Abstract

Described herein are solvates of the Bruton's tyrosine kinase (Btk) inhibitor 1-((R)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one, including crystalline forms, and pharmaceutically acceptable salts thereof. Also disclosed are pharmaceutical compositions that include the solvates, as well as methods of using the solvates, alone or in combination with other therapeutic agents, for the treatment of autoimmune diseases or conditions, heteroimmune diseases or conditions, cancer, including lymphoma, and inflammatory diseases or conditions.

Claims (81)

1 . A solvate of 1-((R)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one, wherein 1-((R)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one is solvated with butyronitrile, 1,2-dimethoxyethane, hexafluorobenzene, acetophenone, chlorobenzene, dimethylacetamide, benzyl acetate, or 1,1,2-trichloroethane, or a mixture thereof.

2 . The solvate of claim 1 , which is in a crystalline form.

31 . The crystalline form of claim 2 , wherein the solvate is an acetophenone solvate that has at least one of the following properties:

(a) an X-ray powder diffraction (XRPD) pattern substantially similar to the one set forth in FIG. 9 ;

(b) an XRPD pattern comprising at least two, at least four, at least six, at least eight, at least ten, or twelve peaks at about 7.6° 2-Theta, about 8.8° 2-Theta, about 15.2° 2-Theta, about 17.6° 2-Theta, about 18.9° 2-Theta, about 19.5° 2-Theta, about 20.4° 2-Theta, about 21.0° 2-Theta, about 21.3° 2-Theta, about 21.8° 2-Theta, about 24.3° 2-Theta, and about 24.8° 2-Theta; g

(c) a differential scanning calorimetry (DSC) thermogram substantially similar to the one set forth in FIG. 10 ;

(d) a DSC thermogram having an endotherm with a peak at about 96° C.;

(e) thermo-gravimetric analysis (TGA) thermogram substantially similar to the one set forth in FIG. 10 ;

(f) unit cell parameters approximately equal to the following at a temperature of approximately 100(2) K:

32 . The crystalline form of claim 31 , wherein the crystalline form has an XRPD pattern substantially similar to the one set forth in FIG. 9 .

The crystalline form of claim 31 , wherein the crystalline form has an XRPD pattern comprising at least two peaks at about 7.6° 2-Theta, about 8.8° 2-Theta, about 15.2° 2-Theta, about 17.6° 2-Theta, about 18.9° 2-Theta, about 19.5° 2-Theta, about 20.4° 2-Theta, about 21.0° 2-Theta, about 21.3° 2-Theta, about 21.8° 2-Theta, about 24.3° 2-Theta, and about 24.8° 2-Theta.

34 . The crystalline form of claim 31 , wherein the DSC thermogram has an endotherm with a peak at about 96° C.

35 . The crystalline form of claim 31 , wherein the crystalline form has a DSC thermogram substantially similar to the one set forth in FIG. 10 .

47 . The crystalline form of claim 2 , wherein the solvate is an acetophenone solvate that has at least one of the following properties:

(a) an XRPD pattern substantially similar to the one set forth in FIG. 13 ;

(b) an XRPD pattern comprising at least two, at least four, at least six, at least eight, or nine peaks at about 6.5° 2-Theta, about 13.0° 2-Theta, about 17.6° 2-Theta, about 18.4° 2-Theta, about 19.9° 2-Theta, about 21.0° 2-Theta, about 21.5° 2-Theta, about 22.1° 2-Theta, and about 23.9° 2-Theta;

(c) a DSC thermogram substantially similar to the one set forth in FIG. 14 ;

(d) a DSC thermogram having an endotherm with a peak at about 127° C.;

(e) thermo-gravimetric analysis (TGA) thermogram substantially similar to the one set forth in FIG. 14 ;

or

(f) combinations thereof.

48 . The crystalline form of claim 47 , wherein the crystalline form has an XRPD pattern substantially similar to the one set forth in FIG. 13 .

49 . The crystalline form of claim 47 , wherein the crystalline form has an XRPD pattern comprising at least two peaks at about 6.5° 2-Theta, about 13.0° 2-Theta, about 17.6° 2-Theta, about 18.4° 2-Theta, about 19.9° 2-Theta, about 21.0° 2-Theta, about 21.5° 2-Theta, about 22.1° 2-Theta, and about 23.9° 2-Theta.

50 . The crystalline form of claim 47 , wherein the DSC thermogram has an endotherm with a peak at about 127° C.

51 . The crystalline form of claim 47 , wherein the crystalline form has a DSC thermogram substantially similar to the one set forth in FIG. 14 .

52 . The crystalline form of claim 47 , wherein the crystalline form has a TGA thermogram substantially similar to the one set forth in FIG. 14 .

62 . The crystalline form of claim 2 , wherein the solvate is a benzyl acetate solvate that has at least one of the following properties:

(a) an XRPD pattern substantially similar to the one set forth in FIG. 17 ;

(b) an XRPD pattern comprising at least two, at least four, at least six, or eight peaks at about 12.8° 2-Theta, about 17.8° 2-Theta, about 18.7° 2-Theta, about 19.2° 2-Theta, about 20.1° 2-Theta, about 20.7° 2-Theta, about 22.1° 2-Theta and about 22.9° 2-Theta;

(c) a DSC thermogram substantially similar to the one set forth in FIG. 18 ;

(d) a DSC thermogram having an endotherm with a peak at about 158° C.;

(e) a thermo-gravimetric analysis (TGA) thermogram substantially similar to the one set forth in FIG. 18 ;

or

(f) combinations thereof.

63 . The crystalline form of claim 62 , wherein the crystalline form has an XRPD pattern substantially the same as shown in FIG. 17 .

64 . The crystalline form of claim 62 , wherein the crystalline form has an XRPD pattern comprising at least two peaks at about 12.8° 2-Theta, about 17.8° 2-Theta, about 18.7° 2-Theta, about 19.2° 2-Theta, about 20.1° 2-Theta, about 20.7° 2-Theta, about 22.1° 2-Theta and about 22.9° 2-Theta.

68 . A pharmaceutical composition comprising a crystalline form of ibrutinib and at least one pharmaceutically acceptable excipient.

74 . A method of treating cancer in a mammal, comprising administering a solvate of 1-((R)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one to the mammal.

75 . The method of claim 74 , wherein the cancer is a B cell malignancy.

76 . A kit comprising the solvate of claim 1 , and instructions for use.
