Abstract

The present invention relates to immunoglobulins that bind FcγRIIb+ cells and coengage the antigen on the cell's surface and an FcγRIIb on the cell's surface, methods for their generation, and methods for using the immunoglobulins.

Claims (14)

1 . An antibody having affinity for CD40 comprising:

a means for binding CD40; and

an IgG1 Fc domain comprising an Fc modification comprising the amino acid substitution S267E in an Fc region, wherein the numbering is according to the EU index, as in Kabat.

2 . The antibody of claim 1 , wherein the Fc modification consists of the amino acid substitution S267E in the Fc.

3 . A pharmaceutical composition comprising the antibody of claim 1 and a pharmaceutically acceptable carrier.

4 . The pharmaceutical composition of claim 3 , wherein the antibody is in a water soluble-form.

5 . The pharmaceutical composition of claim 3 , wherein the composition is formulated for injection.

6 . The pharmaceutical composition of claim 3 , wherein the composition is a sustained release formulation.

7 . The pharmaceutical composition of claim 3 , wherein the composition further comprises a chemotherapeutic agent.

8 . The pharmaceutical composition of claim 7 , wherein the chemotherapeutic agent is a taxane.

9 . A kit comprising the pharmaceutical composition of claim 3 and a chemotherapeutic agent.

10 . The kit of claim 9 wherein the chemotherapeutic agent is a taxane.

11 . A method of activating a B cell or a dendritic cell comprising contacting a B cell or a dendritic cell with the antibody of claim 1 .

12 . A method of activating a B cell or a dendritic cell comprising contacting a B cell or a dendritic cell with the antibody of claim 2 .
