Abstract

Cardiovascular prostheses that include an ECM-mimicking composition, which is adapted to induce modulated healing of damaged or diseased cardiovascular tissue and, thereby, associated structures, when administered thereto. The ECM-mimicking composition includes a polymer component and a pharmacological component. The polymer component includes poly(glycerol sebacate) (PGS). The pharmacological component includes a corticosteroid. The ECM-mimicking composition can also include a biological component having a biologically active agent, such as an exosome, a cytokine and a growth factor.

Claims (24)

1 . A cardiovascular prosthesis, comprising:

an ECM-mimicking composition, said ECM-mimicking composition comprising a polymer component and a pharmacological component,

said polymer component comprising poly(glycerol sebacate) (PGS),

said pharmacological component comprising a biological composition comprising a corticosteroid selected from the group consisting of dexamethasone, dexamethasone phosphate, dexamethasone dipropionate, betamethasone, betamethasone phosphate, prednisolone, fluocinolone acetonide, endrisone, methylprednisolone suleptanate, alclometasone dipropionate, clobetasone butyrate, fluorometholone acetate, isoflupredone acetate, meclorisone, rimexolone and desoximetasone.

2 . The prosthesis of claim 1 , wherein said pharmacological composition further comprises an HMG-CoA reductase inhibitor.

3 . The prosthesis of claim 2 , wherein said HMG-CoA reductase inhibitor comprises (3R,5S,6E)-7-[4-(4-fluorophenyl)-5-(methoxymethyl)-2,6-bis(propan-2-yl)pyridin-3-yl]-3,5-dihydroxyhept-6-enoic acid (cerivastatin).

4 . The prosthesis of claim 1 , wherein said ECM-mimicking composition further comprises a biological component.

5 . The prosthesis of claim 4 , wherein said biological component comprises a biological composition.

The prosthesis of claim 5 , wherein said biological composition comprises extracellular matrix (ECM) from a mammalian tissue source selected from the group consisting of small intestine submucosa (SIS), urinary bladder submucosa (UBS), stomach submucosa (SS), cardiac tissue, placental tissue, amniotic membrane tissue, and mesothelial tissue.

7 . The prosthesis of claim 6 , wherein said biological composition further comprises a biologically active agent.

8 . The prosthesis of claim 7 , wherein said biologically active agent comprises an exosome derived from a cell source selected from the group consisting of mesenchymal stem cells and embryonic stem cells.

9 . The prosthesis of claim 7 , wherein said biologically active agent comprises a cytokine selected from the group consisting of interleukin-8 (IL-8) and interleukin-10 (IL-10).

The prosthesis of claim 7 , wherein said biologically active agent comprises a growth factor selected from the group consisting of a fibroblast growth factor-2 (FGF-2), transforming growth factor alpha (TGF-α), transforming growth factor beta (TGF-β), vascular endothelial growth factor (VEGF), hepatocyte growth factor (HGF), insulin-like growth factor (IGF), nerve growth factor (NGF), platelet derived growth factor (PDGF), tumor necrosis factor alpha (TNF-α) and placental growth factor (PLGF).

11 . A cardiovascular prosthesis, comprising:

a sheet structure comprising an ECM-mimicking composition, said ECM-mimicking composition comprising a polymer component and a pharmacological component,

said polymer component comprising poly(glycerol sebacate) (PGS),

said pharmacological component comprising a biological composition comprising a corticosteroid selected from the group consisting of dexamethasone, dexamethasone phosphate, dexamethasone dipropionate, betamethasone, betamethasone phosphate, prednisolone, fluocinolone acetonide, endrisone, methylprednisolone suleptanate, alclometasone dipropionate, clobetasone butyrate, fluorometholone acetate, isoflupredone acetate, meclorisone, rimexolone and desoximetasone.

12 . The prosthesis of claim 11 , wherein said ECM-mimicking composition further comprises a biological component.

13 . The prosthesis of claim 12 , wherein said biological component comprises a biological composition.

14 . The prosthesis of claim 13 , wherein said biological composition comprises extracellular matrix (ECM) from a mammalian tissue source selected from the group consisting of small intestine submucosa (SIS), urinary bladder submucosa (UBS), stomach submucosa (SS), cardiac tissue, placental tissue, amniotic membrane tissue, and mesothelial tissue.

15 . The prosthesis of claim 13 , wherein said biological composition further comprises a biologically active agent.

16 . The prosthesis of claim 15 , wherein said biologically active agent comprises an exosome derived from a cell source selected from the group consisting of mesenchymal stem cells and embryonic stem cells.

17 . The prosthesis of claim 15 , wherein said biologically active agent comprises a cytokine selected from the group consisting of interleukin-8 (IL-8) and interleukin-10 (IL-10).

18 . The prosthesis of claim 15 , wherein said biologically active agent comprises a growth factor selected from the group consisting of a fibroblast growth factor-2 (FGF-2), transforming growth factor alpha (TGF-α), transforming growth factor beta (TGF-β), vascular endothelial growth factor (VEGF), hepatocyte growth factor (HGF), insulin-like growth factor (IGF), nerve growth factor (NGF), platelet derived growth factor (PDGF), tumor necrosis factor alpha (TNF-α) and placental growth factor (PLGF).
