Engineered uridine phosphorylase variant enzymes
The present invention provides engineered uridine phosphorylase (UP) enzymes, polypeptides having UP activity, and polynucleotides encoding these enzymes, as well as vectors and host cells comprising these polynucleotides and polypeptides. Methods for producing UP enzymes are also provided. The present invention further provides compositions comprising the UP enzymes and methods of using the engineered UP enzymes. The present invention finds particular use in the production of pharmaceutical compounds.
1. An engineered uridine phosphorylase comprising a polypeptide sequence having at least 90% sequence identity to SEQ ID NO: 2, or a functional fragment thereof, wherein the polypeptide sequence of said engineered uridine phosphorylase comprises a substitution at position 80 and wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 2.

7. The engineered uridine phosphorylase of claim 1 , wherein said engineered uridine phosphorylase comprises a polypeptide sequence that is at least 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99% or more identical to SEQ ID NOs: 2, 4, 246, 594, 776, and/or 868.
8. An engineered uridine phosphorylase, wherein said engineered uridine phosphorylase comprises a variant engineered uridine phosphorylase set forth in SEQ ID NOs: 4, 246, 594, 776, and/or 868.

The engineered uridine phosphorylase of claim 1 , wherein said engineered uridine phosphorylase comprises a polypeptide sequence that is at least 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99% or more identical to the sequence of at least one engineered uridine phosphorylase variant set forth in the even numbered sequences of SEQ ID NOs: 4-1196.
10. An engineered uridine phosphorylase, wherein said engineered uridine phosphorylase comprises a polypeptide sequence forth in at least one of the even numbered sequences of SEQ ID NOs: 4 and 246-1196.
11. The engineered uridine phosphorylase of claim 1 , wherein said engineered uridine phosphorylase comprises at least one improved property compared to wild-type Escherichia coli uridine phosphorylase.
12. The engineered uridine phosphorylase of claim 11 , wherein said improved property comprises improved activity on one or more substrates.
13. The engineered uridine phosphorylase of claim 12 , wherein said one or more substrates comprises 5′-isobutyryl ribose-1-phosphate (compound (2)) and/or uracil (compound (3)).
14. The engineered uridine phosphorylase of claim 11 , wherein said improved property comprises improved production of 5′-isobutyryluridine, compound (1).
15. The engineered uridine phosphorylase of claim 1 , wherein said engineered uridine phosphorylase is purified.
16. The engineered uridine phosphorylase of claim 1 , wherein said engineered uridine phosphorylase is part of a multi-enzyme system for producing a nucleoside analogue.
17. A composition comprising at least one engineered uridine phosphorylase of claim 1 .
