Tie-2 activators targeting the Schlemm's canal
Disclosed herein are compounds effective for activation of Tie-2 and inhibition of HPTP-beta. The compounds can provide effective therapy for eye conditions, for example, intraocular pressure, ocular hypertension, and glaucoma.
1. A method for increasing area of a Schlemm's canal in an eye of a subject in need thereof, the method comprising administering to the subject a therapeutically effective amount of a Tie-2 activator, wherein the Tie-2 activator is a compound of the formula:
or a pharmaceutically-acceptable salt or zwitterion thereof, and
wherein administering the Tie-2 activator to the subject increases area of the Schlemm's canal by at least about 10% as compared to absence of administration.
2. The method of claim 1 , wherein administering the Tie-2 activator in the subject increases area of the Schlemm's canal by at least about 12% as compared to absence of administration.
3. The method of claim 1 , wherein the administering is by a unit dosage form, wherein the unit dosage form comprises the Tie-2 activator.
4. The method of claim 3 , wherein the unit dosage form further comprises a pharmaceutically-acceptable excipient.
5. The method of claim 1 , wherein the administering is by a sustained release formulation, wherein the sustained release formulation comprises the Tie-2 activator.
6. The method of claim 1 , wherein the administering is by a solid hydrophobic polymer, wherein the solid hydrophobic polymer comprises the Tie-2 activator.
7. The method of claim 1 , wherein the administering is by a microparticle, wherein the microparticle comprises the Tie-2 activator.
8. The method of claim 1 , wherein the administering is topical.
9. The method of claim 1 , wherein the administering is subcutaneous.
10. The method of claim 1 , wherein the administering is intravitreal.
11. The method of claim 1 , wherein Tie-2 is activated in the Schlemm's canal of the subject upon the administering of the Tie-2 activator.
12. The method of claim 1 , wherein the Tie-2 activator binds to HPTPβ in a Schlemm's canal of the subject.
13. The method of claim 1 , wherein the Tie-2 activator inhibits HPTPβ in a Schlemm's canal of the subject.
14. The method of claim 1 , wherein the Tie-2 activator is a compound of the formula:
or a pharmaceutically-acceptable salt or zwitterion thereof.
15. The method of claim 1 , wherein the increase in the area of the Schlemm's canal is determined by comparison to the area of the Schlemm's canal of the eye at baseline.
16. The method of claim 14 , wherein the increase in the area of the Schlemm's canal is determined by comparison to the area of the Schlemm's canal of the eye at baseline.
