Abstract

An antibody-resin coupling apparatus quickly and efficiently activates resin beads and couples them to antibodies, while preventing breakdown and crosslinking of the beads, thereby improving downstream column purification processes, extending the usable life of the resin beads, and increasing molecule capture efficiency of the resultant resin-antibody complexes, to allow improved isolation and purification of factor VIII molecules or other drug compounds.

Claims (30)

1. A device for coupling antibodies to resin, the device comprising:

a mixing vessel divided into an upper portion and a lower portion by a mesh screen stretched across the mixing vessel, the upper portion comprising an inlet and the lower portion comprising an outlet;

an agitator disposed within the upper portion; and

a dispersion apparatus comprising an elongated tubular structure forming a lumen, the tubular structure comprising:

a proximal portion extending vertically outside of the mixing vessel with an inlet that opens upward;

a distal portion extending horizontally within the upper portion above the agitator and comprising a closed end and a plurality of downward-facing holes; and

an elbow connecting the distal portion and proximal portion.

2. The device of claim 1 , wherein the agitator comprises a rotor and a rotating impeller.

3. The device of claim 2 , wherein the impeller comprises a hub, a first blade, and a second blade, the first and second blades extending in opposite directions perpendicularly from the hub, each blade having a blade axis transverse to a hub axis upon which the hub is configured to rotate.

4. The device of claim 3 , wherein each blade has a fixed pitch.

5. The device of claim 3 , wherein each blade rounded edges.

6. The device of claim 1 , wherein the agitator is configured to impart a force to a fluid in the mixing vessel, wherein the force is directed away from the mesh screen.

7. The device of claim 1 , wherein the holes are arranged in three rows parallel to a longitudinal axis of the distal portion.

8. The device of claim 7 , wherein the three rows are each positioned between 30 and 60 degrees apart.

9. The device of claim 1 , further comprising a valve for opening and closing the inlet.

10. The device of claim 1 , further comprising a support beam configured to support the mesh screen and to prevent the mesh screen from bowing.

11. The device of claim 1 , wherein the mesh screen is removable from the mixing vessel.

12. The device of claim 1 , wherein the elbow comprises a bend in the tubular structure which orients the inlet such that it opens in a direction that is substantially perpendicular from a longitudinal axis of the distal portion.

13. The device of claim 1 , wherein the mesh screen has a pore size between about 5 μm and about 80 μm.

14. A method for activating a resin using the device of claim 1 , the method comprising:

inserting resin beads suspended in water into the mixing vessel comprising the dispersion apparatus, the mesh screen with holes smaller than the resin beads, and the agitator disposed above the mesh screen;

distributing an activation solution comprising CNBr and acetonitrile onto the resin beads via the dispersion apparatus;

stirring the agitator for less than 5 minutes; and

draining the activation solution through the mesh screen, thereby leaving activated resin beads supported on the screen.

15. The method of claim 14 , further comprising washing the activated resin beads with a fluid.

16. The method of claim 15 , wherein washing comprises filling the mixing vessel with the fluid, incubating with stirring, and draining the fluid from the mixing vessel.

17. The method of claim 15 , wherein the fluid is water or solution comprising sodium bicarbonate and sodium chloride.

18. The method of claim 14 , wherein the stirring is performed at between 10 RPM and 40 RPM.

19. The method of claim 18 , wherein the stirring is performed at 20 RPM.

20. The method of claim 18 , wherein the stirring is performed at 35 RPM.
