Methods and Systems For Saccharification of Biomass
Provided are methods and compositions for high yields while using reduced enzyme loads in saccharification and fermentation processes. These methods increase the efficiency of enzymes and result in improved yields and composition of saccharification and fermentation end products.

31 . A method of producing one or more fermentation end-products from a biomass composition comprising lignocellulose, the method comprising:
(a) pretreating the biomass composition to produce a pretreated biomass composition, wherein the pretreating comprises:
(i) hydration of the biomass composition to produce a hydrated biomass composition,
(ii) mechanical size reduction of the hydrated biomass composition to produce solid particles that are less than 10 mm in size, wherein the mechanical size reduction comprises cutting or shearing, and
(iii) heating of the hydrated biomass composition, thereby producing a mixture of a first population of saccharide polymers and the solid particles; and
(iv) subjecting the mixture of the first population of saccharide polymers and the solid particles to steam explosion, thereby producing the pretreated biomass composition; and
(b) hydrolyzing and fermenting the pretreated biomass composition with one or more microorganisms for a time sufficient to produce the one or more fermentation end-products.
32 . The method of claim 31 , wherein at least 50% of the solid particles in the mixture are less than 1.5 mm in size.
33 . The method of claim 31 , wherein at least 50% of the solid particles in the mixture are from about 0.1 mm to about 1 mm in size.
34 . The method of claim 31 , wherein the mechanical size reduction comprises cutting.
35 . The method of claim 34 , wherein the mechanical size reduction comprises cutting with a first rotating cutter and/or a second rotating cutter.
36 . The method of claim 31 , wherein the mechanical size reduction comprises shearing.
37 . The method of claim 31 , wherein the mechanical size reduction and the heating of the hydrated biomass composition are conducted simultaneously.
38 . The method of claim 31 , wherein the hydration comprises hydration of the biomass in an acidic aqueous medium.
39 . The method of claim 38 , wherein the acidic aqueous medium comprises an acid at from about 0.1% to about 5% v/w by dry biomass weight.
40 . The method of claim 38 , wherein the acidic aqueous medium comprises an acid that is sulfuric acid, peroxyacetic acid, lactic acid, formic acid, acetic acid, citric acid, phosphoric acid, hydrochloric acid, sulfurous acid, chloroacetic acid, dichloroacetic acid, trichloroacetic acid, trifluoroacetic acid, oxalic acid, benzoic acid, or a combination thereof.
41 . The method of claim 31 , wherein the first population of saccharide polymers in the mixture comprises oligosaccharides.
42 . The method of claim 31 , wherein the first population of saccharide polymers comprises disaccharides, trisaccharides, tetrasaccharides, pentasaccharides, hexasaccharides, heptasaccharides, octasaccharides, enneasaccharides, decasaccharides, or a combination thereof.
43 . The method of claim 31 , wherein the first population of saccharide polymers comprises glucose residues, xylose residues, mannose residues, galactose residues, rhamnose residues, arabinose residues, or a combination thereof.
44 . The method of claim 31 , wherein the heating of the hydrated biomass composition is at a temperature of from about 100° C. to about 250° C.
45 . The method of claim 31 , wherein the heating of the hydrated biomass composition is performed at a pressure of from about 100 PSIG to about 150 PSIG.
46 . The method of claim 31 , wherein the hydrolyzing and fermenting further comprises treating with one or more enzymes.
47 . The method of claim 46 , wherein the one or more enzymes comprise one or more hemicellulases, one or more cellulases, or a combination thereof.
48 . The method of claim 46 , wherein the one or more enzymes are at a total level from about 1% to about 20% w/w by dry biomass weight.
49 . The method of claim 31 , wherein the solid particles are hydrolyzed with an acid to produce a second population of saccharide polymers.
50 . The method of claim 49 , wherein the second population of saccharide polymers comprises glucose residues.
