Reducing levels of nicotinic alkaloids in plants
Two genes, A622 and NBB1, can be influenced to achieve a decrease of nicotinic alkaloid levels in plants. In particular, suppression of one or both of A622 and NBB1 may be used to decrease nicotine in tobacco plants.
1. A method for producing a tobacco plant comprising:
(a) mutagenizing a plurality of tobacco plant cells to comprise one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1, thereby producing a plurality of mutagenized cells comprising the one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1;
(b) screening plants generated from the mutagenized cells comprising the one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1 for reduced expression of A622; and
(c) selecting one or more of the progeny comprising the one or more mutations in the nucleotide sequence set forth in SEQ ID NO: 1, wherein the progeny has reduced A622 expression and reduced anatabine content as compared to a control tobacco plant,
thereby resulting in a tobacco plant having reduced A622 expression and reduced anatabine content as compared to a control tobacco plant.
2. The method of claim 1 , wherein the mutagenizing comprises introducing into the plurality of tobacco plant cells a reagent for site-directed mutagenesis comprising an oligonucleotide having a targeting sequence with at least 90% identity to the nucleotide sequence set forth in SEQ ID NO: 1 or its complement.
3. A tobacco product produced from the tobacco plant of claim 1 , wherein:
(a) the tobacco product is selected from the group consisting of smoking cessation products, cigarettes, cigarette tobacco, cigars, cigar tobacco, snus, pipe tobacco, and chewing tobacco, and
(b) the tobacco product comprises the mutagenized cells of claim 1 .
4. A product produced from the tobacco plant of claim 1 , wherein:
(a) the product is selected from the group consisting of a food product, food ingredient, feed product, feed ingredient, nutritional supplement, and biofuel, and
(b) the product comprises the mutagenized cells of claim 1 .
5. A method for producing a reduced-anatabine tobacco plant comprising:
(a) introducing into a plant cell of the tobacco plant a nucleic acid construct that expresses a short interfering RNA that suppresses expression of a gene product encoded by SEQ ID NO: 1;
(b) screening the tobacco plant produced by step (a) for reduced expression of a gene product encoded by SEQ ID NO: 1 and reduced anatabine content as compared to a non-transformed control plant; and
(c) selecting the tobacco plant having suppression of a gene product encoded by SEQ ID NO: 1 and reduced anatabine content as compared to a non-transformed control plant;
thereby resulting in a tobacco plant exhibiting suppression of a gene product encoded by SEQ ID NO: 1 and reduced anatabine content as compared to a non-transformed control plant.
6. A tobacco product comprising reduced nicotine tobacco from the tobacco plant of claim 5 , having suppression of a gene product encoded by SEQ ID NO: 1.
7. The tobacco product of claim 6 , wherein the product is selected from the group consisting of a cigarette, cigarette tobacco, a cigar, cigar tobacco, pipe tobacco, chewing tobacco, snuff, snus, leaf tobacco, shredded tobacco, and cut tobacco.
8. Seeds from the plant of claim 5 , wherein the seeds comprise the nucleic acid construct.
9. A tobacco product comprising tobacco from the tobacco plant of claim 5 , having suppression of a gene product encoded by SEQ ID NO: 1.
10. The tobacco product of claim 9 , wherein the tobacco product is selected from the group consisting of a cigarette, cigarette tobacco, a cigar, cigar tobacco, pipe tobacco, chewing tobacco, snuff, snus, leaf tobacco, shredded tobacco, and cut tobacco.
