heteroaryl, alkyloxy or the heterocyclic ring formed by R 6 , R 7 together with the N atom to which they are attached is optionally substituted with a substituent selected from the group consisting of
C 1-6 -alkyl, C 3-8 -cycloalkyl, C 1-6 -alkyloxy, —NR 9 R 10 , carbonyl, —C(═O)—OR 9 , halogen atom, C 1-6 -alkyl substituted with halo, C 1-6 -alkyloxy-C 1-6 -alkyl, aryl, heteroaryl, C 1 -C 6 -alkyl substituted with aryl, cyano, hydroxy, —SR 8 , —SO 2 R 8 , —SO 2 NR 9 R 10 , —C(═O)NR 9 R 10 , and C 1-6 -alkyl substituted with hydroxyl.
3. A compound according to claim 1 , wherein R 6 , R 7 are independently selected from the group consisting of
H, C 1-6 -alkyl, C 2-6 -alkenyl, C 2-6 -alkynyl, C 3-10 -cycloalkyl, C 1-6 -alkyloxy-C 1 -C 6 -alkyl, C 1 -C 6 -alkyl substituted with aryl, C 1 -C 6 -alkyl substituted with heteroaryl, and C 1 -C 6 -alkyl substituted with heterocyclyl, or NR 6 R 7 is NO 2 or
R 6 , R 7 together with the N atom to which they are attached can form a saturated or unsaturated heterocyclic ring having 3 to 12 ring atoms, wherein 1 ring atom is N and wherein 0, 1, 2, or 3 further ring atoms are selected from N, S, and O.
4. A compound according to claim 1 , wherein R 6 , R 7 are independently selected from
H, C 1-6 -alkyl, C 2-6 -alkenyl, C 3-10 -cycloalkyl, C 1-6 -alkyloxy-C 1 -C 6 -alkyl, C 1 -C 6 -alkyl substituted with aryl, and C 1 -C 6 -alkyl substituted with heteroaryl, or NR 6 R 7 is NO 2 or
R 6 , R 7 together with the N atom to which they are attached can form a saturated or unsaturated heterocyclic ring having 3 to 12 ring atoms, wherein 1 ring atom is N and wherein 0, 1, 2, or 3 further ring atoms are selected from N, S, and O.
5. A pharmaceutical composition comprising a compound according to claim 1 and a pharmaceutical acceptable carrier.
6. A method of treating bovine respiratory disease or swine respiratory disease comprising administering an effective amount of a compound according to claim 1 to an animal in need thereof.
7. A method of treating an infection caused by a bacteria in an animal wherein the bacteria is at least one of the bacteria selected from the group Pasteurella multocida, Mannheimia haemolytica and Histophilus somni comprising administering an effective amount of a compound according to claim 1 to an animal in need thereof.
