Abstract

Disclosed herein are compositions comprising tobramycin or a tobramycin derivative that exhibit improvements in permeation of corneal cells, retention in corneal cells, or both. Such compositions can comprise tobramycin or tobramycin derivative(s) complexed to an agent which facilitates improved permeation of corneal cells, retention in corneal cells, or both. The invention also relates to a process of preparing such compositions.

Claims (5)

1. A pharmaceutical composition comprising an effective amount of an ophthalmologically suitable complex, the complex comprising: (a) tobramycin at 0.3 to 3% w/v, and (b) histidine at 0.3-3% w/v, wherein histidine forms a stable complex with tobramycin and the complex detectably promotes the uptake of tobramycin by corneal cells, the retention of tobramycin by corneal cells, or both, as compared to free (noncomplexed) tobramycin.

2. The pharmaceutical composition of claim 1 , wherein tobramycin is present within the composition in a concentration of between 1% and 3% w/v.

3. The pharmaceutical composition of claim 1 , wherein the rate of cornea cell permeation for the complex is at least about 15% faster than tobramycin in noncomplexed form, after about 60 minutes from administration to cornea cells.

The pharmaceutical composition of claim 1 , wherein the concentration of tobramycin permeating the surface of the cornea is at least about 15% greater after 15 minutes from administration than a corresponding concentration of tobramycin after administration of a composition consisting of tobramycin at 0.3% w/v, benzalkonium chloride at 0.01%, boric acid, sodium sulfate, sodium chloride, tyloxapol, sodium hydroxide and/or sulfuric acid, and purified water (TOBREX®).

5. The pharmaceutical composition of claim 1 , wherein an effective amount of the composition prevents detectable or significant growth of bacteria, for a period of at least 6 hours after administration.
