Abstract

CD37-specific bispecific antibody molecules binding to different epitopes of the human CD37 antigen which bispecific antibody molecules have enhanced Fc-Fc interactions upon binding to CD37 on the cell surface. The invention also relates to the monoclonal parental antibodies from which the first or the second binding region of the bispecific antibody molecules is obtained. The invention also relates to pharmaceutical compositions containing these molecules and the treatment of cancer and other diseases using these compositions.

Claims (36)

1. A bispecific antibody comprising a first binding arm and a second binding arm, wherein the first binding arm comprises a first heavy chain and a first light chain comprising a first antigen-binding region, and the second binding arm comprises a second heavy chain and a second light chain comprising a second antigen-binding region, wherein both the first antigen-binding region and second antigen-binding region bind to human CD37, and wherein

(a) the first antigen-binding region comprises a variable heavy chain (VH) region comprising the complementarity determining regions (CDRs) and the framework regions (FRs) within the amino acid sequence set forth in SEQ ID NO: 22 and a variable light chain (VL) region comprising the CDRs and FRs within the amino acid sequence set forth in SEQ ID NO: 29, and

(b) the second antigen-binding region comprises a VH region comprising the CDRs and FRs within the amino acid sequence set forth in SEQ ID NO: 15 and a VL region comprising the CDRs and FRs within the amino acid sequence set forth in SEQ ID NO: 19, and

wherein in each of the VH and VL regions, the CDRs and FRs are arranged from amino-terminus to carboxy-terminus in the following order: FR1, CDR1, FR2, CDR2, FR3, CDR3, FR4.

2. The bispecific antibody of claim 1 , wherein the first binding arm comprises a first human IgG1 heavy chain constant region and the second binding arm comprises a second human IgG1 heavy chain constant region.

The bispecific antibody of claim 1 , wherein, in the first heavy chain, the amino acid residues corresponding to E430 and F405 in a human IgG1 heavy chain according to EU numbering are glycine and leucine, respectively; wherein, in the second heavy chain, the amino acid residues corresponding to E430 and K409 in a human IgG1 heavy chain according to EU numbering are glycine and arginine, respectively; and wherein the first light chain and second light chain comprise a light chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 61.

The bispecific antibody of claim 1 , wherein, in the first heavy chain, the amino acid residues corresponding to E430 and K409 in a human IgG1 heavy chain according to EU numbering are glycine and arginine, respectively; wherein, in the second heavy chain, the amino acid residues corresponding to E430 and F405 in a human IgG1 heavy chain according to EU numbering are glycine and leucine, respectively; and wherein the first light chain and second light chain comprise a light chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 61.

The bispecific antibody of claim 1 , wherein the first heavy chain comprises a first human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 59, and the second heavy chain comprises a second IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 60.

The bispecific antibody of claim 1 , wherein the first heavy chain comprises a first human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 60, and the second heavy chain comprises a second IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 59.

7. The bispecific antibody of claim 1 , wherein both the first binding arm and second binding arm comprise a human kappa light chain constant region.

8. The bispecific antibody of claim 7 , wherein the light chain constant region consists of the amino acid sequence set forth in SEQ ID NO: 61.

9. The bispecific antibody of claim 1 , which is a full-length antibody.

10. The bispecific antibody of claim 1 , wherein the first heavy and light chains and the second heavy and light chains are all inter-connected by disulfide bonds.

A bispecific antibody comprising a first binding arm and a second binding arm, wherein the first binding arm comprises a first heavy chain and a first light chain comprising a first antigen-binding region, and the second binding arm comprises a second heavy chain and a second light chain comprising a second antigen-binding region, wherein both the first antigen-binding region and second antigen-binding region bind to human CD37, and wherein

(a) the first antigen-binding region comprises a variable heavy chain (VH) region consisting of the amino acid sequence set forth in SEQ ID NO: 22 and a variable light chain (VL) region consisting of the amino acid sequence set forth in SEQ ID NO: 29, and

(b) the second antigen-binding region comprises a VH region consisting of the amino acid sequence set forth in SEQ ID NO: 15 and a VL region consisting of the amino acid sequence set forth in SEQ ID NO: 19.

12. The bispecific antibody of claim 11 , wherein the first heavy chain comprises a first human IgG1 heavy chain constant region and the second heavy chain comprises a second human IgG1 heavy chain constant region.

13. The bispecific antibody of claim 11 , wherein both the first binding arm and second binding arm comprise a human kappa light chain constant region.

14. The bispecific antibody of claim 11 , wherein, in the first heavy chain, the amino acid residues corresponding to E430 and F405 in a human IgG1 heavy chain according to EU numbering are glycine and leucine, respectively; wherein, in the second heavy chain, the amino acid residues corresponding to E430 and K409 in a human IgG1 heavy chain according to EU numbering are glycine and arginine, respectively; and wherein the first light chain and second light chain comprise a light chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 61.

The bispecific antibody of claim 11 , wherein, in the first heavy chain, the amino acid residues corresponding to E430 and K409 in a human IgG1 heavy chain according to EU numbering are glycine and arginine, respectively; wherein, in the second heavy chain, the amino acid residues corresponding to E430 and F405 in a human IgG1 heavy chain according to EU numbering are glycine and leucine, respectively; and wherein the first light chain and second light chain comprise a light chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 61.

The bispecific antibody of claim 11 , wherein the first heavy chain comprises a first human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 59, and the second heavy chain comprises a second IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 60.

The bispecific antibody of claim 11 , wherein the first heavy chain comprises a first human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 60, and the second heavy chain comprises a second IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 59.

18. The bispecific antibody of claim 11 , which is a full-length antibody.

19. The bispecific antibody according to claim 11 , wherein the first heavy and light chains and the second heavy and light chains are all inter-connected by disulfide bonds.

20. A bispecific antibody comprising a first binding arm and a second binding arm, wherein the first binding arm comprises a first heavy chain and a first light chain comprising a first antigen-binding region, and the second binding arm comprises a second heavy chain and a second light chain comprising a second antigen-binding region, wherein both the first antigen-binding region and second antigen-binding region bind to human CD37, and wherein

(a) the first antigen-binding region comprises a variable heavy chain (VH) region consisting of the amino acid sequence set forth in SEQ ID NO: 22 and a variable light chain (VL) region consisting of the amino acid sequence set forth in SEQ ID NO: 29, and

(b) the second antigen-binding region comprises a VH region consisting of the amino acid sequence set forth in SEQ ID NO: 15 and a VL region consisting of the amino acid sequence set forth in SEQ ID NO: 19,

wherein the bispecific antibody comprises a first human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 59 and a second human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 60, and

wherein the first light chain and second light chain comprise a light chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 61.

21. The bispecific antibody of claim 20 , wherein the first heavy and light chains and the second heavy and light chains are all inter-connected by disulfide bonds.

22. A bispecific antibody comprising a first binding arm and a second binding arm, wherein the first binding arm comprises a first heavy chain and a first light chain comprising a first antigen-binding region, and the second binding arm comprises a second heavy chain and a second light chain comprising a second antigen-binding region, wherein both the first antigen-binding region and second antigen-binding region bind to human CD37, and wherein

(a) the first antigen-binding region comprises a variable heavy chain (VH) region consisting of the amino acid sequence set forth in SEQ ID NO: 22 and a variable light chain (VL) region consisting of the amino acid sequence set forth in SEQ ID NO: 29, and

(b) the second antigen-binding region comprises a VH region consisting of the amino acid sequence set forth in SEQ ID NO: 15 and a VL region consisting of the amino acid sequence set forth in SEQ ID NO: 19,

wherein the bispecific antibody comprises a first human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 60 and a second human IgG1 heavy chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 59, and

wherein the first light chain and second light chain comprise a light chain constant region consisting of the amino acid sequence set forth in SEQ ID NO: 61.

23. The bispecific antibody of claim 22 , wherein the first heavy and light chains and the second heavy and light chains are all inter-connected by disulfide bonds.
